The effects of
1-antitrypsin (
1,-AT),
1,-acid glycoprotein (
1AGP), and haptoglobin (Hp), the main constituents of-globulin and which belong to acute phase proteins, on NK activity were examined using K562 cells as the NK target cells. Among the three proteins,
1,-AT and
1AGP had inhibitory effects on NK activity for fast target K562 cells. The,-AT preparations having the same protein concentration and a different trypsin inhibitory capacity (TIC) had an equal effect. Although
1AT and
1,-AGP equally reduced the NK activity, the mechanism involved in the reduction differed, in that the effect of
1,-AT directed toward NK cells reduced their binding capacity with the target cells,
1,-AGP probably interacts with a cytotoxic factor secreted from NK cells following effector-target interaction. These studies suggest that each of the acute-phase proteins, which increase following inflammation, inhibits NK cell function by two distinct mechanisms. 相似文献

Interferon (IFN)-gamma was produced with a high yield in cultures of human peripheral mononuclear cells by combined stimulation with OK-432 and staphylococcal enterotoxin B. Human mononuclear cells cultured in serum-free medium produced several times as much IFN as those in RPMI-1640 medium containing 10% fetal bovine serum. A synergistic effect of OK-432 and staphylococcal enterotoxin B on the production of IFN-gamma was demonstrated. Ultrogel AcA54 column chromatography of crude IFN showed a single peak with an apparent molecular weight of 43,000. Our production system for human IFN-gamma offers a feasible approach to preparation of large quantities of purified IFN-gamma for structure studies, antibody production, and clinical applications. 相似文献

The first Japanese En(a-) individual (T.N.) was found by screening red cells from 250,000 Japanese blood donors with monoclonal anti-Ena. His serum contained no atypical antibodies and his partial red cell phenotype was M-N-S+s-, although a trypsin- resistant N antigen was detected. His red cells were En(a-) and Wr(b-), as determined by various human and mouse monoclonal antibodies. The absence of glycophorin A (GPA) and the presence of apparently normal glycophorin B (GPB) were demonstrated by immunoblotting with antibodies to the extracellular and cytoplasmic domain of GPA and to epitopes common to GPA and GPB. Sialic acid levels of T.N.'s intact red cells were substantially lower than those of control MN cells. Isolated rat Kupffer cells produced a factor which stimulated the synthesis of
2-macroglobulin (2M) in primary cultured rat hepatocytes. Although Kupffer cells placed in culture produced the factor without stimulation by lipopolysaccharide (LPS), the LPS-stimulated cells produced larger amounts of the factor. On the other hand, the production of the factor was inhibited by addition of actinomycin D. The induction of2M synthesis by cultured hepatocytes was enhanced in the presence of dexamethasone (Dex), in that hepatic synthesis of2M increased by addition of the factor alone and with Dex 1.5 and three- to four-fold, respectively. The factor was nondialyzable and stable at 60°C for 30 min. When the factor was fractionated using the molecular sieve method, the activity recovered in the fraction had a molecular weight of over 30,000. 相似文献

Screening tests for HIV and HTLV-I in donated blood were introduced into Japan in Nov. 1986. I) The incidence of HIV-antibody (Ab) among donated blood in Japan is hitherto as low as 1-2/500,000. However, those Ab tests used at present may overlook HIV-infected donors before of their seroconversion. A newly developed HIV-antigen (Ag) test has yielded positive results in 4 out of 29 (13.8%) of our HIV-Ab+ hemophiliacs (all in the AC stage), although none of the 27 HIV-Ab negatives were HIV-Ag+. The rate of Ag+ was higher in the group showing disappearance of p 24-band on the Western blot (WB). The isolation of HIV was successful in 6 out of 13 (46.2%) AC hemophiliacs, even in three with negative HIV-Ag. The usefulness of these new tests for screening should be estimated from the standpoint of the cost/benefit balance. In future, the screening for HIV-2 may also be needed. II) Gelatin particle agglutination test (PA) could detect not only IgG- but also IgM-HTLV-I Ab, while the latter is overlooked by ELISA. On the other hand, a few of false negatives by PA could be observed. Anti-HTLV-I Ab was found in 2% of our donors, the frequency being higher among those who had previously received blood transfusions. Oxygen-related free radicals have been suggested as a cause of aging and various diseases, for example, various cancers and rheumatoid arthritis. A radical scavenger as an antioxidant has been sought in foods. Fish sauces are traditional Asian fermented seasonings. Using the luminol chemiluminescence method, the peroxyl radical scavenging capability of fish sauces was examined. From the IC50 values, many fish sauces have been shown to have a strong scavenging capability as well as soy sauces. A scavenging mechanism is also proposed. 相似文献
