外源性AA引起兔动脉条收缩,呈剂量依赖性;EPA抑制AA收缩血管亦呈浓度依赖性;DHA对AA收缩血管作用无明显影响。破坏血管内皮后AA收缩血管作用大为减弱,EPA抑制AA收缩血管作用也几乎消失。吲哚美辛能阻断AA收缩兔主动脉条的作用。兔主动脉6-keto-PGF_1α、TXB₂及其比值随AA浓度升高而增加,低剂量EPA对前列腺素类代谢无明显影响,较大剂量时则降低上述指标。     

Renal thorium deposition associated with transitional cell carcinoma: radiologic demonstration in two patients

Abdominal radiography, excretory urography, retrograde pyelography, and computed tomography were performed in two patients who had undergone retrograde pyelography with thorium dioxide (Thorotrast) approximately 40 years ago. Both patients developed a transitional cell carcinoma due to suburothelial thorium deposition. Typical thorium densities were demonstrated at CT in the peripelvicalyceal area as well as in retroperitoneal lymph nodes. Elderly patients in whom radiographic examination reveals retained Thorotrast in the kidney should be followed up because of the high risk of renal carcinoma.     

A comparison of two radioimmunoassays for 7-hydroxychlorpromazine: rabbit polyclonal antibodies vs. mouse monoclonal antibodies

Two radioimmunoassays (RIAs) were developed for 7-hydroxychlorpromazine (7-OHCPZ), which is a pharmacologically active chlorpromazine (CPZ) metabolite. One of the RIAs used polyclonal antibodies produced in rabbits immunized with a 7-OHCPZ-protein conjugate, which was prepared by coupling 7-hydroxy-N-(2-carboxyethyl)desmethylchlorpromazine to bovine serum albumin by a mixed anhydride method (90% yield). The other RIA was based on mouse monoclonal antibodies produced by hybridomas against the same conjugate. The mouse monoclonal antibodies were considerably more specific than the rabbit polyclonal antibodies. There was little interference with the measurement of 7-OHCPZ by RIA based on mouse monoclonal antibodies even when the samples were spiked with 7-OHCPZ in the presence of five times excess of CPZ and two major metabolites, CPZ sulfoxide and CPZ-N-oxide. By contrast, there was a significant increase in the apparent concentration of 7-OHCPZ when the same samples were assayed by RIA based on polyclonal antibodies. The RIA based on mouse monoclonal antibodies was applied, together with an RIA for CPZ to determine the concentrations of 7-OHCPZ and CPZ in plasma samples from two healthy volunteers after they had received a single 50 mg p.o. dose of CPZ. To our knowledge, this is the first time that plasma 7-OHCPZ concentrations, measured up to 24 hr after a single dose of CPZ, are reported in humans.     

Klatskin tumors of the bile ducts: sonographic appearance

The authors present 3 cases of surgically proved Klatskin tumor diagnosed by ultrasound alone. Sonographic features of these tumors include (a) dilatation of the intrahepatic biliary ducts but not the extrahepatic duct, (b) non-union of the right and left hepatic ducts, and (c) small, solid masses at the hepatic hilus. Local spread to the liver may also be seen. If the pancreas appears normal and no primary tumor can be found, Klatskin tumor can be diagnosed with a high degree of accuracy.     

Dose-response relationship of DNA and recombinant fowlpox virus prime-boost HIV vaccines: implications for future trials

Estimating effective doses of novel HIV vaccines is challenging. Dose-response analyses of DNA and fowlpox virus HIV vaccines showed that 1 mg of DNA vaccine and 5 x 10(7)pfu of fowlpox virus booster was immunogenic in macaques. However, this dose was poorly immunogenic in humans. When adjusted for body surface area, the human dose studied was equivalent to a poorly immunogenic lower dose in monkeys. These data provide a rationale for guiding dosing in future trials of HIV vaccine technologies.
