Purpose

We investigated the local contribution of nasopharyngeal epithelial cancer cells to the inflammatory process.

Materials and methods

THP-1 monocytes were treated with phorbol 12-myristate 13-acetate to induce the production of differentiated macrophages (D-THP-1), which were subsequently activated by lipopolysaccharide (LPS) (10?ng/ml). The production of pro-inflammatory cytokines in D-THP-1 cells was detected by ELISA and qRT-PCR. The effects of conditioned media harvested from LPS-treated D-THP-1 cells were investigated with regard to cell proliferation (MTT), production of pro-inflammatory cytokines (ELISA) and activation of NF-??B and STAT3 (western blot) in the nasopharyngeal epithelial cancer cell line 5?C8F.

Results

LPS induced the production of the pro-inflammatory cytokines TNF-?? (875.1?±?68.31?pg/ml), IL-6 (42.2?±?5.32?pg/ml), IL-1?? (9.6?±?1.34?pg/ml) and IL-8 (19.3?±?3.47?pg/ml) in D-THP-1 cells significantly (P?P?P?P?Conclusions Nasopharyngeal epithelial cancer cells may play a significant role in maintaining and amplifying the inflammation process via activation of NF-??B and STAT3 pathway and through the local production of pro-inflammatory cytokines, which recruit and activate additional immune cells in the nasopharyngeal path and promote tumour progression.  相似文献

Abstract: Melatonin is an important natural oncostatic agent, and our previous studies have found its inhibitory action on tumor angiogenesis, but the mechanism remains unclear. It is well known that vascular endothelial growth factor (VEGF) plays key roles in tumor angiogenesis and has become an important target for antitumor therapy. Pancreatic cancer is a representative of the most highly vascularized and angiogenic solid tumors, which responds poorly to chemotherapy and radiation. Thus, seeking new treatment strategies targeting which have anti‐angiogenic capability is urgent in clinical practice. In this study, a co‐culture system between human umbilical vein endothelial cells (HUVECs) and pancreatic carcinoma cells (PANC‐1) was used to investigate the direct effect of melatonin on the tumor angiogenesis and its possible action on VEGF expression. We found HUVECs exhibited an increased cell proliferation and cell migration when co‐cultured with PANC‐1 cells, but the process was prevented when melatonin added to the incubation medium. Melatonin at concentrations of 1 μm and 1 mm inhibited the cell proliferation and migration of HUVECs and also decreased both the VEGF protein secreted to the cultured medium and the protein produced by the PANC‐1 cells. In addition, the VEGF mRNA expression was also down‐regulated by melatonin. Taken together, our present study shows that melatonin at pharmacological concentrations inhibited the elevated cell proliferation and cell migration of HUVECs stimulated by co‐culturing them with PANC‐1 cells; this was associated with a suppression of VEGF expression in PANC‐1 cells.  相似文献

目的探讨质子泵抑制剂对胃食管反流性咽喉炎的治疗作用及其安全性,为胃食管反流性咽喉炎的临床治疗提供指导依据。方法计算机检索PubMed、CENTRAL、EMbase、the ISI Web of Knowledge databases、VIP、CNKI、CBM和WANFANG数据库,查找所有探讨质子泵抑制剂对胃食管反流性咽喉炎治疗作用的随机对照试验,检索时限均为建库至2012年3月2日。同时手检纳入文献的参考文献。按纳入排除标准由3名研究者独立进行RCT的筛选、资料提取和质量评价后,采用RevMan 5.1软件进行Meta分析,并采用GRADE系统进行证据质量评价。结果共纳入8个研究,591例患者。Meta分析结果显示:质子泵抑制剂治疗胃食管反流性咽喉炎的总体有效率[OR=7.79,95%CI(2.20,27.51),P<0.05]高于对照组,咳嗽症状缓解效率[OR=2.29,95%CI(0.83,6.35),P>0.05],与对照组比较差异不显著,内镜检查好转率[OR=18.83,95%CI(1.86,190.60),P<0.05],高于对照组。结论质子泵抑制剂对胃食管反流性咽喉炎的治疗作用较对照组明显;但因纳入研究较少、质量参差不齐、样本含量小、各研究方法变异性大,建议临床上审慎选择使用;需要更多高质量、大样本、多中心的随机对照试验进一步论证。  相似文献

目的分析中国18家医院的14万例年龄相关性白内障患者角膜前表面散光的分布特征。方法回顾性系列病例研究。连续性收集2015年7月至2018年10月于中国18家爱尔眼科医院就诊的40岁以上年龄相关性白内障患者143889例(143889只右眼)的眼部生物学参数资料。角膜前表面散光度数和轴向、前房深度、角膜屈光力、眼轴长度等眼球参数采用IOLMaster 500测量,获取3次测量结果的平均值。各医院将资料整理分析后提交给武汉爱尔眼科医院进行总体分析。非正态分布数据以M(P25~P75)表示;采用Mann-Whitney检验、Kruskal-Wallis检验、χ2检验等分析角膜前表面散光度数和轴向在不同性别、年龄、前房深度、角膜屈光力、眼轴长度中的分布差异。结果143889例患者中女性84319例,男性59570例;年龄为72(65~78)岁;角膜散光度数为0.84(0.51~1.33)D,散光度数≥0.75 D者80895例(56.22%),散光度数≥1.00 D者57304例(39.83%)。女性角膜散光度数为0.87(0.53~1.37)D,男性为0.82(0.50~1.29)D(U=-14.891);女性顺规散光比例为33.26%(28046/84319),逆规散光比例为49.08%(41385/84319);男性顺规散光比例为34.26%(20408/59570),逆规散光比例为46.91%(27945/59570)(χ2=70.913),差异均有统计学意义(均P<0.05)。随年龄增加,角膜散光度数先由0.94(0.57~1.48)D减少至0.75(0.46~1.18)D,后又增大至1.19(0.74~1.79)D,差异有统计学意义(H=1263.438,P<0.05),变化的转折在61~70岁。随着年龄的增大,顺规散光比例减小[由77.50%(396/511)减少至12.50%(3/24)],逆规散光比例增大[由11.15%(57/511)增大至79.07%(34/43)],斜向散光比例变化不大[17.02%(16/94)至19.92%(245/1230)],分布差异有统计学意义(χ2=10174.496,P<0.05)。前房越浅,角膜散光度数越大,由0.82(0.51~1.31)D增大至1.05(0.61~1.56)D;逆规散光比例越大,由47.32%(60207/127227)增大至51.69%(184/356),差异均有统计学意义(H=409.961,χ2=120.995;均P<0.05)。平均角膜屈光力越大,角膜散光度数越大,由0.80(0.49~1.33)D增大至0.95(0.58~1.53)D;逆规散光比例越小,由52.84%(4963/9392)减小至39.97%(9023/22577),差异均有统计学意义(H=808.562,χ2=752.147;均P<0.05)。不同眼轴长度相比,当眼轴长度>25.00 mm时,角膜散光度数最大,为1.04(0.62~1.65)D;逆规散光比例最大,为49.00%(10964/22376),差异均有统计学意义(H=2071.198,χ2=131.130;均P<0.05)。结论年龄相关性白内障患者角膜前表面散光轴向以逆规散光为主。随着年龄的增大,角膜散光度数有先减小后增加的趋势。65岁为顺规散光向逆规散光变化的转折点。前房越浅,角膜前表面散光度数以及逆规散光比例越大。当眼轴长度>25.00 mm时,角膜前表面散光度数和逆规散光比例最大。  相似文献

探讨肺表面活性物质(PS)对新生儿重症肺炎患儿肺部氧合情况及血气指标的影响。选取新生儿重症肺炎患儿62例,随机分为治疗组和对照组,各31例。所有患儿均进行常规治疗,接呼吸机,治疗组患儿肺部接受PS治疗。比较2组患儿治疗前后肺部氧合情况,对治疗组PS治疗前后血气指标进行分析,并统计2组平均机械通气时间、氧暴露时间及死亡病例和原因。治疗后10 min、30 min、60 min时2组患儿血氧饱和度(SpO2)均较治疗前升高,且治疗组明显高于对照组(P<0.01);治疗后24 h、48 h、72 h时2组患儿氧合指数(OI)均降低,且治疗组明显低于对照组(P<0.01);治疗组患儿经PS治疗后30 min、60 min时血氧分压/吸入氧浓度比值(PO2/FiO2)及动脉/肺泡氧分压比值(a/APO2)均呈先升高后降低趋势,但均明显高于治疗前(P<0.01);治疗组平均机械通气时间和平均氧暴露时间均明显低于对照组(P<0.01);2组病死率比较有显著性差异(P<0.05)。PS可以显著改善新生儿重症肺炎患儿肺部氧合状态及低氧血症,值得临床推广使用。  相似文献

Recycling of reclaimed asphalt pavement (RAP) has gradually emerged as a hot topic in the current research of building materials. Manufacturing the recycled asphalt mixture with a high RAP content still remains a major challenge due to the problem of inferior water damage resistance and fatigue cracking resistance. The aim of this study is to evaluate the servicing performance of recycled asphalt mixture with high percentage of RAP and assess its environmental impact on volatile organic compounds (VOCs) emission. To further explore the intrinsic factor on the performance of a recycled asphalt mixture, the mixed asphalt binder with a different content of extracted RAP binder was firstly investigated. The results show that the larger the content of RAP binder, the lower the mechanical indexes and VOCs emission. There exists an internal relationship between the chemical composition and VOCs release behavior with the variation of the recycled asphalt binder content. Based on the results of mixed recycled asphalt binder, the road servicing performance of a rather high utilization of RAP (i.e., 50%, 60%, and 70%) was assessed. It was found that the reuse of RAP aggregates has little influence on the volume performance of recycled asphalt mixture, and servicing performances still meet the construction requirements in spite of a descending trend. Moreover, a significant reduction effect on VOCs emission was found in the mixing stage of recycled asphalt mixture, indicating that the VOCs emission can be decreased by 94.82% when the content of RAP aggregates increases to 70%. The recycling of RAP with a high content contributes to the sustainable development of road engineering and the construction of green pavements.  相似文献

目的 运用精益管理(lean management,LM)理论,对出院带药进行流程再造。方法 通过使用价值流图(Value Stream Mapping)和7种浪费(Seven Wastes)2种LM工具对原出院带药流程进行分析,通过多部门协作对流程进行再造。结果 新流程的实施减少了出院带药流通环节,降低出错机率,保证了患者用药安全;减轻了医护人员的工作强度,减少了人力资源成本,缩短了患者的出院时间,提高了患者和医务人员的满意度,并让患者享受到药师专业的用药指导服务。结论 新流程提供了高品质、方便快捷的出院带药服务,保证了患者用药安全。  相似文献
