In the present study effect of dietary restriction with and without leucine supplementation was observed on body and liver weights, and liver protein status, in adult rats. Animals were fed on two diets ad lib or were on 50 per cent and 25 per cent intakes. Dietary restriction resulted in loss of body and liver weights, hepatic protein, free-alpha-amino nitrogen and RNA contents and liver cell size (liver weight/DNA ratio). When compared with the control group, the decrease in these parameters was more in the dietary restricted leucine supplemented group. However, hepatic DNA content was not changed with the change in dietary regimen. The results suggest that leucine supplementation with dietary restriction may be more harmful for the animal than dietary restriction alone.  相似文献

Susceptibility of Campylobacter jejuni to twenty-three antimicrobial agents   总被引:7,自引:0,他引:7  
Twenty-three antimicrobial agents, including 4 new broadspectrum beta-lactam antibiotics were tested against 50 clinical isolates of Campylobacter jejuni. The activity of metabolites of metronidazole and tinidazole was also tested. Minimum inhibitory concentrations (MIC) were determined by agar dilution. beta-lactamase production was detected by a chromogenic cephalosporin method. All strains were susceptible to erythromycin, clindamycin, chloramphenicol, furazolidone, aminoglycosides (including gentamicin), cefotaxime and NF-thienamycin. All isolates were resistant to penicillin, cephalothin, cefoperazone, vancomycin, rifampicin and trimethoprim; beta-lactamase was detected in 2% of isolates. Some strains were resistant and others sensitive to the other drugs tested, which included ampicillin, moxalactum tetracycline, metronidazole and tinidazole. The 'hydroxy' metabolites of metronidazole and tinidazole were more active than the parent compounds.  相似文献

Summary: Purpose: Ictal perfusion single photon emission computed tomography (SPECT), using HMPAO, has been shown to localize epileptic foci in ~90% of studies. Unfortunately, HMPAO decomposes rapidly, precluding the performance of ictal studies. Ethyl cysteinate dimer (ECD) is a SPECT perfusion agent recently approved by the Food and Drug Administration. After preparation, this compound is stable for ~6 h. facilitating the performance of ictal studies. Methods: In a prospective, open-label, uncontrolled, non randomized study, we evaluated the potential benefits of the use of 99mTc-ECD SPECT for lateralization of the epileptic focus. Ten consecutive adult epilepsy surgery candidates were studied with ictal and interictal 99mTc-ECD SPECT. Results: The mean delay between seizure onset and ictal SPECT injection was 23.2 s. The mean seizure duration was 84.1 s. Ictal studies agreement between the epilepsy focus and area of hyperperfusion was evident in 8 of 10 cases. In one case, SPECT was lateralized in a patient with bilateral temporal lobe epilepsy (TLE); however, hyperperfusion was observed on the same side of that particular seizure. In another case, there was location disagreement. Interictal SPECT showed focal hypoperfusion in three cases. Conclusions: 99mTc-ECD proved to be an optimal tracer for ictal studies. Although this is a small series, the results of ictal and interictal findings using 99mTc-ECD are similar to those reported with 99mTc-HMPA0. Because 99mTc-ECD has a longer decomposition time, true ictal studies are easier to obtain. This new tracer will probably allow the use of ictal SPECT to become widely accepted in most epilepsy centers.  相似文献

The level of myosin heavy chain (MHC) alpha mRNA and of MHC-beta mRNA was quantitated in the rat heart using a specific cDNA probe. In hypothyroid and diabetic hearts MHC-beta mRNA predominates, whereas in normal hearts MHC-alpha mRNA represents 80% of all MHC mRNA. Administration of 0.2 mg T3/100 g body wt. to hypothyroid rats led to an increase in MHC-alpha mRNA beginning at 3 h after injection and continued to rise until at 24 h control level of MHC-alpha mRNA were reached. In contrast, after administration of 2 units regular insulin to diabetic rats, MHC-alpha mRNA levels showed a small but significant increase already 30 min after insulin administration reaching a peak at 3 h and returning to diabetic values 5 h after insulin. The T3 response of other cardiac mRNAs was quantitated using in vitro translation, separation of 35S methionine labeled translational products and their quantitation by digital matrix photometry. An mRNA (spot 72b) coding for a translational product with a Mr 81,000 and pI of 5.4 showed a 3-fold increase in its level 1 h after T3 administration. In view of the rapid response of spot 72b and the early response of MHC-alpha mRNA to insulin, it is currently unclear if the T3 response of MHC-alpha mRNA represents a primary effect of T3.  相似文献

Experiments were conducted to determine whether photoreceptor outer segment components contain precursors for lipofuscin fluorophores that accumulate in the retinal pigment epithelium (RPE) during senescence. Intravitreal injection of the lysosomal protease inhibitor leupeptin caused a rapid accumulation of inclusions with lipofuscin-like autofluorescence in the RPE of albino rats. These inclusions appeared to be derived from photoreceptor outer segments, which are normally phagocytosed and degraded by the RPE. The tripeptide leu-gly-gly, which is similar to leupeptin except that it does not inhibit proteolysis, had no effect on RPE autofluorescent pigment content. Likewise, netilmicin, a purported inhibitor of lysosomal phospholipases, did not enhance autofluorescent pigment deposition in the RPE. These findings are consistent with other experiments suggesting that RPE age-pigment fluorophores are derived from molecular components of the photoreceptor outer segments. The effect of leupeptin suggests that outer segment proteins may be among the precursors for these fluorophores. The RPE appears to differ from other cell types that accumulate lipofuscin in that the majority of the precursors for this pigment in the RPE are taken up by phagocytosis rather than being generated within the cells themselves.  相似文献

The real and imaginary components, C'(p) and C'(p), of the complex heat capacity, C*(p)=C'(p)-iC"(p) of supercooled, ultraviscous melt of acetaminophen have been measured at different temperatures during cooling through its vitrification range and during heating through its glass-softening range by using a modulation frequency of 3.3 mHz. From these data, the distribution of relaxation time parameter, beta, and a characteristic (calorimetric or configurational) relaxation time, tau(cal), have been determined. A constant value of 0.65 for beta fits the data, and tau(cal) varies with the temperature according to the Vogel-Fulcher-Tammann equation, tau(cal) = 10(-12.95) exp[1813/(T - 240.5)]. This relation differs significantly from the one deduced by others in which the configurational entropy theory was used to deduce tau(cal). The C'(p) and C'(p) values measured during the cooling of its ultraviscous melt and during the heating of its glassy state show a small hysteresis only at low temperatures. These investigations also provide a comparison of calorimetric and dielectric relaxation times in ultraviscous acetaminophen and highlight the role of faster modes of relaxation at low temperatures in organic, molecular glasses that can help in a better understanding of the crystal nucleation process in glasses at T below their T(g).  相似文献
