人毛乳头细胞凝集性生长差异表达基因cDNA文库的构建

目的 构建毛乳头细胞凝集性生长状态下差异表达基因的消减cDNA文库。方法 分别提取凝集性生长和非凝集性生长状态下毛乳头细胞中的总RNA，应用SMART cDNA合成技术和抑制性消减杂交技术分离毛乳头细胞凝集性生长状态下差异表达基因的cDNA片段并建立消减cDNA文库；利用PCR对随机挑选的100个白色菌落进行插入片段的验证，对其中证实有插入片段的30个克隆进行cDNA斑点杂交验证。结果 所构建的消减cDNA文库扩增后得到320个阳性克隆，随机挑选的100个阳性克隆中95％的均有长度在100—600bp的插入片段，cDNA斑点杂交验证显示27个(90．0％)克隆为阳性。结论 结合SMART cDNA合成技术，应用抑制性消减杂交成功地构建了毛乳头细胞凝集性生长差异表达基因消减cDNA文库，为进一步筛选和克隆毛乳头细胞凝集性生长相关基因奠定了基础。     

Inhibition on Telomerase Activity and Cytotoxic Effects by Cisplatin in Cultured Human Choroidal Melanoma Cells

Purpose: To study the changes of telomerase activity and cytotoxic effects by Cisplatin; cis-dichlorodiamine platinum (CDDP) in cultured human choroidal melanoma. Material and Methods: The primary cultured human choroidal melanoma cells were cultured in the presence and absence of CDDP with different concentration and time respectively. The toxic effects were evaluated by MTT and the level of telormarse was detected by PCR-ELISA assay. And the relationship between telomerase activity and cytotoxic effects were analyzed by a correlation analysis.Results: Following the increase of the concentration and the time of CDDP, gradually repressed telomerase activity was detected in cultured cells. Meanwhile, the restrain rate of the cells increased. The telomerase activity at 24h and 1μg/ml was repressed significantly compared with the control cells. However, the appearance of cell death lagged behind the decreasing of telomerase.Conclusions: CDDP is an effective telomerase inhibitor in cultured choroidal melan     

硅胶管套接法治疗陈旧性周围神经损伤

我院自1998年6月-2000年9月间,用硅胶管套接法治疗10例陈旧性周围神经损伤患者,取得满意疗效,现报告如下.……     

眼内异物合并外伤性白内障的联合手术治疗：附88例分析

眼内异物合并外伤性白内障是眼外伤常见的致盲原因之一,各地报告的致盲率为20%～48.9%。本文对88例眼内异物合并外伤性白内障患者采用联合手术治疗,收到较好疗效,术前视力0.05以下占85.2%,术后矫正视力0.05以下占27.3%,矫正视力0.06～1.5占73.7%,其中矫正视力0.6～1.5者占22.8%。根据异物的性质、在眼内部位和晶体损伤情况选择不同的手术方式一次联合手术摘出异物和白内障,术后采用中西药配合治疗。联合手术可提高疗效,减少术后并发症.减少手术次数,减轻病人的痛苦,缩短病程,减少医疗费用.眼科学报1993;9:216—218.     

Psychoactive substance use in three sites in China: gender differences and related factors

One year prevalence rates far psychoactive substance use are reported based on a community survey in Hunan, Helongjiang and Jiangsu. Data were gathered on more than 14 000 respondents (15–65 years old). The drinking rates ranged from 58.3% to 82.6% for men and from 16.3% to 31.4% for women. Smoking rates ranged from 64.9% to 68.1% for men and from 0.1% to 20.5% for women. Most drinkers were light users and most smokers were heavy users, Illicit drug use was found in the Hunan site. For both frequency and quantity of psychoactive substance use, women are lighter consumers than males (except for the use of minor tranquilizers and analgesics). Psychosocial factors related to drinking and smoking are also investigated in this study.     

LncRNA-MAFG-AS1在结直肠癌中的表达及临床意义

目的 探讨LncRNA-MAFG-AS1在结直肠癌中的表达及临床意义。方法 选取我院行手术切除的结直肠癌组织及对应的癌旁正常组织各80例,应用定量即时聚合酶链锁反应(quantitative real time polymerase chain reaction, qRT-PCR)检测结直肠癌组织及癌旁正常组织中LncRNA-MAFG-AS1的表达水平,并分析其与结直肠癌临床病理特征之间的关系,所有患者随访截止日期到2019年12月31日,应用Kaplan-Meier(K-M)法进行生存分析,采用Cox回归模型分析结直肠癌预后的危险因素。结果 结直肠癌组织中LncRNA-MAFG-AS1相对表达量为2.398±0.214,高于癌旁正常组织的1.032±0.132,差异有统计学意义(P<0.05)。当患者TNM分期更高、中高分化及存在淋巴结转移和远期转移时,LncRNA-MAFG-AS1过表达(P<0.05)。K-M生存分析显示高表达患者无进展生存时间(progression-free survival, PFS)为(20.36±3.21)个月,低表达患者为(29.71±4....     

超声心动图和下肢静脉超声联合血浆D-二聚体对孕产妇肺栓塞的临床诊断价值

目的分析和评价超声心动图和下肢静脉超声联合血浆D-二聚体对孕产妇肺栓塞（pulmonary thromboembolism, PE）的临床诊断和预测价值。方法回顾性收集2019年5月—2021年5月同济大学附属第一妇婴保健院收治120例临床疑诊PE的孕产妇临床资料,按照肺动脉断层血管造影（computed tomographic pulmonary angiographic, CTPA）的结果分为阴性组和阳性组,比较和评价超声心动图、下肢静脉超声、血浆D-二聚体单独及联合诊断孕产妇PE的临床诊断和预测效能。结果临床特征比较： 两组间年龄、孕次、产次、受孕方式、受孕胎数、终止妊娠方式、是否早产、既往妇产科手术史、既往外科手术史、是否合并高血压、糖尿病、肥胖及BNP等指标比较差异均无统计学意义（均P>0.05）。两组间仅血浆D-二聚体比较差异有统计学意义（t=-3.598,P＜0.001）。超声心动图、下肢静脉超声和血浆D-二聚体升高单独及联合诊断孕产妇PE的灵敏度分别为45.00%、53.33%、95.00%和85.00%,特异度分别为83.33%、91.67%、21.67%和81.67%,阳性预测值分别为72.97%、86.49%、54.81%和82.26%,阴性预测值分别为60.24%、66.27%、81.25%和84.48%。与单独诊断相比较,联合诊断显著提高灵敏度（χ2=49.669,P＜0.001）、特异度（χ2=88.413,P＜0.001）、阳性预测值（χ2=20.629,P＜0.001）和阴性预测值（χ2=11.169,P=0.01）。多因素Logistic回归分析提示超声心动图阳性（OR=18.266,P＜0.001）、下肢静脉血栓（OR=45.482,P＜0.001）及D-二聚体升高（OR=6.595,P=0.019）与孕产妇PE发生显著相关,ROC曲线分析显示单独及联合诊断的曲线下面积（AUC）分别为0.642、0.725、0.583和0.833。结论超声心动图和下肢静脉超声联合血浆D-二聚体对孕产妇PE具有良好的诊断和预测价值,为临床早期预防、诊断和治疗提供可靠依据。     

调节性B淋巴细胞在视神经脊髓炎谱系疾病患者外周血中的表达和意义

背景 调节性B淋巴细胞(Breg)是一类具有负向免疫调节作用的细胞,在多种自身免疫性疾病中表达异常,视神经脊髓炎谱系疾病(NMOSD)系自身免疫性疾病,目前关于Breg在NMOSD患者外周血中的表达和意义的研究较少。目的 研究Breg在NMOSD患者外周血中的表达和意义。方法 选取2019年6月—2021年12月新疆医科大学第一附属医院神经内科收治的NMOSD患者55例作为NMOSD组,选取同期本院体检正常的健康志愿者50例作为对照组。运用流式细胞仪检测两组研究对象外周血中CD19⁺CD24^hiCD38^hi Breg、CD19⁺CD24^hiCD27⁺Breg的表达。采用酶联免疫吸附测定(ELISA)法检测和比较两组研究对象血清中白介素10(IL-10)、白介素35(IL-35)和转化生长因子β1(TGF-β1)的水平。结果 NMOSD组患者CD19⁺CD24^hiCD38^hi Breg、CD1...     

康复新液联合奥美拉唑对HP阴性胃溃疡的疗效分析

目的分析康复新液联合奥美拉唑治疗幽门螺杆菌(HP)阴性胃溃疡患者的疗效。 方法选取HP阴性胃溃疡患者108例,随机均分为对照组和联合组。对照组接受奥美拉唑针剂治疗,联合组接受康复新液联合奥美拉唑针剂治疗。比较两组临床疗效、血清胃蛋白酶原(PG)、高迁移率族蛋白B1(HMGB1)、细胞炎症因子水平及不良反应发生率。 结果联合组治疗总有效率高于对照组(P＜0.05)。与治疗前比较,两组治疗后PGⅠ、PGⅡ、HMGB1、白细胞介素-6、肿瘤坏死因子-α及C反应蛋白均有降低,且联合组较对照组降低更明显(P＜0.05)。两组患者治疗期间总不良反应发生率比较,差异无统计学意义(P＞0.05)。 结论康复新液联合奥美拉唑针剂可提高HP阴性胃溃疡患者的临床疗效,值得临床应用推广。     

幽门螺杆菌对SGC-7901细胞增殖凋亡及p27kipl表达的影响

Objective To explore the influence of Helicobacter pylori (Hp) on the proliferation, apoptosis and expression of p27kipl protein of gastric epithelial cells in vitro. Methods SC, C-7901 cell pro-liferation was examined by flow cytometry after incubation with different concentration of NCTC11637. The effect of NCTC11637 on cell apoptosis was also evaluated by flow cytometryo And the expression of p27kipl of gastric epithelial cells was detected by immunohistochemical analysis and in situ hybridization, respectively. Results Cell proliferation was enhanced when co-incubated with the Hp in low concentration (3.4 x 104 to 1.9 x 105 CFU/ml) but inhibited in higher concentration (≥4.8×106CFU/ml). However, cell apoptosis was increase when co-incubated with the Hp in high concentration (≥9.6 ×105 CFU/ml) showing concen-tration dependent picture. In addition, the co-incubation of SGC-7901 with Hp led to decrease of the expres-sion of p27kipl protein but not mRNA in a Hp concentration dependent way. Conclusion Hp could effect the gastric epithelial cells apoptosis and proliferation directly and influence the expression of p27kips protein which might facilitate gastric carcinoma.
