After retinoic acid treatment, a large percentage of cells of the human embryonal carcinoma cell line NT2/D1 differentiate into neuronal cells. We demonstrate here that the differentiated cells, but not the undifferentiated cells, contain high levels of neurofilament mRNA. We have also measured mRNA, protein, and activity levels of two kinases, cAMP-dependent protein kinase (PKA) and protein kinase C (PKC), in order to explore the role of protein kinases in the establishment of the differentiated state. RNA levels for the catalytic (C alpha and C beta) subunits of PKA increased after differentiation. Total PKA activity levels increased 7-fold in the differentiated cells. Parallel with this, a rise in the level of catalytic subunit protein occurred. A 12-fold induction of Type 2 (beta) PKC mRNA levels was observed after neuronal differentiation. Increases in PKC activity and in Type 2 (beta) and Type 3 (alpha) PKC protein levels also accompanied differentiation. These changes in PKA- and PKC-specific RNA levels and enzyme activity may be necessary for production and maintenance of the differentiated state in these cells.  相似文献

An integrating Rn daughter dosimeter using conventional Dy-doped CaSO4 thermoluminescent (TL) material has been developed for use as a personal monitor. The dosimeter was found to have a linear response over the range 0.5 WL-h to 25 WL-h and to be insensitive to the state of equilibrium of the Rn daughters. The use of the dosimeter to monitor both 222Rn and 220Rn daughters is described. The limiting sensitivity of the dosimeter, at a sampling rate of 1 L min-1, was determined to be 0.1 WL-h for 222Rn daughters and 0.5 WL-h for 220Rn daughters. The application of the thermoluminescent dosimeter (TLD) to occupational monitoring of 222Rn and 220Rn daughters is discussed.  相似文献

This experiment represents the first time that it has been possible to measure a body fluid compartment by direct means during spaceflight. Based on the results observed in the five crewmen in this study, it is concluded that TBW decreases by 3.4% after 1 to 3 days of exposure to microgravity in the Space Shuttle. Some individuals appear to undergo this decrease within 24 hours. This effect may be enhanced by decreased water intake due to nausea associated with SMS.  相似文献

Analogues of dioctanoylglycerol (diC8) and 1-oleoyl-2-acetylglycerol (OAG) containing an iodoaryl group have been synthesized and shown to compete with [3H]phorbol dibutyrate [( ([3H]PDBu) for binding to protein kinase C in a crude rat brain preparation. Phorbol diesters have been shown to bind specifically to protein kinase C and the PDBu receptor has been copurified with protein kinase C activity. All three diacylglycerol analogues were comparable to OAG in binding affinity. In an assay of protein kinase C activation, the diC8 analogue was more active than the OAG analogues, thus demonstrating greater structural specificity under the conditions of this assay.  相似文献

Copper deficiency in the laying hen resulted in anemia and the production of eggs which were abnormal in size and shape. Many of the eggs had shells which were wrinkled and rough in texture. There was also an increase in the number of shell-less eggs. Examination of malformed egg shells using the scanning electron microscope revealed ultrastructural changes in the mammillary layer of the shell. The effect of copper deficiency on shell formation was attributed to the shell membranes which were altered in color, appearance, and physical consistency. Amino acid analysis of the membranes indicated that the membranes from copper deficient hens were characterized by an increase in lysine content. This suggests that copper is necessary for the formation of lysine derived cross-links in a manner similar to that which occurs in connective tissue. The exact nature of these cross-links is unknown at this time.  相似文献
