Bone marrow stromal cells (MSCs) increase vascular endothelial growth factor (VEGF) expression and promote angiogenesis after stroke. Angiopoietin-1 (Ang1) and its receptor Tie2 mediate vascular integrity and angiogenesis as does VEGF and its receptors. In this study, we tested whether MSC treatment of stroke increases Ang1/Tie2 expression, and whether Ang1/Tie2 with VEGF/ vascular endothelial growth factor receptor 2 (VEGFR2) (Flk1), in combination, induced by MSCs enhances angiogenesis and vascular integrity. Male Wistar rats were subjected to middle cerebral artery occlusion (MCAo) and treated with or without MSCs. Marrow stromal cell treatment significantly decreased blood-brain barrier (BBB) leakage and increased Ang1, Tie2, and occludin (a tight junction protein) expression in the ischemic border compared with MCAo control. To further test the mechanisms of MSC-induced angiogenesis and vascular stabilization, cocultures of MSCs with mouse brain endothelial cells (MBECs) or astrocytes were performed. Supernatant derived from MSCs cocultured with MBECs significantly increased MBEC expression of Ang1/Tie2 and Flk1 compared with MBEC alone. Marrow stromal cells cocultured with astrocytes also significantly increased astrocyte VEGF and Ang1/Tie2 expression compared with astrocyte culture alone. Conditioned media from MSCs alone, and media from cocultures of MSCs with astrocytes or MBECs, all significantly increased capillary tube-like formation of MBEC compared with control Dulbecco's modified Eagle's medium media. Inhibition of Flk1 and/or Ang1 significantly decreased MSC-induced MBEC tube formation. Knockdown of Tie2 expression in MBECs significantly inhibited MSC-induced tube formation. Our data indicate MSC treatment of stroke promotes angiogenesis and vascular stabilization, which is at least partially mediated by VEGF/Flk1 and Ang1/Tie2.  相似文献

To extend the signal longevity of magnetically excited spins in flowing fluids while in a state of global coherent free precession (GCFP), a refocusing radiofrequency (RF) pulse and bipolar gradient waveforms were combined with the GCFP sequence. The data demonstrate that RF refocusing in the presence of flowing blood is possible, but the improvement in signal amplitude depends on the static magnetic field homogeneity along the direction of motion and the displacement of the spins between the excitation and the RF refocusing pulse, as well as displacement during subsequent RF refocusing pulses. The least amount of phase dispersion and thus the longest lasting signal is obtained with the shortest echo spacing where only one line of data is recorded between two RF refocusing pulses. This approach was successfully used in a phantom and in vivo to image fast and slow blood flow. Depending on the experimental conditions, signal persistence is improved significantly compared to playing the same sequence without RF refocusing, but the improvement is limited by the product of blood flow velocity and the time between RF refocusing pulses.  相似文献

目的 探讨螺旋cT功能重建在骨与关节外伤中的应用,提高重建技术的使用能力和诊断水平。方法 对使用双层螺旋CT,SSD,MPR,MIP等多种方式重建检查的49例复杂的骨与关节外伤病例进行回顾性分析。结果 49例骨关节外伤重建成像,其中,颅底3例,颌面部7例,颈、胸、腰、骶椎共17例,肩胛骨3例,髋关节8例,膝关节11例。SSD在显示碎骨片明显移位的空间关系方面较MIP,MPR为佳,它可以直观地、立体地、清晰地、多角度显示骨关节损伤,而MIP在显示骨折细节方面较优,尤其在外伤中颅窝观察有关神经孔方面。结论 螺旋CT的SSD,MPR和MIP技术的综合应用,对复杂的骨折、关节损伤的显示效果良好,具有较高的临床应用价值。  相似文献

目的探讨经尿道等离子双极电切术(PKRP)治疗良性前列腺增生(BPH)术后对阴茎勃起功能的影响。方法PKRP、经尿道前列腺电切术(TURP)治疗BPH各95例,评价其术后1个月、3个月及1年阴茎勃起功能。结果PKRP、TURP术后1个月、3个月及1年阴茎勃起功能障碍(ED)发生率分别是3.2%、2.1%、2.1%和13.7%、10.5%、9.5%。结论PKRP术后ED发生率比TURP低,是一种对阴茎勃起功能较为安全的治疗良性前列腺增生的方法。  相似文献

对诊断PADAM的血清游离睾酮界限值的初探   总被引:1,自引:1,他引:0  
目的探寻诊断中老年男子部分雄激素缺乏综合征(PADAM)血清游离睾酮(FT)的界限值。方法根据60例45岁以上健康男性完成伊斯坦布尔症状量表(ISS)与视听性刺激(AVSS)时阴茎勃起监测结果进行分组,计算AVSS勃起功能正常组推算的游离睾酮(CFT)值的参考范围。结果AVSS勃起功能正常组CFT的中位数为0.37(0.20~0.52)nmol/L,异常组的中位数为0.25(0.12~0.40)nmol/L,z=2.25,P<0.05,差别有显著性意义。两组性功能症状的差别也有极显著性意义(P<0.01)。CFT的界限值是0.20nmol/L。结论研究用ISS和AVSS勃起功能正常对象的血清FT值,可为诊断PADAM建立界限值。  相似文献

目的 :了解急性胰腺炎患者血清免疫抑制酸性蛋白 (IAP)的临床意义。方法 :应用免疫扩散法检测 32例患者和 2 0例健康人中的IAP水平。结果 :正常组 (2 0例 )、轻症组 (2 0例 )和重症组 (12例 )的血清IAP在入院时分别为(32 5± 10 5 )mg/L、(40 4± 15 1)mg/L和 (5 75± 14 5 )mg/L ,重症组血清IAP水平高于正常组 (P <0 .0 1) ,也高于轻症组 (P <0 .0 5 )。其次重症组IAP值在入院后第 3、5、7天与轻症组比较仍明显升高 ,两组有统计上的差异性。结论 :检测IAP可以作为了解急性胰腺炎炎症程度的指标  相似文献

We report a rare chronic encapsulated intracerebral haematoma (CEICH). A 52-year-old man had two seizures. Unenhanced computed tomography scanning of the head revealed a hypodense tumour with clusters of calcification in the left temporal lobe. Magnetic resonance imaging of the brain showed a left temporal tumour with a hypointense centre and hyperintense periphery on T(1)-weighted imaging and heterogeneous hypointensity on T(2)-weighted imaging. The tumour was heterogeneously enhanced after gadolinium injection. Craniotomy was carried out and a CEICH in the left temporal lobe was completely excised. No vascular anomaly was found. The tumour was histologically confirmed to be a CEICH. The patient recovered well after the operation. In this report, we describe this rare case and discuss the characteristics of CEICH.  相似文献

The mechanisms underlying neurologic deficits and delayed neuronal death after ischemia are not fully understood. In the present study, we report that transient cerebral ischemia induces accumulation of ubiquitinated proteins (ubi-proteins) in postsynaptic densities (PSDs). By immunoelectron microscopy, we demonstrated that ubi-proteins were highly accumulated in PSD structures after ischemia. On Western blots, ubi-proteins were markedly increased in purified PSDs at 30 minutes of reperfusion, and the increase persisted until cell death in the CA1 region after ischemia. In the resistant DG area, however, the changes were transient and significantly less pronounced. Deposition of ubi-proteins in PSDs after ischemia correlates well with PSD structural damage in the CA1 region as viewed by electron microscopy. These results suggest that the ubiquitin-proteasome system fails to repair and remove damaged proteins in PSDs. The changes may demolish synaptic neurotransmission, contribute to neurologic deficits, and eventually lead to delayed neuronal death after transient cerebral ischemia.  相似文献
