糖尿病血管病变是常见的糖尿病并发症之一,可危及全身各个器官,是糖尿病患者死亡的重要原因。目前常规治疗手段对严重的糖尿病血管病变的效果有限。血管平滑肌细胞(VSMC)在糖尿病血管病变的进程中发挥重要作用,因此,通过调控VSMC来逆转糖尿病血管病变有广泛的应用前景。笔者就VSMC的生物学特征及其在糖尿病中的表型转换特点与VSMC调控应用于糖尿病的治疗策略进行综述。 相似文献

The mechanism for stem cell‐mediated improvement following acute myocardial infarction has been actively debated. We support hypotheses that the stem cell effect is primarily paracrine factor‐linked. We used a heparin‐presenting injectable nanofibre network to bind and deliver paracrine factors derived from hypoxic conditioned stem cell media to mimic this stem cell paracrine effect. Our self‐assembling peptide nanofibres presenting heparin were capable of binding paracrine factors from a medium phase. When these factor‐loaded materials were injected into the heart following coronary artery ligation in a mouse ischaemia‐reperfusion model of acute myocardial infarction, we found significant preservation of haemodynamic function. Through media manipulation, we were able to determine that crucial factors are primarily < 30 kDa and primarily heparin‐binding. Using recombinant VEGF‐ and bFGF‐loaded nanofibre networks, the effect observed with conditioned media was recapitulated. When evaluated in another disease model, a chronic rat ischaemic hind limb, our factor‐loaded materials contributed to extensive limb revascularization. These experiments demonstrate the potency of the paracrine effect associated with stem cell therapies and the potential of a biomaterial to bind and deliver these factors, pointing to a potential therapy based on synthetic materials and recombinant factors as an acellular therapy. Objective:To reveal the mechanism and effect of SU5416 in the treatment of mouse Lewis cancer in vivo.Methods:Lewis cell was transplanted into groin of C57/B6 mouse by subcutaneous injection,then SU5416 was administrated intraperitoneally to investigate the impact of SU5416 on tumor angiogenesis and growth in vivo.32 mice were treated with SU5416 at two different doses every day until the end-point.As a control,8 mice received no treatment and 8 mice were treated with vehicle (DMSO) only after implantation.Results:Median survival in the treated group was statistically longer compared to that in the control groups (P<0.05) and no significant systemic adverse was observed.Histological analysis of the treated tumors showed an increase in necroses and reduced in angiogenesis compared to the control tumors.Furthermore,the percent of apoptotic cells increased in the treated tumors by FCM,the expressions of VEGF and KDR had no change after SU5416 administration by western blot,Conclusion:SU5416 may be useful therapeutics drug that specifically inhibits the enzymatic activity of KDR kinase and could down regulate the tumor angiogenesis. 相似文献

目的:探索中医面色诊的规范化采集环境和客观识别方法,实现中医面色诊的计算机自动识别。方法:在中医理论指导下,比较各种人造光源,建立面色诊采集环境和临床面色判读量表;采集典型面色样本,结合图像处理、模式识别、人工智能等多种信息处理方法对面部颜色信息进行识别。结果:①研制了符合中医特点的以LED光源为照明光源的面色诊采集暗箱;②确定了符合中医理论的,临床专家认可的中医面部五色分类判读量表;③建立了面色诊颜色客观自动识别的方法,确定各类面色在色彩空间中的范围,初步研制中医面色识别分析系统。结论:该方法实现了中医面色诊的计算机自动识别,克服了传统中医望面色无客观量化标准和存在主观性的不足,增强了中医临床信息采集的规范化和准确性,提高了辨证诊断的科学性。 相似文献

自体外周血干细胞移植治疗老年人慢性下肢缺血临床研究 总被引:1,自引:0,他引:1
目的:评价外周血干细胞移植治疗老年人慢性下肢缺血的临床疗效。方法:应用自体外周血干细胞移植治疗慢性下肢缺血10例,其中下肢动脉硬化闭塞症4例,糖尿病足3例,血管闭塞性脉管炎3例。患肢12条中左5条,右7条。均予G-CSF 300μg/d皮下注射,连用4~6天行外周血干细胞动员,当外周血白细胞计数>30×109/L时,用血细胞分离机单采干细胞,配成干细胞悬液。部分干细胞悬液直接注射至患肢肌肉内,其余干细胞悬液经球囊导管技术灌注至患肢末梢动脉内。结果:患肢疼痛和冷感觉改善率为100%。间歇性跛行3例行走距离明显增加。移植后1个月,5例7条下肢数字减影血管造影结果示均有新生侧支血管形成。结论:外周血干细胞移植治疗慢性下肢缺血安全、简单、有效。 相似文献

Enhancement of contraction of rat mesenteric artery by acteoside: role of endothelial nitric oxide 总被引:1,自引:0,他引:1
The present study describes the role of endothelium in the vascular response to purified acteoside from Ligustrum purpurascens in rat mesenteric arteries. In endothelium-intact rings, acteoside (3-50 micromol/L) enhanced phenylephrine-induced contraction without affecting the maximum response. This enhancement was absent in endothelium-denuded rings. Pretreatment with nitric oxide synthase (NOS) inhibitors, N(G)-nitro-L-arginine (L-NNA, 100 micromol/L) and N(G)-nitro-L-arginine methyl ester (L-NAME, 100 micromol/L), or a selective guanylyl cyclase inhibitor, 1H-[1,2,4]oxadiazolo[4,2-alpha]quinoxalin-1-one (ODQ, 10 micromol/L), increased both the sensitivity of vasoconstriction to phenylephrine and the maximal response. The enhancing effect of acteoside (30 micromol/L) was abolished in the presence of L-NAME, L-NNA, or ODQ. Tetraethylammonium (TEA(+), 3 mmol/L), a putative K(+) channel blocker, also abolished the effect of acteoside. CaCl2 (0.01-10 mmol/L) induced contractions in 50 mmol/L K(+)-containing Krebs solution. Neither acteoside nor TEA(+) affected CaCl2-induced contraction in elevated K(+) solution. Acteoside (30 micromol/L) attenuated acetylcholine-induced endothelium-dependent relaxation. Acteoside did not influence relaxation induced by exogenous NO donors, hydroxylamine or sodium nitroprusside, in endothelium-denuded rings. Acteoside did not alter endothelium-independent relaxation induced by forskolin or NS 1619. The present results indicate that acteoside enhanced the evoked vasoconstriction, mainly through inhibition of endothelial NO production/release and inhibition of NO-mediated TEA(+)-sensitive activation of K(+) channels. 相似文献

目的 探讨对左髂静脉压迫综合征患者外科处理的适应证、手术方法和围手术期的处理经验。方法 左下肢静脉曲张或慢性肿胀46例,行介入治疗32例,进腹行髂静脉松解、右髂动脉后置吻合8例,髂静脉病变段切除、人工血管原位移植6例,同时或二期手术处理大隐静脉及股静脉瓣膜41例。急性左下肢深静脉血栓形成36例,取栓后行左髂静脉病变段切除、人工血管原位移植25例,髂一下腔静脉搭桥11例。随访6~66个月。结果 无手术死亡和急性肺动脉栓塞发生。效果良好。结论 左髂静脉病变是左下肢静脉曲张或肿胀和急性左下肢深静脉血栓形成的重要原因,应尽早处理。介入治疗或外科手术治疗能取得良好的效果。 相似文献
