A novel coumarin‐based fluorescence probe has been constructed for the selective and sensitive detection of hydrogen sulfide (H2S). This probe displays high sensitivity and linearity to H2S in degassed PBS buffers and fetal bovine serum. It reacts with H2S with high selectivity over Cys, GSH, and other anions. Meanwhile, successful detection of H2S in living cells was also demonstrated.  相似文献

This paper presents the study on the hydrogen evolution reaction (HER) of the silicon nanowire (SiNW)-based surfaces. Large-area SiNWs with different lengths were fabricated on the silicon surfaces by a cost effective and scalable wet-etching method. The SiNW-based surfaces promoted the photoelectrocatalytical performance of the electrodes due to the increased effective surface area for electrolyte diffusion and the fast release of hydrogen bubbles that formed on the electrodes. In addition, at different applied potentials, the nanostructured electrodes showed different behaviour that depended on the SiNWs'' with different lengths and morphologies. For example, surfaces with longer SiNWs performed better in the low potential region, while surfaces with shorter SiNWs presented improved performance in the high potential region. The findings in this study provide new insights into designing electrodes with desired nanostructures for improved HER performance.

Ongoing studies have determined that the gut microbiota is a major factor influencing both health and disease. Host genetic factors and environmental factors contribute to differences in gut microbiota composition and function. Intestinal dysbiosis is a cause or a contributory cause for diseases in multiple body systems, ranging from the digestive system to the immune, cardiovascular, respiratory, and even nervous system. Investigation of pathogenesis has identified specific species or strains, bacterial genes, and metabolites that play roles in certain diseases and represent potential drug targets. As research progresses, gut microbiome–based diagnosis and therapy are proposed and applied, which might lead to considerable progress in precision medicine. We further discuss the limitations of current studies and potential solutions.  相似文献

Isoelectric focusing on slabs of acrylamide gel was adapted for the screening of abnormal hemoglobins, the characterization of 70 human variants, and the study of minor fractions of normal hemoglobin. The screening method was as fast and inexpensive as conventional techniques, allowed the simultaneous analysis of some 50 samples of whole blood, and yielded resolution superior to that obtained by other methods with hemolysates. Among the 70 variants, 31 mutants could not be separated from HbS by cellulose acetate electrophoresis. The characterization technique of electrofocusing allowed us to distinguish between most variants. Only one mutant, Hb Galveston, could be confused with HbS. Hb Koln, the most frequent unstable mutant, exhibited a special pattern. HbA1C was separated from HbA. Preliminary results indicate that quantitation of HbA1C by gel scanning is feasible.  相似文献

目的提出日本血吸虫病肝脏超声显像肝实质图像的分类。方法利用原研究课题曾在洞庭湖的华容、汉寿、澧县中度流行区3个临湖自然村的居民及同期在本院住院的晚期血吸虫病人超声显像检查图像中的2918例肝脏图片,进行分类分型。结果肝实质图像有光点正常91例、增粗1757例、光斑121例、光条467例、光带482例等5类。其形状有光点正常91例、增粗1757例、光片59例、光斑62例、细条323例、粗条144例、细带280例、粗带202例等8型。结论超声显像诊断日本血吸虫病图像可分为5类8型。  相似文献

Bacterial pathogenicity islands (PAI) often encode both effector molecules responsible for disease and secretion systems that deliver these effectors to host cells. Human enterohemorrhagic Escherichia coli (EHEC), enteropathogenic E. coli, and the mouse pathogen Citrobacter rodentium (CR) possess the locus of enterocyte effacement (LEE) PAI. We systematically mutagenized all 41 CR LEE genes and functionally characterized these mutants in vitro and in a murine infection model. We identified 33 virulence factors, including two virulence regulators and a hierarchical switch for type III secretion. In addition, 7 potential type III effectors encoded outside the LEE were identified by using a proteomics approach. These non-LEE effectors are encoded by three uncharacterized PAIs in EHEC O157, suggesting that these PAIs act cooperatively with the LEE in pathogenesis. Our findings provide significant insights into bacterial virulence mechanisms and disease.  相似文献

Bioavailability of oral melatonin in humans   总被引:6,自引:0,他引:6  
We administered crystalline melatonin (80 mg) in gelatin capsules to 5 young male volunteers and measured serum and urinary melatonin levels at intervals. Changes in serum melatonin levels were best described by a biexponential equation with an absorption constant (ka) of 1.72 h-1 (half-life = 0.40 h) and an elimination constant (ke1) of 0.87 h-1 (half-life = 0.80 h). Peak serum melatonin levels, ranging from 350 to 10,000 times those occurring physiologically at nighttime, were observed 60-150 min after its administration, remaining stable for approximately 1.5 h. The fraction of ingested melatonin that was absorbed, estimated from the area under the curve describing serum melatonin concentrations as a function of time after melatonin administration (the concentration-time curve), varied by 25-fold among subjects. 3 additional volunteers received three melatonin-containing capsules (80 mg each) at 60-min intervals. This regimen extended the duration of elevated serum melatonin levels to 4-6 h. Melatonin excretion closely paralleled serum melatonin levels until 9 h after the hormone's administration, after which urinary levels tended to be higher than those predicted from serum levels. However, the area under the concentration-time curve for serum melatonin correlated well (r = 0.96) with the cumulative melatonin excretion during the initial 15 h after melatonin's administration, indicating that either approach can be used to estimate the absorption of orally administered melatonin.  相似文献
