Background and purpose:

The ionic mechanisms underlying nitrergic inhibitory junction potentials (IJPs) in gut smooth muscle remain a matter of debate. Recently, it has been reported that opening of TWIK-related K+ channel 1 (TREK-1) K+ channels contributes to the nitrergic IJP in colonic smooth muscle. We investigated the effects of TREK-1 channel blockers on nitrergic neurotransmission in mouse and opossum lower oesophageal sphincter (LOS) circular smooth muscle (CSM).

Experimental approach:

The effects of TREK-1 channel blockers were characterized pharmacologically in murine and opossum gut smooth muscle using conventional intracellular and tension recordings.

Key results:

In LOS, L-methionine depolarized the resting membrane potential (RMP) but did not inhibit the nitrergic IJP. Cumulative application of theophylline hyperpolarized the RMP and inhibited the nitrergic IJP concentration dependently. The induced membrane hyperpolarization was prevented by pre-application of caffeine, but not by 1H-[1,2,4]oxadiazolo-[4,3-a]quinoxalin-1-one. 8-Br-cAMP significantly hyperpolarized membrane potential and increased the amplitude of the nitrergic IJP. In opossum LOS muscle strips, L-methionine increased resting tone but had no effect on nerve-mediated LOS relaxation. On the other hand, theophylline markedly inhibited tone. In CSM from mouse proximal colon, L-methionine caused modest inhibition of nitrergic IJPs.

Conclusions and implications:

TREK-1 channels were not involved in the nitrergic IJP in LOS CSM. Not only does L-methionine have no effect on the nitrergic IJP or LOS relaxation, but the effect of theophylline appears to be due to interruption of Ca2+-releasing pathways (i.e. caffeine-like effect) rather than via blockade of TREK-1 channels.  相似文献

Immunological cross-reactions were found between each of the four major virus-specific polypeptides of Marek's disease virus (MDV) strains CVI 988, K and HPRS-16/att, and herpesvirus of turkey (HVT) Fc126 by one-dimensional (1D) gel analysis of immunoprecipitates from the lysate or culture medium of infected cells. The results, however, did not allow a serotype classification of MDV and HVT strains. Comparison of the two-dimensional (2D) gel patterns of virus-specific polypeptides of nine MDV and HVT strains with different biological properties revealed many similarities. Strain CVI 988 provided the reference pattern, consisting of 35 polypeptides, 18 of which were glycosylated. Based on their similarities in migration characteristics (size, charge, and heterogeneity of spots) during 2D gel electrophoresis, 11 virus-specific polypeptides or polypeptide complexes were identified in the patterns of nearly all virus strains analyzed. One of these polypeptides was glycoprotein gp5, the putative A antigen of MDV and HVT, which was also detected in the medium of cells infected with HPRS-16/att, a strain which was reported to have lost its capacity for production of A antigen. In addition to the similarities mentioned above, differences were found in migration behavior of virus-specific polypeptides (complexes) p4/p5/p6, gp3, gp5, and gp8/gp9, which were confirmed by coelectrophoresis experiments. The most conspicuous difference was found between three patterns of gp5 which seem to be characteristic for three groups of viruses: (I) high- and low-virulent MDV strains and their attenuated variants; (II) avirulent and apparently nononcogenic MDV strains; (III) herpesviruses of turkey. The minor differences found for the other polypeptides mentioned above further substantiated this molecular-biological classification of MDV and HVT strains and, in addition, enabled the differentiation of two HVT strains within molecular-biological group III.  相似文献

The introduction of the Nordmøre grid to shrimp trawls has reduced the issue of bycatch to that of small-sized species and juveniles that are able to pass through the grid and enter the small-meshed cod end together with the targeted shrimp. This study estimated the size- and species-selective performance of the Nordmøre grid in the configuration most often applied by fishermen and made a preliminary exploration of the effects of reducing the length of the guiding funnel in front of the grid and mounting light-emitting diodes (LEDs) around the escape exit. Experimental fishing trials were conducted in the Barents Sea to assess the size-selective properties of a 19-mm bar spacing Nordmøre grid, mandatory in this Norwegian trawl fishery targeting deepwater shrimp Pandalus borealis (also known as northern shrimp), and its potential improvement. Results were obtained for the target species and four bycatch species: redfish Sebastes spp., Haddock Melanogrammus aeglefinus, Atlantic Cod Gadus morhua, and American Plaice Hippoglossoides platessoides. In general, very few deepwater shrimp were found to escape through the escape exit, although the quantity increased slightly at larger sizes. Between 80% and 100% of the bycatch species up to a species-specific size passed through the grid and entered the cod end. A short guiding funnel decreased this for Haddock significantly by increasing the fraction of small Haddock seeking the escape exit. Further, adding LEDs around the escape exit significantly negated this effect. For the other bycatch species, the results indicated similar trends but were not statistically significant. However, considering that only a few fishing hauls were conducted using the short guiding funnel with or without mounting LEDs calls for some caution when attributing these changes to the configuration of the Nordmøre grid, and these preliminary results should be followed up by further testing before definitive conclusions are made.

Necrotizing fasciitis is a severe soft tissue infection that can involve skin, subcutaneous fat, fascia and muscle. It can result in devastating sequelae including tissue necrosis, sepsis, toxic shock syndrome, cardiopulmonary collapse and death. To control rapidly spreading necrosis, early diagnosis and aggressive surgical treatment with extensive radical debridement of the affected areas is necessary, as well as systemic administration of broad-spectrum antimicrobials and, very often, intensive care support.The subatmospheric negative pressure dressing has been previously used in acute and complex wounds management. The concept of using vacuum-assisted closure dressing as another management component is presented in the current article.  相似文献

Background

Timing is critical for efficient hepatitis A vaccination in high endemic areas as high levels of maternal IgG antibodies against the hepatitis A virus (HAV) present in the first year of life may impede the vaccine response.

Objectives

To describe the kinetics of the decline of anti-HAV maternal antibodies, and to estimate the time of complete loss of maternal antibodies in infants in León, Nicaragua, a region in which almost all mothers are anti-HAV seropositive.

Methods

We collected cord blood samples from 99 healthy newborns together with 49 corresponding maternal blood samples, as well as further blood samples at 2 and 7 months of age. Anti-HAV IgG antibody levels were measured by enzyme immunoassay (EIA). We predicted the time when antibodies would fall below 10 mIU/ml, the presumed lowest level of seroprotection.

Results

Seroprevalence was 100% at birth (GMC 8392 mIU/ml); maternal and cord blood antibody concentrations were similar. The maternal antibody levels of the infants decreased exponentially with age and the half-life of the maternal antibody was estimated to be 40 days. The relationship between the antibody concentration at birth and time until full waning was described as: critical age (months) = 3.355 + 1.969 × log10(Ab-level at birth). The survival model estimated that loss of passive immunity will have occurred in 95% of infants by the age of 13.2 months.

Conclusions

Complete waning of maternal anti-HAV antibodies may take until early in the second year of life. The here-derived formula relating maternal or cord blood antibody concentrations to the age at which passive immunity is lost may be used to determine the optimal age of childhood HAV vaccination.  相似文献

Migraine headaches and depression often co-occur within individuals, and both syndromes run in families. However, knowledge about how these disorders relate across generations, as well as how migraine relates to other forms of psychopathology, is sparse. This study examined risk for migraine among female adolescent offspring of parents with different types of psychopathology. The sample was drawn from the Minnesota Twin Family Study, a community-based study of adolescents and their families ( n  = 674, 17-year-old female adolescents and their biological parents). Diagnoses of maternal, paternal and offspring major depression, antisocial behaviour, alcohol dependence and drug dependence were based on structured interviews. Migraine headaches in each family member were assessed via interviews with the mother. Parental depression, antisocial behaviour and drug dependence were associated with offspring migraine. These associations mostly remained significant even when parental migraine and the corresponding type of psychopathology in offspring were adjusted for. In contrast, there were no significant associations between parental psychopathology and offspring stomach problems, indicating that these associations did not extend to all offspring somatic symptoms. These results emphasize the need to look at antisocial behaviour and substance-related problems when examining associations between migraine and psychopathology, and indicate that more research on inter-generational links between migraine and psychopathology is needed.  相似文献

Introduction

The Intra-Aortic Balloon Pump (IABP) is frequently used to mechanically support the heart. There is evidence that IABP improves microvascular flow during cardiogenic shock but its influence on the human microcirculation in patients deemed ready for discontinuing IABP support has not yet been studied. Therefore we used sidestream dark field imaging (SDF) to test our hypothesis that human microcirculation remains unaltered with or without IABP support in patients clinically ready for discontinuation of mechanical support.

Methods

We studied 15 ICU patients on IABP therapy. Measurements were performed after the clinical decision was made to remove the balloon catheter. We recorded global hemodynamic parameters and performed venous oximetry during maximal IABP support (1:1) and 10 minutes after temporarily stopping the IABP therapy. At both time points, we also recorded video clips of the sublingual microcirculation. From these we determined indices of microvascular perfusion including perfused vessel density (PVD) and microvascular flow index (MFI).

Results

Ceasing IABP support lowered mean arterial pressure (74 ± 8 to 71 ± 10 mmHg; P = 0.048) and increased diastolic pressure (43 ± 10 to 53 ± 9 mmHg; P = 0.0002). However, at the level of the microcirculation we found an increase of PVD of small vessels <20 μm (5.47 ± 1.76 to 6.63 ± 1.90; P = 0.0039). PVD for vessels >20 μm and MFI for both small and large vessels were unaltered. During the procedure global oxygenation parameters (ScvO2/SvO2) remained unchanged.

Conclusions

In patients deemed ready for discontinuing IABP support according to current practice, SDF imaging showed an increase of microcirculatory flow of small vessels after ceasing IABP therapy. This observation may indicate that IABP impairs microvascular perfusion in recovered patients, although this warrants confirmation.  相似文献
