目的 研究AML1-ETO9a (AE9a)异构体在伴有t(8;21)的M2型急性髓系白血病( AML-M2)患者中表达比例的变化及其临床意义.方法 应用RT-PCR方法筛选了44例初诊AE9a与AML1-ETO(AE)融合基因共表达的伴有t(8;21)的AML-M2患者.应用实时定量PCR技术监测这44例多次随访标本中AE9a在AE9a与AE两者总和中百分比的变化.结果 伴有t(8;21)的44例初诊AML-M2患者中AE表达量高于AE9a(中位CT值分别为20.48和21.54),两者表达量呈正相关(r =0.900);44例初诊患者经首次标准方案化疗后AE与AE9a表达量均下降,但初治后AE9a在总体中百分比较初诊时上升(P<0.05);患者经1个疗程标准方案化疗后,未达完全缓解(CR)组的AE9a百分比明显高于CR组(中位值分别为36.81%和7.93%,P<0.05);复发患者缓解期间AE9a百分比高于未复发患者(中位值分别为22.89%和2.43%,P<0.05);缓解期间,17例复发患者中11例AE9a百分比在AE表达低水平时即出现升高甚至异常升高.结论 AE9a与AE共表达于伴有t(8;21)的AML-M2患者中,并且AE表达量高于AE9a,两者呈正相关.AE9a异构体对标准化疗的敏感性较AE差.在缓解期监测AE9a在AE及AE9a两者总体中百分比的变化比单独监测AE融合基因能更早地预示疾病复发趋势,从而可提前进行临床干预,降低患者的复发率,提高患者的长期生存率. 相似文献

The current study was designed to observe chondrogenic differentiation of adipose derived stem cells (ASCs) on fibrous polyglycolic acid (PGA) scaffold stabilized with polylactic acid (PLA), and to further explore the feasibility of using the resulting cell/scaffold constructs to repair full thickness articular cartilage defects in non-weight bearing area in porcine model within a follow-up of 6 months. Autologous ASCs isolated from subcutaneous fat were expanded and seeded on the scaffold to fabricate ASCs/PGA constructs. Chondrogenic differentiation of ASCs in the constructs under chondrogenic induction was monitored with time by measuring the expression of collagen type II (COL II) and glycosaminoglycan (GAG). The constructs after being in vitro induced for 2 weeks were implanted to repair full thickness articular cartilage defects (8 mm in diameter, deep to subchondral bone) in femur trochlea (the experimental group), while scaffold alone was implanted to serve as the control. Histologically, the generated neo-cartilage integrated well with its surrounding normal cartilage and subchondral bone in the defects of experimental group at 3 months post-implantation, whereas only fibrous tissue was filled in the defects of control group. Immunohistochemical and toluidine blue staining confirmed the similar distribution of COL II and GAG in the regenerated cartilage as the normal one. A vivid remolding process with post-operation time was also witnessed in the neo-cartilage as its compressive moduli increased significantly from 50.55% of the normal cartilage at 3 months to 88.05% at 6 months. This study investigated the in vitro effects of akermanite, a new kind of Ca-, Mg-, Si-containing bioceramic, on the attachment, proliferation and osteogenic differentiation of human adipose-derived stem cells (hASCs). Parallel comparison of the cellular behaviors of hASCs on the akermanite was made with those on beta-tricalcium phosphate (beta-TCP). Scanning electron microscope (SEM) observation and fluorescent DiO labeling were carried out to reveal the attachment and growth of hASCs on the two ceramic surfaces, while the quantitative assay of cell proliferation with time was detected by DNA assay. Osteogenic differentiation of hASCs cultured on the akermanite and beta-TCP was assayed by ALP expression and osteocalcin (OCN) deposition, which was further confirmed by Real-time PCR analysis for markers of osteogenic differentiation. It was shown that hASCs attached and spread well on the akermanite as those on beta-TCP, and similar proliferation behaviors of hASCs were observed on the two ceramics. Both of them exhibited good compatibility to hASCs with only minor cytotoxicity as compared with the tissue culture plates. Interestingly, the osteogenic differentiation of hASCs could be enhanced on the akermanite compared with that on the beta-TCP when the culture time was extended to approximately 10 days. 背景:单纯使用克氏针或外固定架治疗桡骨远端骨骺骨折的临床实践中,仍存在桡骨短缩、骨折移位等固定后并发症,尤其在不稳定骨骺骨折情况下。克氏针主要治疗桡骨远端骨折,对其骨骺骨折使用相对较少。
目的:观察外固定支架结合经皮克氏针复位固定微创治疗桡骨远端不稳定性骨骺骨折的临床效果。
方法:对90例桡骨远端骨骺骨折患儿行固定治疗,在不切开情况下微创闭合复位骨折骨骺,随机分成2组,对照组采用单纯跨腕关节外固定支架固定方案;观察组采用跨腕关节外固定支架结合经皮克氏针闭合帮助复位固定骨骺骨折方案。固定后行腕关节功能锻炼,分别于固定后9周及24个月随访观察。对比两组患者固定后中远期的临床疗效、腕关节功能恢复及X射线检查情况。
结果与结论:固定后24个月随访按Cooney标准评定腕关节功能,对照组优良率77%,X射线评定优良率63%;观察组腕关节功能优良率93%,X射线评定优良率为90%,两组差异具有显著性意义(P < 0.01)。两组骨骺骨折患者均获临床骨愈合,腕关节均功能恢复。提示外固定支架结合克氏针闭合复位微创治疗桡骨远端不稳定骨骺骨折,可应用克氏针辅助复位掌倾角及尺偏角,治疗简单,固定可靠,腕关节功能恢复良好,骨骺畸形愈合并发症少,固定后基本生活学习功能恢复正常,疗效稳定满意,其临床疗效明显优于单纯跨外固定支架固定。 相似文献

目的 观察脐血T淋巴细胞及其亚群低剂量辐射 (LDR)效应。方法 对脐血T淋巴细胞及其亚群分离、培养 ,LDR后用3 H 胸腺嘧啶核苷 (3 H TdR)掺入法观察其DNA合成和抗肿瘤细胞K562 活性 ,集落培养观察脐血干细胞造血能力。结果 脐血T淋巴细胞及其亚群CD4、CD8、CD19细胞经 2、5、10、2 0cGy的LDR与 0cGy(对照组 )比较 ,3 H TdR掺入明显增加 ,5cGy的LDR与 0cGy比较 ,3 H TdR掺入分别为 14 3 2 %、14 6 8%、14 5 4 %、15 7 9%。对K562 细胞杀伤作用经 2、5、10、2 0cGy的LDR均有明显增高 ,5cGy的LDR与 0cGy比较杀伤率和NK相对活性分别为 38 3%和 172 0 %。造血干细胞集落数无差别 ,5cGy的LDR与 0cGy比较分别为 12 2和 12 1个。结论 适宜的LDR对脐血T淋巴细胞和亚群的DNA合成有刺激作用 ,能增加体外抗肿瘤细胞K562活性 ,并保持脐血干细胞造血能力 相似文献

为了探讨14-3-3ζ蛋白在中枢神经系统内的分布,本文采用纯化的基因重组型14-3-3ζ蛋白免疫Balb/c小鼠,用杂交瘤技术制备抗14-3-3ζ蛋白单克隆抗体;Western blot及免疫组织化学法鉴定抗体。结果显示:制备的单克隆抗体能特异性识别基因重组14-3-3ζ蛋白和大鼠脑匀浆32kD蛋白。免疫组化染色结果显示,14-3-3ζ蛋白在大鼠中枢神经系统广泛分布,在皮层、海马、杏仁核和基底前脑的免疫阳性染色较强,免疫阳性产物位于神经元胞浆内。本研究结果提示,14-3-3ζ蛋白可能具有多样化的生理功能。 相似文献

Autosomal recessive nonsyndromic hearing impairment (ARNSHI) segregating in three unrelated, large consanguineous Pakistani families (PKDF528, PKDF859 and PKDF326) is linked to markers on chromosome 12q14.2-q15. This novel locus is designated DFNB74 . Maximum two-point limit of detection (LOD) scores of 5.6, 5.7 and 2.6 were estimated for markers D 12 S 313, D 12 S 83 and D 12 S 75 at θ = 0 for recessive deafness segregating in these three families. Haplotype analyses identified a critical linkage interval of 5.35 cM (5.36 Mb) defined by D 12 S 329 at 74.58 cM and D 12 S 313 at 79.93 cM. DFNB74 is the second ARNSHI locus mapped to chromosome 12, but the physical intervals do not overlap with one another. A locus contributing to the early onset, rapidly progressing hearing loss of A/J mice ( ahl4 , age-related hearing loss 4) was reported to map to chromosome 10 in a region of conserved synteny to DFNB74 , suggesting that ahl4 and DFNB74 may be due to mutations of the same gene in these two species. 相似文献
