Glioblastoma (GBM) is a highly malignant neurological tumor that has a poor prognosis. While pyroptosis affects cancer cell proliferation, invasion and migration, function of pyroptosis-related genes (PRGs) in GBM as well as the prognostic significance of PRGs remain obscure. By analyzing the mechanisms involved in the association between pyroptosis and GBM, our study hopes to provide new insights into the treatment of GBM. Here, 32 out of 52 PRGs were identified as the differentially expressed genes between GBM tumor versus normal tissues. And all GBM cases were assigned to 2 groups according to the expression of the differentially expressed genes using comprehensive bioinformatics analysis. The least absolute shrinkage and selection operator analysis led to the construction of a 9-gene signature, and the cancer genome atlas cohort of GBM patients were categorized into high risk and low risk subgroups. A significant increase in the survival possibility was found in low risk patients in comparison with the high risk ones. Consistently, low risk patients of a gene expression omnibus cohort displayed a markedly longer overall survival than the high risk counterparts. The risk score calculated using the gene signature was found to be an independent predictor of survival of GBM cases. Besides, we observed significant differences in the expression levels of immune checkpoints between the high risk versus low risk GBM cases, providing instructive suggestions for immunotherapy of GBM. Overall, the present study developed a new multigene signature for prognostic prediction of GBM.  相似文献

The changes of microstructure, magnetostriction properties and hardness of the Fe73Ga27−xAlx alloy and (Fe73Ga27−xAlx)99.9La0.1 alloy (x = 0, 0.5, 1.5, 2.5, 3.5, 4.5) were studied by doping Al into the Fe73Ga27 and (Fe73Ga27)99.9La0.1 alloy, respectively. The results show that both the Fe73Ga27−xAlx alloy and (Fe73Ga27−xAlx)99.9La0.1 alloy are dominated by the A2 phase, and the alloy grains are obvious columnar crystals with certain orientations along the water-cooled direction. A proportion of Al atoms replaced Ga atoms, which changed the lattice constant of the alloy, caused lattice distortion, and produced vacancy effects which affected the magnetostriction properties. La atoms were difficult to dissolve in the matrix alloy which made the alloy grains smaller and enhanced the orientation along the (100) direction, resulting in greater magneto-crystalline anisotropy and greater tetragonal distortion, which is conducive to improving the magnetostriction properties. Fe73Ga24.5Al2.5 alloy has a saturation magnetostrictive strain of 74 ppm and a hardness value of 268.064 HV, taking into account the advantages of saturated magnetostrictive strain and high hardness. The maximum saturation magnetostrictive strain of the (Fe73Ga24.5Al2.5)99.9La0.1 alloy is 115 ppm and the hardness is 278.096 HV, indicating that trace La doping can improve the magnetostriction properties and deformation resistance of Fe-Ga alloy, which provides a new design idea for the Fe-Ga alloy, broadening their application in the field of practical production.  相似文献

Purpose Spinal metastases of lung cancer (SMLC) usually have high degree of malignancy and require surgical treatment. However, there are several controversies about the efficacy of surgery. This study aimed to investigate factors predicting prognosis of SMLC after surgery‐based comprehensive treatment.MethodsA cohort of 112 cases of SMLC who underwent surgical treatment between 2009 and 2020 were retrospectively reviewed and analyzed. The surgical strategies included total en‐bloc spondylectomy, debulking surgery, palliative decompression, and vertebral augmentation procedures. The patients were regularly followed‐up. Survival analysis was performed, as well as analysis of the patients'' neurological recovery, pain relief, and improvement of Karnosky performance score (KPS). Cox regression was used to analyze influencing factors of survival time, and Kaplan–Meier method was performed in survival analysis.ResultsThe cohort included 63 males and 49 females, with an average age of 60.6 ± 10.6 years. Median survival time was 16 months. A total of 86.7% of paralysis patients'' neurological function recovered and 83.9% of patients with low KPS score (10–40) improved. Surgical method was significantly correlated with improvement of neurological function (p < 0.001) and KPS (p < 0.001). The mean bleeding volume was 502 ml and operative time was 170 min. The survival rates at 3, 6, 12, 24, and 36 months were 92.0%, 80.4%, 63.4%, 63.4%, and 22.6%, respectively. Postoperative Frankel grade (p < 0.001), postoperative KPS score (p = 0.001), and application of molecular targeted drugs (p < 0.001) were significantly correlated with survival time in univariate analysis, while application of molecular targeted drugs was an independent predictor for a longer survival by a multivariate analysis.ConclusionSurgery‐based comprehensive treatment brought a fair outcome, with elongated survival time. Surgery can significantly improve patients'' neurological function and physical performance status. Adjuvant targeted therapy is an independent positive factor for patients'' survival.  相似文献

目的:探讨诱型一氧化氮合酶(iNOS)与人皮肤自然衰老的关系.方法:收集整形外科植皮手术中非曝光部位的皮肤标本60例,采用反转录(RT)-PCR和免疫组化的方法检测皮肤组织中iNOS的表达水平,根据年龄分为青少年组、中年组和老年组3组,分析不同年龄组之间iNOS表达水平的差异.结果:青少年组、中年组和老年组iNOS mRNA表达的相对量分别为0.372 7±0.037 1、0.601 4±0.052 5和0.711 0±0.050 7,3组之间的差异具有统计学意义(P<0.05);免疫组化示iNOS蛋白在青少年组主要为低度阳性,中年组主要为中度阳性.老年组主要为中高度阳性,3组之间的差异亦具有统计学意义(P<0.05).结论:iNOS在皮肤组织中的表达量随年龄增长而增多,其可能与人皮肤的自然衰老有密切关系.  相似文献

背景 抑郁障碍患者的主观认知功能与客观认知功能存在差异。目前,关于主客观认知功能差异影响因素的研究有限。目的 探索抑郁障碍患者主观和客观认知功能的差异及其影响因素,为进一步理解抑郁障碍患者认知功能受损情况提供参考。方法 纳入2022年1月13日—2023年12月11日在成都市第四人民医院门诊就诊或住院治疗的、符合《精神障碍诊断与统计手册(第5版)》(DSM-5)抑郁障碍诊断标准的77例患者为研究对象。采用蒙哥马利-艾森贝格抑郁量表(MADRS)评定患者抑郁症状严重程度,采用抑郁感知缺陷问卷(PDQ-D)和中国简版神经认知成套测验(C-BCT)分别评定患者的主观认知功能和客观认知功能,采用席汉残疾量表(SDS)评定患者的社会功能,以临床总体印象量表-疾病严重程度量表(CGI-SI)评定患者病情严重程度。采用Pearson相关分析考查年龄、受教育年限、MADRS总评分、SDS总评分、CGI-SI评分与主客观认知功能及其差异的相关性。采用多元线性回归探索主客观认知功能差异的影响因素。结果 是否用药的抑郁障碍患者PDQ-D总评分和主客观认知功能差异(D值)比较差异均有统计学意义(t=-4.2...  相似文献

目的:探讨音乐疗法联合右美托咪定对腹腔镜胆囊切除术(LC)患者炎症的影响。方法:选择2020年10月~2021年4月本院因胆囊良性疾病全身麻醉下择期行LC手术患者60例,随机分为两组,每组30例:对照组给予右美托咪定治疗;试验组给予音乐疗法+右美托咪定治疗。记录两组术中情况和生命体征;采用视觉模拟评分法(VAS)评估患者术后疼痛程度;用ELISA法测定血浆中TNF-α、IL-8、IL-10的浓度。结果:两组患者的ASA分级、性别、年龄、体重指数、麻醉时长、手术时长、输液量、出血量比较差异均无统计学意义。试验组在出手术室时以及术后8h、24h、48h的VSA评分与对照组同时间点相比较均明显降低。接受音乐疗法后,试验组的MAP、HR与对照组同时间点相比较均明显降低;试验组TNF-α、IL-8的浓度与对照组同时间点相比较均明显降低;试验组IL-10的浓度与对照组同时间点相比较明显升高。结论:音乐疗法联合右美托咪定可以更好的缓解LC患者术后疼痛,降低炎症反应。  相似文献

Based on the theory of magnetoacoustic coupled dynamics, the purpose of this paper is to evaluate the dynamic stress concentration near an elliptical opening in exponential-gradient piezomagnetic materials under the action of antiplane shear waves. By the wave function expansion, the solutions for the acoustic wave fields and magnetic fields can be obtained. Stress analysis is performed by the complex function method and the conformal mapping method, which are used to solve the boundary conditions problem, and is used to express the dynamic stress concentration coefficient (DSCC) theoretically. As cases, numerical results of DSCCs are plotted and discussed with different incident wave numbers and material parameters by numerical simulation. Compared with circular openings, elliptical openings are widely used in material processing techniques and are more difficult to solve. Numerical results show that the dynamic stress concentration coefficient at the elliptical opening is strongly dependent on various parameters, which indicates that the elliptical opening is more likely to cause crack and damage to exponential-gradient piezomagnetic materials.  相似文献

BackgroundCough is one of the most common symptoms of coronavirus disease 2019 (COVID-19). However, the prevalence of persistent cough in recovered patients with COVID-19 during a longer follow-up remained unknown. This study aims to investigate the prevalence, and risk factors for postinfectious cough in COVID-19 patients after discharge.MethodsWe conducted a follow-up study for 129 discharged patients with laboratory-confirmed COVID-19 in two large hospitals located in Hubei Province, China from January 2020 to December 2020. Baseline demographics, comorbidities and smoking history were extracted from the medical record. Current symptoms and severity were recorded by a uniform questionnaire. Spirometry, diffuse function and chest computed tomography (CT) were performed on part of patients who were able to return to the outpatient department at follow-up.ResultsThe median (interquartile range) follow-up time was 8.1 (7.9–8.5) months after discharge. The mean (standard deviation) age was 51.5 (14.9) years and 57 (44.2%) were male. A total of 27 (20.9%) patients had postinfectious cough (>3 weeks), 6 patients (4.7%) had persistent cough by the end of follow-up, including 3 patients with previous chronic respiratory diseases or current smoking. Other symptoms included dyspnea (6, 4.7%), sputum (4, 3.1%), fatigue (4, 3.1%), and anorexia (4, 3.1%) by the end of follow-up. Thirty-six of 41 (87.8%) patients showed impaired lung function or diffuse function, and 39 of 50 (78.0%) patients showed abnormal CT imaging. Patients with postinfectious cough demonstrated more severe and more frequent cough during hospitalization (P<0.001), and more chronic respiratory diseases (P=0.01). In multivariate logistic regression analysis, digestive symptoms during hospitalization [odds ratio (OR) 2.95, 95% confidence interval (CI): 1.10–7.92] and current smoking (OR 6.95, 95% CI: 1.46–33.14) were significantly associated with postinfectious cough of COVID-19.ConclusionsA small part of patients developed postinfectious cough after recovery from COVID-19, few patients developed chronic cough in spite of a higher proportion of impaired lung function and abnormal lung CT image. Current smoking and digestive symptoms during hospitalization were risk factors for postinfectious cough in COVID-19.  相似文献

背景 默认网络内楔前叶功能活动与抗抑郁药物的疗效有关。然而,楔前叶功能网络与抗抑郁药物早期疗效的关系仍不清楚。 目的 探索抑郁障碍患者楔前叶功能连接(FC)与抗抑郁药物早期疗效的关系,以期寻找预测抗抑郁药物早期疗效的神经生物标志物。 方法 连续纳入2017年7月—2019年2月在四川大学华西医院心理卫生中心就诊的、符合《精神障碍诊断与统计手册(第5版)》(DSM-5)诊断标准的47例抑郁障碍患者。采集患者基线期静息态功能磁共振(rs-fMRI)数据及临床信息。患者接受2周抗抑郁药物治疗,根据治疗2周时16项抑郁症状快速自评量表(QIDS-SR16)评分减分率是否≥20%,将患者分为早期改善组( n=27)和未改善组( n=20)。以双侧楔前叶为种子点,计算楔前叶与全脑FC值,比较两组基线期楔前叶FC的差异。采用Pearson相关分析考查差异有统计学意义的脑区的FC值与QIDS-SR16评分及其减分率之间的相关性。 结果 早期改善组左侧楔前叶与左侧中央前回的FC值、右侧楔前叶与右侧梭状回的FC值均高于未改善组(GRF校正, P<0.01)。抑郁障碍患者左侧楔前叶与左侧中央前回的FC值、右侧楔前叶与右侧梭状回的FC值与QIDS-SR16总评分减分率均呈正相关( r=0.475、0.297, P均<0.05)。 结论 基线期较低的左侧楔前叶与左侧中央前回、右侧楔前叶与右侧梭状回的FC与较差的抗抑郁药物早期疗效有关,楔前叶FC可能是预测抗抑郁药物早期疗效的潜在指标。  相似文献
