目的 筛选 7,8 二羟 9,10 环氧苯并芘 (BPDE)转化细胞与正常细胞的基因表达差异。方法 以BPDE转化支气管上皮细胞 (16HBE)细胞为检测子 (tester) ,正常 16HBE细胞为驱动子 (driver) ,应用抑制性消减杂交方法构建cDNA消减文库 ,经 2次消减杂交和 2次PCR后 ,将巢式PCR产物插入TA载体克隆 ,随机挑选克隆进行鉴定和测序 ,并用斑点杂交法验证其同源性 ,将所得序列进行GenBank的BLAST分析。结果 发现了 7个在BPDE转化细胞中高表达的基因 ,为翻译延长因子 1α1、线粒体基因、溶解物载体家族 2 5、EphA2、酪氨酸 3 加单氧酶 色氨酸 - 5 -加单氧酶活化蛋白、乳酸脱氢酶A、TRIM2 8等 ,尚有一条在GenBank的已知基因中找不到对应序列 ,在EST序列中找到全长对应序列。结论 发现的高表达基因可能在BPDE的致癌机制中起作用 ,另外 ,可能有未知基因与BPDE的恶性转化作用有关。  相似文献

目的 分析鼻咽恶性淋巴瘤的误诊原因,探讨减少误诊的措施。方法对我院1990至2002年间住院治疗的120例鼻咽恶性淋巴瘤病人进行回顾性分析。结果120例鼻咽恶性淋巴瘤中有43例误诊,误诊率高达35.8%。结论鼻咽恶性淋巴瘤易发生临床误诊和病理误诊,全面掌握鼻咽恶性淋巴瘤的临床表现特点、重视鼻咽的检查和活检、注意影像学检查、结合实验室检查及免疫组化等是减少鼻咽恶性淋巴瘤误诊的有效措施。  相似文献

二甲基亚砜对培养人视网膜色素上皮细胞增殖的影响   总被引:3,自引:2,他引:3  
目的 探讨细胞分化诱导剂及端粒酶抑制剂二甲基亚砜 (dimethylsulfoxide ,DMSO)对人视网膜色素上皮细胞增殖的抑制作用。方法 本研究分实验组和对照组 ,对照组弃原培养液 ,加无血清培养液继续培养 ,实验组用不同浓度的DMSO 5、10、2 0、4 0mL·L-1分别作用于人RPE细胞 2 4、4 8、72、96h ,用MTT法检测DMSO对hRPE细胞生长的影响 ,流式细胞仪检测DMSO对hRPE细胞周期的影响。结果  10mL·L-1DMSO处理的hRPE细胞从作用 4 8h开始 ,OD值 ( 0 .5 37± 0 .0 10 )小于对照组 ( 0 .6 4 7± 0 .0 2 2 ) ,有显著性差异 (P <0 .0 0 1) ,细胞生长受到抑制 ,且随浓度增加及作用时间延长 ,抑制越显著。流式细胞仪检测 2 0mL·L-1DMSO作用 72h细胞周期发生明显变化 ,G1期细胞由4 1 31%增加至 5 9.6 5 % ,S期由 32 .0 9%减至 2 1.2 1% ,G2 期由 2 6 .6 5 %减至 19 14 % ,细胞生长被阻滞在G1期 ,生长受到抑制。结论 DMSO能阻滞hRPE细胞于G1期 ,抑制hRPE细胞增殖且呈剂量依赖性。  相似文献

The aim of this animal study was to investigate the effects of intravitreal injection of indocyanine green (ICG) applied in macular hole surgery on retinal functional, morphology, and histologic changes. Eighteen (18) New Zealand albino rabbits were divided equally into three groups (6 rabbits in each). In Group A, both eyes of each rabbit were vitrectomized by perflouropropane gas compression. One (1) month later, 0.1 cc of different doses of ICG was injected into the vitreous in the left eyes. In the right eyes, 0.1 cc of balanced salt solution was injected intravitreally, allowing them to serve as control eyes. In Group B, the same doses of ICG were injected intravitreally. ICG was washed out by fluid-fluid exchange 3 minutes after injection. In Group C, the same doses of ICG were injected intravitreally in nonvitectomized eyes. Scotopic and photopic electroretinogram (ERG) recordings and indirect ophthalmoscopy examinations were performed to detect any functional and morphologic changes. Rabbit eyes were enucleated 4 months after ICG injections to observe histologic changes. Significant decreased of scotopic and photopic ERG amplitude and marked histologic changes were noted in eyes injected with 0.5 mg/cc and 0.1 mg/cc of ICG in nonvitrectomized eyes (Group C). In vitrectomized eyes (Group A), decreased scotopic and photopic ERGs and mild histologic changes were noted in eyes injected with 0.5 mg/cc, but no histologic changes were noted in eyes injected with 0.1 mg/cc. There was a transient, mild decrease in scotopic and photopic ERGs and no morphologic changes were noted in the eyes with fluid-fluid exchange (Group B). The toxicity of intravitreous ICG is dose- and time-dependent. ICG at 0.5 mg/cc, with short exposure time, is recommended in macular hole surgery.  相似文献

Information is lacking in the literature on the precise anatomy of the iliolumbar ligament and its individual differences. The morphologic pattern, length, and width of the iliolumbar ligament were determined in 56 embalmed lumbosacral spines from human cadavers. It was possible to classify the iliolumbar ligament into two groups: Type A (74 ligaments), in which anterior and posterior ligaments had separate courses; and Type B (32 ligaments), in which anterior and posterior ligaments moved together as one band. The angle of the posterior iliolumbar ligament in Type A was oriented significantly more posteriorly than that in Type B. The posterior iliolumbar ligament was significantly shorter and oriented more posteriorly in male anatomic specimens than in female ones.  相似文献

BACKGROUND: Both static and dynamic factors are responsible for glenohumeral joint stability. We hypothesized that dynamic factors could potentially operate throughout the entire range of glenohumeral motion, although capsuloligamentous restraints (a static factor) have been thought to be primarily responsible for stability in the end-range of motion. The purpose of this study was to quantitatively compare the dynamic glenohumeral joint stability in the end-range of motion (the position of anterior instability) with that in the mid-range by investigating the force components generated by the rotator cuff muscles. METHODS: Ten fresh-frozen shoulders from human cadavera were obtained, and all soft tissues except the rotator cuff were removed. The glenohumeral capsule was resected after the rotator cuff muscles had been released from the scapula. A specially designed frame positioned the humerus in 60 degrees of abduction and 45 degrees of extension with respect to the scapula. The compressive and shear components on the glenoid were measured before and after a constant force was applied individually to each muscle with the humerus in five different positions (from neutral to 90 degrees of external rotation). The dynamic stability index, a new biomechanical parameter reflecting these force components and the concavity-compression mechanism, was calculated. The higher the dynamic stability index, the greater the dynamic glenohumeral stability. RESULTS: In the mid-range of motion, the supraspinatus and subscapularis provided higher dynamic stability indices than did the other muscles (p < 0.05). On the other hand, when the position of anterior instability was simulated in the end-range of motion, the subscapularis, infraspinatus, and teres minor provided significantly higher dynamic stability indices than did the supraspinatus (p < 0.005). CONCLUSIONS: The rotator cuff provided substantial anterior dynamic stability to the glenohumeral joint in the end-range of motion as well as in the mid-range. CLINICAL RELEVANCE: A glenohumeral joint with a lax capsule and ligaments might be stabilized dynamically in the end-range of motion if the glenoid concavity is maintained and the function of the external and internal rotators, which are efficient stabilizers in this position, is enhanced.  相似文献

目的 探索儿童鞍区肿瘤的有效手术治疗方法。方法 回顾性分析53例儿童鞍区肿瘤患者的临床资料,其中颅咽管瘤29例,生殖细胞瘤7例,垂体瘤11例,表皮囊肿2例,星形细胞瘤1例,恶性畸胎瘤1例,神经鞘瘤1例,蛛网膜囊肿1例。29例患儿行经额下硬脑膜入路,13例经改良翼点入路,11例经蝶窦入路,16例行脑室一腹腔分流术。结果 肿瘤全切除32例;次全切除16例;大部切除5例。术中22例可见垂体柄,均予保留。术后出现多饮多尿27例,其中20例在术后2周内恢复正常;出现高钠血症7例,低钠血症7例,均在2周内恢复正常,术后死亡2例。随访30例,其中3例术后1年内复发而再次手术,2例行γ-刀治疗,25例患儿术后可参加正常学习。结论 选择合适的手术入路,并注意保护下丘脑结构,防止其供血动脉损伤,是儿童鞍区肿瘤手术治疗的原则和取得较好效果的关键。  相似文献

BACKGROUND: Osteopenia of prematurity is a known source for morbidity in preterm infants. Premature infants have shown favorable outcomes in response to massage and physical activity. Whether such intervention can stimulate bone formation or decrease bone resorption is yet to be determined. OBJECTIVE: To test the hypothesis that massage combined with physical activity can stimulate bone formation and ameliorate bone resorption in premature infants. DESIGN/METHODS: A prospective double-blinded randomized trial was conducted at the Neonatal Intensive Care Unit of Ain Shams University in Cairo, Egypt. Thirty preterm infants (28 to 35 weeks' gestation) were randomly assigned to either control group (Group I, n=15) or intervention group (Group II, n=15). Infants in the intervention group received a daily protocol of combined massage and physical activity. Serum type I collagen C-terminal propeptide (PICP) and urinary pyridinoline crosslinks of collagen (Pyd) were used as indices for bone formation and resorption, respectively. PICP and Pyd were measured at enrollment and at discharge for all subjects. t-Test, ANOVA and linear regression analysis were used for statistical analyses. RESULTS: There was no difference between groups I and II in gestational age (32.1+/-1.8 vs 31.5+/-1.4 weeks) or birth weight (1.429+/-0.148 vs 1.467+/-0.132 g). In the control group, serum PICP decreased over time from 82.3+/-8.5 to 68.78+/-14.6 (p<0.01), while urinary Pyd increased from 447.7+/-282.8 to 744.9+/-373.6 (p<0.01) indicating decreased bone formation and increased bone resorption, respectively. In the intervention group, serum PICP increased over time from 62.5+/-13.8 to 73.84+/-12.9 (p<0.01). Urinary Pyd also increased over time from 445.7+/-266.5 to 716.8+/-301.8 (p<0.01). In a linear regression model including gestational age and intervention, serum PICP increased significantly in the intervention group (regression coefficient 18.8+/-4.6, p=0.0001) while urinary Pyd did not differ between groups (regression coefficient=5.6+/-114.3, p=0.961). CONCLUSIONS: A combined massage and physical activity protocol improved bone formation (PICP) but did not affect bone resorption (Pyd). Pyd increased over time in both groups, possibly due to continuous bone resorption and Ca mobilization.  相似文献

目的探讨含高单不饱和脂肪酸的特殊类型肠内营养制剂(Glucerna)对2型糖尿病患者血糖、血脂等方面的影响。方法将60例行肠内营养支持的2型糖尿病患者随机分为对照组和研究组,采用间歇喂饲法,分别经鼻胃管重力滴注两种不同的肠内营养制剂(Nutrison和Glucerna),并于营养支持前、支持后第7、14天测定两组患者的空腹血糖(FBG)、餐后2小时血糖(2hBG)、糖化血红蛋白(GHb)、血清甘油三酯(TG)、胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C)等指标。结果研究结束时,对照组FBG(7.57±2.12)mmol/L和2hBG(10.48±2.36)mmol/L比研究前略有升高,但差异不显著(P>0.05),GHb(7.47±1.22)%无明显改变(P>0.05)。研究组FBG和2hBG在营养支持后第7天已出现下降,到研究结束时分别降至(6.41±1.98)mmol/L和(9.20±2.38)mmol/L,明显低于研究前和对照组(P<0.05);GHb(7.05±0.78)%与研究前和对照组相比虽有所下降,但差异不显著(P>0.05)。整个研究期内两组患者血脂水平无明显变化(P>0.05)。结论高单不饱和脂肪酸型肠内营养制剂能够降低2型糖尿病患者的血糖水平,尤其是餐后血糖水平,在临床上比标准配方更适合糖尿病患者。  相似文献
