Characterization of the processed form of a ubiquitous protein displaying a variable membrane organization in erythroid cells

We recently identified a group of proteins that are present in all hematopoietic cells but are organized in the cell membrane of erythrocytes in a lineage-specific fashion (Blood 61:803, 1983). One of these polypeptides has a mol wt of approximately 37,000 (p37T) when translated in vitro from messenger RNA (mRNA) extracted from the erythroleukemic K562 cell line. The membrane-associated form of the p37 translation product has been analyzed in detail here. When detergent lysates prepared from biosynthetically labeled K562 cells were reacted with an antiserum containing anti-p37T antibodies, one of the proteins immunoprecipitated had a nominal mol wt of 36,000 to 37,000 (p37M). Several results suggest that this protein is homologous to the p37 translation product: (1) the protein, like the mRNA coding for the p37 translation product, was expressed in cell lines with diverse differentiated phenotypes; (2) the antigenic determinant(s) on p37M and p37T are oriented to the inner surface of the erythrocyte membrane while being oriented to the outer surface of erythroleukemic cells; and (3) one-dimensional peptide maps show homology between p37M and p37T. P37M does not appear to possess an N-terminal leader sequence that is proteolytically cleaved as the molecule is inserted into the membrane. In addition, p37M is not glycosylated.     

Seroprevalence of syphilis amongst pregnant women attending antenatal clinics in a rural hospital in north west Ethiopia.

OBJECTIVE--To determine the seroprevalence of syphilis in pregnant women attending antenatal care clinics in a rural hospital in Ethiopia and describe their characteristics so that timely and effective care can be offered. SUBJECTS--Pregnant women attending antenatal care clinics at a rural hospital. METHODS--A cross sectional study was conducted at a rural hospital in north west Ethiopia in September 1994. Data were collected on socio demographic characteristics and past history of sexually transmitted diseases of antenatal care attenders from the records of the hospital. Sera from these pregnant women were examined for syphilis using the VDRL test. VDRL positivity was cross tabulated with socio demographic variables and past history of sexually transmitted diseases. RESULTS--Two hundred and seventy pregnant women were included in the study. Median age of the respondents was 25.2 years. A substantial majority were orthodox Christians (97.4%) and married (92.6%). Town dwellers constituted 58.2% of the antenatal care attenders. Only 4.7% of the women started attending antenatal care clinics during the first trimester. Thirty seven women (13.7%) were found to be VDRL positive. Past history of sexually transmitted diseases was significantly associated with VDRL positivity (p < 0.05). CONCLUSIONS--High VDRL positivity rate is observed in this study. Certain risk factors may be responsible for current and past episodes of sexually transmitted diseases. There is a need for improvement of antenatal care activities at different levels of health care. Appropriate strategies should be devised for prevention and control of sexually transmitted diseases in women of reproductive age groups and the general population.     

Adventitial cystic disease of the popliteal artery

In a 27-year-old man with claudication, computed tomography (CT) demonstrated a popliteal cyst and an adventitial cyst of the popliteal artery. Adventitial cystic disease was unexpected because the patient had a diminished pedal pulse on plantar flexion of the foot, a physical finding suggesting popliteal artery entrapment syndrome. CT was useful in the initial examination of this young patient with suspected popliteal vascular disease because of its accuracy and noninvasiveness.
