Glutamine (Gln) is required for intestinal mucosal homeostasis, and it can promote triglyceride absorption. The intestinal mucosal mast cells (MMCs) are activated during fat absorption. This study investigated the potential role of Gln on fat absorption-induced activation of MMCs in rats. Lymph fistula rats (n = 24) were studied after an overnight recovery with the infusion of saline only, saline plus 85 mM L-glutamine (L-Gln) or 85 mM D-glutamine (D-Gln), respectively. On the test day, rats (n = 8/group) were given an intraduodenal bolus of 20% Intralipid contained either saline only (vehicle group), 85 mM L-Gln (L-Gln group), or 85 mM D-Gln (D-Gln group). Lymph was collected hourly for up to 6 h for analyses. The results showed that intestinal lymph from rats given L-Gln had increased levels of apolipoprotein B (ApoB) and A-I (ApoA-I), concomitant with an increased spectrum of smaller chylomicron particles. Unexpectedly, L-Gln also increased levels of rat mucosal mast cell protease II (RMCPII), as well as histamine and prostaglandin D2 (PGD2) in response to dietary lipid. However, these effects were not observed in rats treated with 85 mM of the stereoisomer D-Gln. Our results showed that L-glutamine could specifically activate MMCs to degranulate and release MMC mediators to the lymph during fat absorption. This observation is potentially important clinically since L-glutamine is often used to promote gut health and repair leaky gut.  相似文献

目的观察苦参、土槿皮、黄柏、百部、黄连、虎杖、黄芩、大黄、藿香、五倍子及其混合液体外抑制念珠菌的效果。方法参照1997年美国国家临床实验标准化委员会M27-A方案中的酵母菌微量稀释法进行药敏试验。结果同一种中药水煎剂及不同的单味中药水煎剂对不同的念珠菌标准菌株的最低抑菌浓度(minimuminhibitory concentration,MIC)各不相同,中药混合液对5种标准菌株的MIC最低,均≤0.031 25g/ml。结论不同的中药对不同的念珠菌的抑制作用各不相同,复方对念珠菌的抑制作用优于单味中药。  相似文献

Persistent generalized low bone mineral density (BMD) has been reported in patients with adolescent idiopathic scoliosis (AIS).However,the exact mechanisms and causes of the low BMD in AIS patients are largely unknown.The purpose of this study was to examine the relationship between the receptor activator of NF-κB ligand (RANKL)/osteoprotegerin (OPG) levels in osteoblasts (OBs) from AIS patients with low BMD and with comparison made between the patients and controls.Twenty AIS patients and eight age-matched controls were included in the present study.The BMD of lumbar spine and proximal femur was measured in all subjects.OBs from the cancellous bone of each subject was harvested and primarily cultured.The mRNA and protein expression of RANKL and OPG in OBs was detected by RT-PCR and Western blotting.The results showed BMD was lower in AIS patients than in controls.A significantly higher mRNA and protein expression of RANKL was observed in OBs from AIS patients,while no significant difference was found in the expression of OPG between AIS patients and controls.As a result,RANKL/OPG ratio in patients with AIS was remarkably higher than controls.Our study preliminarily demonstrated expression of RANKL was higher in OBs from AIS patients with low BMD as compared with controls,suggesting the unbalanced RANKL/OPG ratio caused by an over-expression of RANKL in OBs may be responsible for the low BMD in AIS patients.  相似文献

检测人表皮生长因子(hEGF)在胃癌组织中的含量,初步探讨其临床意义。用放射免疫分析法测定52例胃癌切除标本和18例正常胃粘膜组织中hEGF的含量。结果:对照组为1.49±0.95ng/g湿重,胃癌组织为4.29±2.35ng/g湿重,差异显著(P<0.01)。在胃癌组织中,hEGF含量进展期高于早期,弥漫型高于肠型,淋巴结转移阳性者高于阴性者,近期随访发现胃癌患者死亡组的hEGF含量较高。结论:胃癌组织可产生过量的hEGF,并与肿瘤浸润、组织分化程度、淋巴结转移等相关,提示胃癌组织hEGF测定可作为判断胃癌生物学行为及了解患者预后的指标。  相似文献

目的:探讨血清白细胞介素-13(IL-13)与肺功能检测在儿童毛细支气管炎的临床意义。方法:采用双抗体夹心酶联免疫吸附试验(ELISA)对15例毛细支气管炎急性期、10例恢复期患儿外周血行血清IL-13水平测定;并与10例健康对照组儿童比较。结果:毛细支气管炎恢复期组血清IL-13水平较急性期组明显下降(P〈0.01),但两组均明显高于对照组(P均〈0.01);毛细支气管炎恢复期组肺功能指标中呼出75%潮气量时的呼气流速/潮气呼气峰流速(25/PF)及潮气呼气中期流速/潮气吸气中期流速(ME/MI)均较急性期组明显升高(P均〈0.01),但25/PF仍低于对照组(P〈0.01),而ME/MI与对照组比较无显著性差异(P〉0.05)。毛细支气管炎急性期组血清IL-13水平与25/PF呈显著负相关(r=0.38,P〈0.05),与ME/MI无相关性(r=-0.002,P〉0.05)。结论:IL-13参与毛细支气管炎发病过程,降低其水平有助于毛细支气管炎的痊愈;毛细支气管炎患儿急性期肺功能主要表现为小呼吸道阻塞性通气功能障碍。  相似文献

目的:通过<公共关系学>课程的教学,提高大学生的公关意识和能力.方法:在3所大学采用理论教学、课堂讨论、无记名问卷调查等方式开展教学.结果:405名学生的问卷调查显示:选择<公共关系学>教学能"逐步树立公关意识、形象意识"者占95.1%(385/405);能"提高学生综合素质"者占93.1%(377/405);能"提高学生的观察反应能力"者占82.5%(334/405);能"提高学生的自控应变能力"者占78.3%(317/405).结论:公关理念能增强公关意识,主动塑造自身良好社会形象;能培养公关技能,提高大学生的社会适应能力.  相似文献

Objective:The aims of this study were to examine the prognostic value of SHP-1 in breast cancer, its roles in the regulation of breast cancer cell growth and metastasis, and the underlying mechanisms.Methods:Tumor specimens from 160 patients with breast cancer and 160 noncancerous tissues were used to examine the expression of SHP-1 and to analyze its association with overall survival through Kaplan–Meier and multivariate Cox regression analyses. RNA sequencing data and the expression and clinical importance of SHP-1 in breast cancer were evaluated with data from The Cancer Genome Atlas. In vitro and in vivo assays were performed to elucidate the effects of SHP-1 on breast cancer cell proliferation and invasion. Confocal immunofluorescence and GST pulldown assays were used to demonstrate the interaction between SHP-1 and epidermal growth factor receptor, as well as its downstream pathways. Immunohistochemistry and The Cancer Genome Atlas database were used to investigate the clinical association between SHP-1 and EGFR in human breast cancer.Results:SHP-1 expression was associated with better survival in patients with breast cancer, whereas SHP-1 expression was negatively correlated with EGFR in human breast cancer. Ectopic SHP-1 expression significantly suppressed breast cancer cell proliferation, migration, and invasion. SHP-1 knockdown induced a more invasive phenotype and accelerated cell growth. Mechanistically, EGFR, a protein directly interacting with SHP-1, mediates the SHP-1-induced inactivation of Ras/Erk/GSK3β signaling and its downstream effectors.Conclusions:SHP-1 is an important prognostic biomarker in patients with breast cancer, and the SHP-1-EGFR axis is a promising target for treatment.  相似文献
