Effects of anti-CXCR4 monoclonal antibody 12G5 on proliferation and apoptosis of human acute myelocytic leukemia cell line HL-60

Objective:To investigate the expression of CXCR, on HL-60 cell line and the proliferation, apoptosis of HL-60 cell line cocultured with bone marrow stromal cells, so as to assess the possibility of 12G5. an anti-CXCR4 monoclonal antibody, in eradicating the minimal residual disease. Methods:The activity of SDF-1 was inhibited by 10μg/ml 12G5. After treatment with 12G5. the status of adhesion was observed, and the adhesion rates, apoptosis and cell cycles were detected after 24 h of treatment. Cell growth rates were measured by trypan blue exclusion. Cell growth curve was plotted, and the expression of PCNA and apoptosis related protein including PCNA, Bcl-2 and Fas were detected with immunohis-tochemical technique. Results :(1) There was middling degree expression of CXCR4 on HL-60 membrane. From 0 h to 6 h, as the time of 12G5 incubation along, the expression of CXCR4 decreased gradually. (2) After treatment for 24 h, the adhesion rates in the experiment group and the control were (39. 4±7. 9)% and (51. 4±5. 9)%, respectively. (3)After treatment for 24 h, the percentage of HL-60 cells in G0/G1 phase were (55. 21±4. 9)%, and that in S phase and G2/M phase were (30. 40±4. 1)% and (14. 39±5.2)%, respectively, with the corresponding proportions being (44. 67±2. 2) % , (45. 30±3. 7)% . and (10. 03±2. 6)% in the control. (4) The percentage of apoptotic HL-60 cells was (8. 95±1. 7)% in the experiment group, compared to (3. 97±2. 4)% in the control. (5)The survival rates of HL-60 cells decreased markedly at 48 h to 96 h, and the proliferation slowed down at this time duration. (6)The expression of PCNA and Bcl-2 down-regulated significantly, but the Fas protein expression was up-regulated. Conclusion :12G5 could inhibit the capability of adhesion and proliferation of HL-60 cells and it can induce more cells to enter G0/G1 phase and promote apoptosis. It may be helpful by inhibiting the bioactivity of SDF-1 with 12G5 in the therapy of marrow residual disease.     

PRIMARY STUDY OF REPAIRING PERIPHERAL NERVE GAP WITH PORCINE SMALL INTESTINAL SUBMUCOSA

Objective To investigate the role of porcine small intestinal submucosa （SIS） conduit in axonal regeneration of rat sciatic nerve with a 10 mm gap. Methods Forty-eight rats were randomly divided into three groups （n=16）. Following a 10 mm gap was made in one side of the sciatic nerve of each rat; previously prepared SIS and auto-nerve graft were interposed into the gap to reconnect the proximal and distal ends of the nerve, respectively. In the control group, the nerve gap remained unconnected. The samples of the SIS, graft, and distal nerve in group 1 and group 2 were harvested at 6 weeks and 10 weeks after operation, respectively. Axonal regeneration was evaluated by histology, electrophysiology, and quantitated by using computer-analyzed image.Results Regenerative nerve fibers were evident which contained much myelinated axons and grew over the gap in the SIS conduits at 10 weeks. Electrophysiological examination and computer-analyzed image showed that axonal regeneration in the SIS group was similar to that in the auto-nerve grafting group at 10 weeks. Conclusion SIS as a conduit possesses the ability for axonal regeneration of the peripheral nerve, thereby having a potential to be an alternative bio-material instead of the autograft to repair the peripheral nerve gap.     

维持性血液透析患者的营养需求

随着透析技术的不断发展，透析疗治已逐步成为治疗终末期肾脏疾病的最主要方法。我困每年终末期肾脏病的发病率达1／100000，其中血液透析患者占92％，且年增长率高达18．7‰，透析患者往往存在营养不良的问题，而且显得日趋突出。这不仅影响血液透析患者的预后和生活质量，而且也是并发症和病死率增加的一个重要因素，所以要改善患者的营养状况和提高患者的生活质量，需要医护人员、患者和家庭的共同努力，更多地了解掌握营养需求的有关知识。     

浅谈大学生英语自主学习能力的提高

大学生学习英语的最终目的是能够运用所学语言。满足工作、生活中听、说、读、译等方面多层次的需要。学生这些英语能力的获得必须通过大量的训练．这需要大学生进行英语自主性学习，而英语学习是一个长期的过程，英语自主学习能力的培养对他们以后英语水平的提高和成功的职业发展有着深远的影响。本文在自主学习理论的基础上，设计、发放调查问卷．通过对调查问卷的分析总结，根据调查结果提出培养学生的自主学习能力的方法。     

鞘内注射胶质细胞代谢抑制剂对背根节压迫大鼠痛阈的影响

目的研究鞘内注射（i．t．）氟代柠檬酸（FC）和米诺四环素（MC）对背根节慢性压迫模型（CCD）大鼠痛阈的影响。方法采用大鼠CCD模型，48只SD大鼠鞘内置管，随机分为6组（n=8），分别为：sham组、CCD组、PBS组（0．01mmol／L PBS 20ul，i．t．）、FC组（1umol/L FC20ul，i．t．）、MC组（5g／L MC20ul，i．t．）和MC＋FC组（终浓度1umol／L FC、5g／LMC混合液20ul，i．t．）。CCD术后每天给药1次，分别于术前（基础值）及术后1、3、5、7、9、11、13、14天采用Von—Frey filaments法和热辐射法评定大鼠机械刺激缩足反射阈值（PWMT）和热刺激缩足反射潜伏期（PWTL）。结果与基础痛阈值相比，除sham组第1天有明显下降后恢复外，其余各组大鼠术后的PWMT及PWTL均出现明显下降（P〈0．05）。CCD术后第3天开始，FC、MC、MC＋Fc组与CCD、PBS组相比，大鼠PWMT及PWTL显著升高（P〈0．05），FC组低于MC、MC＋FC组而高于CCD、PBS组（P〈0．05）。结论CCD大鼠鞘内注入本实验剂量胶质细胞代谢抑制剂后，小胶质细胞抑制剂米诺四环素比星形胶质细胞抑制剂氟代柠檬酸更好地抑制了大鼠的慢性疼痛。     

丹参对维持性血液透析患者AOPP、MDA和SOD的干预作用

目的研究透析和丹参对维持性血液透析（MHD）患者氧化应激指标的影响。方法36例MHD半年以上的患者随机分为常规透析组和透析加丹参组，同时18例健康成人作为正常对照组。分别测定各组基础以及透析组1、2和3个月的血浆氧化应激指标（AOPP、MDA和SOD）的水平。结果透析患者的AOPP、MDA明显高于正常对照组（P〈0．01）、SOD显著低于正常对照组（P〈0．01）。常规透析组3个月时AOPP、MDA较服药前增加（P〈0．05），SOD降低（P〈0．05）。丹参组在1、2和3个月时所测MDA、AOPP明显低于服药前以及同时点常规透析组（P〈0．01），而SOD则显著增高（P〈0．01），但3个月时AOPP、MDA仍高于正常对照组（P〈0．01），SOD低于正常对照组（P〈0．01）。结论丹参能有效地改善MHD患者的氧化应激，血液透析可能加重MHD患者的氧化应激。

原发性小血管炎并发自发性肾破裂1例

1病例报告患者男,72岁,因“眼睑浮肿10天,少尿4天”于2006年5月30日入院。患者不明原因双眼睑浮肿伴疲倦、乏力,4天前尿量减少至少尿、无尿,伴活动后心慌、气紧。2月前因“眼红”在当地县医院诊断为“结膜炎”。入院查体:T36.5℃、BP160/100mmHg,精神萎靡,全身皮肤呈多形红斑,颜面及眼睑轻度浮肿,球结膜充血,双肺呼吸音粗糙,无干、湿性啰音,心界不大,心率100次/分,无杂音,肝、脾肋下未及,双肾区轻叩痛,双下肢Ⅰ度凹陷性水肿。实验室检查:血常规WBC9.84×109/L、Hb88g/L、PLT230×109/L;尿常规Pro( )、WBC13个/HP、RBC18个/HP;血清…

输精管粘堵剂的毒理学研究

本文报道输精管粘堵剂(504P)的代谢与毒理研究结果。动物实验结果表明:504部分地被机体吸收后较快排出体外,无长期蓄积作用。504P、504对动物无明显急毒作用;对所测生理指标、血细胞及生化指标亦无明显影响;组织学观察见堵塞处输精管腔纤维化闭塞,其近端输精管精子淤积;致癌试验表明该剂无致癌作用;Ames试验、染色体畸变分析、微核试验及显性致死试验表明该剂无致突变作用。
