Introduction

This study examined the identity of the microbiome of deep dentinal caries and its correlation with the inflammation status of caries-induced pulpitis.

Methods

Seventy-five cases were diagnosed based on the American Association of Endodontics's diagnostic criteria and divided into 4 groups: normal pulp with deep caries (NP; n = 13), reversible pulpitis with only cold-evoked pain (CRP; n = 17), reversible pulpitis with both cold/heat-evoked pain (CHRP; n = 24), and symptomatic irreversible pulpitis (SIP; n = 21). Samples were sequenced by 16S rDNA. Alpha and beta diversity were determined. Linear discriminant analysis effect size (LEfSe) analysis was used to detect intergroup differences, and receiver operating characteristic (ROC) curves were generated to assess the role of the caries microbiome in caries-induced pulpitis.

Results

The 16S rDNA sequencing yielded 9100 operational taxonomic units. Lactobacillus had the highest relative abundance at the genus level among the 4 groups. There were significant differences in the distribution of the microbiome among the groups. In an alpha diversity analysis, species richness differed between the CRP group and the other groups. In a beta diversity analysis, the distribution of microorganisms in the SIP group was significantly different from those in the other 3 groups. LEfSe analysis indicated substantial differences in the microbiome among the groups, and the areas under the ROC curves (AUC) were all high (AUC: 0.734–0.952).

Conclusions

Characterization of the caries microbiome has the potential to become an auxiliary method for the diagnosis of pulpitis. This finding may prompt new research on diagnostic strategies for caries-induced pulpitis.  相似文献

OBJECTIVES: The aim of the present study was to prepare nano-sized calcium fluoride (CaF(2)) that could be used as a labile F reservoir for more effective F regimens and as an agent for use in the reduction of dentin permeability. METHODS: Nano-sized CaF(2) powders were prepared using a spray-drying system with a two-liquid nozzle. The properties of the nano-CaF(2) were studied and the effectiveness of a fluoride (F) rinse with nano-CaF(2) as the F source was evaluated. The thermodynamic solubility product of the nano-CaF(2) solution was determined by equilibrating the nanosample in solutions presaturated with respect to macro-CaF(2). Reactivity of the nano-CaF(2) was assessed by its reaction with dicalcium phosphate dehydrate (DCPD). F deposition by 13.2 mmol/L F rinse with the nano-CaF(2) as the F source was determined using a previously published in vitro model. RESULTS: X-ray diffraction (XRD) analysis showed pattern of low crystalline CaF(2). BET measurements showed that the nano-CaF(2) had a surface area of 46.3m(2)/g, corresponding to a particle size of 41nm. Transmission electron microscopy (TEM) examinations indicated that the nano-CaF(2) contained clusters comprising particles of (10-15) nm in size. The nano-CaF(2) displayed much higher solubility and reactivity than its macro-counterpart. The CaF(2) ion activity product (IAP) of the solution in equilibrium with the nano-CaF(2) was (1.52+/-0.05)x10(-10), which was nearly four times greater than the K(sp) (3.9 x 10(-11)) for CaF(2). The reaction of DCPD with nano-CaF(2) resulted in more F-containing apatitic materials compared to the reaction with macro-CaF(2). The F deposition by the nano-CaF(2) rinse was (2.2+/-0.3)mug/cm(2) (n=5), which was significantly (p<0.001) greater than that ((0.31+/-0.06)mug/cm(2)) produced by the NaF solution. SIGNIFICANCE: The nano-CaF(2) can be used as an effective anticaries agent in increasing the labile F concentration in oral fluid and thus enhance the tooth remineralization. It can also be very useful in the treatment for the reduction of dentin permeability.  相似文献

目的研究集落刺激因子(CSF-1)对体外培养的人牙囊细胞骨保护素(OPG)表达的影响。方法原代培养人牙囊细胞,传代至第3代,制备细胞爬片,进行OPG免疫组化染色;3~6代人牙囊细胞长至80%时,用胰酶消化,离心,重悬,制成单细胞悬液,接种到细胞培养皿,细胞长满至80%时,培养基中加入25ng/ml的CSF-1,孵育0.5、1、3、6h,分别收集上清液,ELISA法检测OPG蛋白分泌量的变化,提取总RNA进行RT-PCR,检测OPG mRNA的表达变化。结果正常人牙囊细胞OPG蛋白表达阳性;25ng/ml的CSF-1可降低人牙囊细胞OPG的表达,共同孵育1h,OPG表达降至最低(P<0.05)。结论牙囊细胞表达OPG,同时在牙齿萌出的特定时期,CSF-1通过下调人牙囊细胞OPG,减弱对破骨细胞形成的抑制作用,促进破骨细胞分化成熟,调节牙齿萌出。  相似文献

目的:观察三维放疗计划优化技术减少头颈部癌放射性口干症的效果。方法:对三类头颈部癌(肿瘤位中线、偏一侧、近中线偏一侧)按常规放疗和优化技术分别作三维放疗计划。放疗优化技术包括放射进路优化、多野、适形放疗和加楔形板。对患者的肿瘤、腮腺和脊髓作三维剂量分布和剂量一容积DVH直方图分析。20例头颈部癌放疗患者分成常规放疗组和新技术(优化)组行放射治疗,记录放疗前、放疗中、放疗后及随访期患者的口干症评分和全唾液量。结果:肿瘤位一侧及近中线偏一侧时,常规和新技术放疗时健侧腮腺放射剂量分别为51.8-64.3Gy和0.4-1.4Gy;肿瘤位中线时常规和新技术放疗腮腺放射剂量分别为51.8Gy和24.7Gy;放疗结束时,新技术组口干症发生率1/12(8.3%)。随访4-6月后降为0,放疗结束时常规放疗组口干症发生率7/8(87.5%),随访4-6月后为4/8(50%)。结论:三维放疗计划优化技术可以明显减少头颈部癌患者放疗后口干症的发生。  相似文献

A Bi2Te3 VA–VIA group compound thin film was prepared via the route of electrochemical atomic layer epitaxy in this paper. The dependence of thin film formation on the electrochemical conditions (such as deposition potential control, supporting electrolyte and substrate) was studied, and the formation process of Bi2Te3 film was determined. The results show that Bi upd on Pt occurs at more negative potential in a HNO3 supporting electrolyte than in a HClO4 supporting electrolyte, and the Te upd peak moves to a more negative potential in HClO4 supporting electrolyte solution than that in HNO3 solution; Both Te and Bi upd on an Ag substrate occur at more negative potentials than that on a Pt substrate. In order to reach steady state deposition, a potential adjustment is necessary for the first 30 or more cycles of each component. After deposition of this initial ‘buffer layer’, the potentials can be kept constant for the remaining cycles. The effect of the slope of the shift of potential used to deposit the first 30 atomic layers of Bi and Te on the deposit has also been investigated. The deposit exhibits a two phase mixture of excess elemental Bi and Bi2Te3 compound when the slope is larger than ?4 mV/p (p indicates per cycle); a single-phase Bi2Te3 compound was obtained at a slope of ?6 mV/p, and Bi4Te3 compound also appears in the deposit along with Bi2Te3 when the slope is decreased to a more negative value of ?10 mV/p.  相似文献

目的对比评价临床常用的3种类型(不同长度、直径)微型种植钉的生物力学特点。方法应用种植钉48枚,新西兰大白兔8只。采用拉出试验,扭转强度试验,在即刻与愈合4周后进行生物力学测量。结果长度为6mm,直径为1.9mm的种植钉相对于同等长度,直径为1.5mm的种植钉在“种植后即刻”与“愈合4周”时,最大剪切力均表现出显著性优势。愈合4周后,直径为1.9mm的种植体在最大扭转强度上表现出显著性的优势。结论在骨质量较差的部位选择较大直径的种植体有利于种植体的稳定。  相似文献

Monoclonal antibodies (mAbs) were used to determine the presence and distribution of immune cells including lymphocytes, macrophages and Langerhans cells, in normal periodontal ligament, periapical granulomata, periapical cysts and dental developmental cysts. Isolated T-lymphocytes, but not B-lymphocytes, were detected in specimens of non-inflamed periodontal ligament. Increased numbers of T and B lymphocytes were found in all of the lesions examined. Monocytes/macrophages were associated with most periapical granulomata, dental developmental cysts and all periapical cysts. Langerhans cells, intraepithelial lymphocytes, and monocytes/macrophages were not detected in the rests of Malassez but were found in some epithelia within periapical granulomata and in most epithelial linings of odontogenic cysts. Increased numbers of immune cells were seen around proliferative epithelia and adjacent to the epithelial linings of cysts. Epithelium, particularly that of odontogenic cysts, showed positive reactions for HLA-Dr, lysozyme and for α-1 antitrypsin. The presence of immune cells in periapical granulomata and odontogenic cysts, suggests that cell-mediated and humoral immunoreactions occur in these lesions and may be associated with the epithelial proliferation within the periapical lesions.  相似文献

Objectives

To investigate the effects of acidic and neutral 30% hydrogen peroxide (HP) on human tooth enamel in terms of chemical structure, mechanical property, surface morphology and tooth colour.

Methods

Twenty-seven human dental blocks were obtained from premolars and randomly divided into three groups (n = 9): Group acidic HP (30% HP, pH ≈ 3.6), Group neutral HP (30% HP, pH ≈ 7.0) and Group DW (distilled water, pH ≈ 6.8). Attenuated total reflection-Fourier transform infrared spectroscopy (ATR-FTIR), Raman spectroscopy, atomic force microscopy (AFM) investigation, microhardness test and colour measurements were carried out before and after treatments. ATR-FTIR and Raman spectroscopy were analysed and then the carbonate:mineral ratio (C:M), Raman absolute intensity (RAI), Raman relative intensity (RRI), and laser-induced fluorescence intensity (FI) were obtained for evaluation.

Results

The C:M, percentage microhardness and percentage RRI of group acidic HP decreased more significantly than those of group neutral HP (P = 0.02, P = 0.001, P < 0.001, respectively) and group DW (P = 0.01, P = 0.008, P < 0.001, respectively). Whilst group neutral HP and group DW had no statistical difference in above terms (P = 0.818, P = 0.528, P = 0.158, respectively). Significant morphological alterations were observed in group acidic HP. Group acidic HP and neutral HP had no significant difference in percentage FI (P = 0.652) and ΔE (P = 0.906).

Conclusions

This study suggested that neutral 30% HP had the same efficiency in tooth bleaching and it caused less deleterious effects on enamel than acidic 30% HP.  相似文献
