Many adjuvant trials have been undertaken in an attempt to reduce the risk of recurrence among patients who undergo surgical resection for locally advanced renal cancer. In the present study we investigated the effects of phenolic compounds present in Hypericum perforatum against neuronal excitotoxicity and mitochondrial dysfunction. Quercetin, kaempferol and biapigenin significantly reduced neuronal death caused by 100 microM kainate plus 100 microM N-methyl-D-aspartate. The observed neuroprotection was correlated with prevention of delayed calcium deregulation and with the maintenance of mitochondrial transmembrane electric potential. The three compounds were able to reduce mitochondrial lipid peroxidation and loss of mitochondrial transmembrane electric potential caused by oxidative stress induced by ADP plus iron. Moreover, biapigenin was also able to significantly affect mitochondrial bioenergetics and decrease the capacity of mitochondria to accumulate calcium. Taken together, the results suggest that the neuroprotective action induced by quercetin and kaempferol are mainly mediated by antioxidant effects, whereas biapigenin mainly affects mitochondrial bioenergetics and calcium uptake.  相似文献

Between June 1989 and June 1992, 144 patients participated in sequential clinical trials using peripheral blood progenitor cells (PBC) as their sole source of hematopoietic rescue following high-dose chemotherapy. All patients had received prior extensive combination chemotherapy and had marrow defects that precluded autologous bone marrow transplantation (ABMT). PBC were collected according to a single apheresis protocol. The initial 86 patients (group 1) had PBC collected without mobilization. Beginning in April 1991, PBC were mobilized solely with recombinant human granulocyte-macrophage colony-stimulating factor (rHuGM-CSF). Thirty-four patients (group 2) received rHuGM-CSF at a dose of 125 micrograms/m2/d by continuous intravenous infusion, and 24 patients (group 3) received rHuGM-CSF at a dose of 250 micrograms/m2/d by continuous intravenous infusion. Patients underwent at least six aphereses and had a minimum of 6.5 x 10(8) mononuclear cells (MNC)/kg collected. Cytokines were not routinely administered immediately after transplantation. A median of nine aphereses were required to collect PBC in group 1 and seven aphereses for groups 2 and 3 (P = .03). The time required to recover 0.5 x 10(9)/L granulocytes after transplant was significantly shorter (P = .0004) for the mobilized groups; the median time to recovery was 26 days for group 1, 23 days for group 2, and 18 days for group 3. Transplantation of PBC mobilized with rHuGM-CSF resulted in a shorter time to platelet (P = .04) and red blood cell (P = .01) transfusion independence. Mobilization with rHuGM-CSF alone resulted in efficient collection of PBC, that provided rapid and sustained restoration of hematopoietic function following high-dose chemotherapy. Mobilization of PBC with rHuGM-CSF alone is an effective method for patients who have received prior chemotherapy and have bone marrow abnormalities.  相似文献

To evaluate the feasibility of chronic atrial pacing (AAI) in sick sinus syndrome (SSS), 22 patients (pts) with bradytachycardia syndrome (BTS) and 17 patients with only bradyarrhythmias (BA) were studied on the incidence of supraventricular tachycardias (SVT) and occurring AV block. A scoring system based on symptoms of SVT was developed (grade 0–5). All patients had proven normal AV conduction before PM implantation. In the BTS-group, nine patients (41%) had symptomatic SVT at the end of follow-up (mean 53 months), despite drug therapy. These patients had a high SVT score on entry (mean 3.2). High degree AV block occurred in three patients. Although in the BA-group SVT arose in six patients (35%), there was only one symptomatic patient at the end of follow-up (mean 36 months). In this group, only one patient developed high degree AV block. Atrial stimulation should be considered as a reliable therapy in patients with SSS and low SVT score before PM implantation when normal AV conduction is present.  相似文献
