Motility of unicellular organisms occurred early in evolution with the emergence of cilia and flagella. In vertebrates, motile cilia are required for numerous functions such as clearance of the airways and determination of left-right body asymmetry. Ependymal cells lining the brain ventricles also carry motile cilia, but their biological function has remained obscure. Here, we show that ependymal cilia generate a laminar flow of cerebrospinal fluid through the cerebral aqueduct, which we term as 'ependymal flow'. The axonemal dynein heavy chain gene Mdnah5 is specifically expressed in ependymal cells, and is essential for ultrastructural and functional integrity of ependymal cilia. In Mdnah5-mutant mice, lack of ependymal flow causes closure of the aqueduct and subsequent formation of triventricular hydrocephalus during early postnatal brain development. The higher incidence of aqueduct stenosis and hydrocephalus formation in patients with ciliary defects proves the relevance of this novel mechanism in humans. 相似文献

The purpose of this study was to evaluate the clinical usefulness of a commercial ligase-based gene amplification method (LCxMycobacterium tuberculosis test; Abbott Laboratories, USA) for detection ofMycobacterium tuberculosis. The tuberculosis infection rate among clinical samples was 10.6%. The sensitivity, specificity, and positive and negative predictive values were 23.5%, 100%, 100%, and 91.7%, respectively, with the fluorochrome auramine stain; 32.4%, 100%, 100%, and 92.6%, respectively, with culture; and 76.5%, 95.8%, 68.4% and 97.2%, respectively, with the gene amplification method. When only samples from patients without current or previous treatment were studied, the sensitivity was 36.4% with the auramine stain, 63.6% with culture, and 100% with the gene amplification assay. The mean treatment time for culture-negative and assay-negative samples was greater than that of culture-negative and assay-positive samples. The LCxMycobacterium tuberculosis test is a sensitive method for detection and identification ofMycobacterium tuberculosis. It produces few false-positive results. However, as it can remain positive after the culture becomes negative, it is not recommended for evaluation of treatment efficiency. 相似文献

This paper reports the annual dynamics of wild rabbit fleas in a study site located in the Middle Ebro Valley, northeastern Spain. Fleas collected directly from wild rabbits included the species Spilopsyllus cuniculi (Dale), Xenopsylla cunicularis (Smit), Echidnophaga iberica (Ribeiro, Lucientes, Osácar, and Calvete), Caenopsylla laptevi (Beaucournu, Gil-Collado and Gilot), and Pulex irritans (L.). Monthly collections of adult and larval fleas made from within the first meter of selected burrow entrances also yielded fleas belonging to the same five species. Larval specimens of X. cunicularis, E. iberica, and C. laptevi were also found. Spilopsyllus cuniculi, a winter species that can only breed during the rabbit breeding season, was common on hosts from November to April. Xenopsylla cunicularis and E. iberica were summer species, whereas C. laptevi was abundant during the autumn and winter. Xenopsylla cunicularis and E. iberica larvae were found in burrows only during April and May, whereas those of C. laptevi were collected from October to January. The data suggested that X. cunicularis and E. iberica might diapause during the egg stage whereas C. laptevi diapauses during the pupal stage. 相似文献

The effects of 8-bromoguanosine 3:5-cyclic monophosphate (8Br-cGMP), a membrane-permeant activator of protein kinase G (PKG), were studied on rat and human connexin43 (Cx43), the most abundant gap junction protein in mammalian heart, which were exogenously expressed in SKHep1 cells. Under dual whole-cell voltage-clamp conditions, 8Br-cGMP decreased gap junctional conductance (gj) in rat Cx43-transfected cells by 24.0±3.7% (mean±SEM, n=5), whereas gj was not affected in human Cx43-transfected cells by the same treatment. The relaxation of gj in response to steps in transjunctional voltage observed in rat Cx43 transfectants was best fitted with three exponentials. Time constants and amplitudes of the decay phases changed in the presence of 8Br-cGMP. Single rat and human Cx43 gap junction channels were resolved in the presence of halothane. Under control conditions, three single-channel conductance states (j) of about 20, 40–45 and 70 pS were detected, the events of the intermediate size being most frequently observed. In the presence of 8Br-cGMP, the
j distribution shifted to the lower size in rat Cx43 but not in human Cx43 transfectants. Immunoblot analyses of Cx43 in subconfluent cultures of rat Cx43 or human Cx43 transfectants showed that 8Br-cGMP did not induce changes in the electrophoretic mobility of Cx43 in either species. However, the basal incorporation of [32P] into rat Cx43 was significantly altered by 8Br-cGMP, whereas this incorporation of [32P] into human Cx43 was not affected. We conclude that 8Br-cGMP modulates phosphorylation of rat Cx43 in SKHep1 cells, but not of human Cx43. This cGMP-dependent phosphorylation of rat Cx43 is associated with a decreased gj, which results from both an increase in the relative frequency of the lowest conductance state and a change in the kinetics of these channels. 相似文献

ABSTRACT To determine the prevalence of low scores on two neuropsychological tests commonly used to evaluate learning and memory in children. 6,030 healthy children from 10 countries in Latin America and Spain were administered Rey–Osterrieth Complex Figure (ROCF) and the Test de Aprendizaje y Memoria Verbal–Infantil (TAMV-I). Results showed that low scores are common when multiple neuropsychological outcomes (tests and/or scores) are evaluated in healthy individuals. Clinicians should consider the higher probability of low scores in a given individual when evaluating learning and memory using various sets of scores to reduce false-positive diagnoses of cognitive deficits in pediatric populations. 相似文献

Three previously healthy children developed gastroenteritis which led within a few days to systemic infections, two cases of bacteremia and one of meningitis. A lactose-fermentingSalmonella virchow strain was isolated from cerebrospinal fluid and blood cultures. In one case, this strain was also isolated from stool cultures. All the children had been fed the same milk formula. There was no other relationship between them. To be clinically useful as indices reflective of altered physiological function consequent to interventions in patients with
chronic obstructive pulmonary disease (COPD), the time constant (τ) and steady-state amplitude of the kinetic responses for
oxygen uptake (
) carbon dioxide output (
) ventilation (
) and heart rate (HR) have to be appropriately differentiable and reproducible. We therefore assessed the reproducibility
of τ and steady state amplitude values in 41 patients with severe COPD [mean (SD)] [forced expiratory volume in 1 s=41 (7)%
predicted], aged 64 (5) years. Of the total, 6 of the patients (15%) did not produce breath-by-breath data of sufficient quality
to warrant kinetic analysis. The remaining 35 patients completed two moderate-intensity 10 min square-wave exercise tests
separated by 2 h, both before and after an endurance training programme. Tests were conducted on an electromagnetically-braked
cycle ergometer at an exercise intensity corresponding to 80% of the estimated lactate threshold (θLa) or 50% of peak oxygen uptake if θLa was insufficiently differentiable. Breath-by-breath measurements of

and HR were averaged into 10 s bins and the on-transient response kinetics were estimated using a mono-exponential model.
Analysing the pre-training and the post-training test 1 and test 2 comparisons together, the test 1 –test 2 differences were
not significantly different from 0 for either τ or A. The standard deviation of the test 1 –test 2 differences allowed us
to define the magnitude of change that would reach statistical significance. For τ, this averaged some 8, 10, 11 and 8 s,

and HR, respectively, for a one-tailed paired-comparisons test (i.e. appropriate for assessing hypothesised improvements resulting
from an intervention); for a two-tailed comparison, the differences were approximately 2 s greater. The corresponding one-tailed
values for A were 100 ml·min–1, 95 ml·min–1, 2.5 1·min–1 and 4 beats·min–1, respectively; the two-tailed values were 10%–15% greater. We therefore conclude that both τ and A for moderate-intensity
exercise can be reproducibly estimated in patients with COPD when the data set provides a sufficiently large amplitude of
response and sufficiently low sample variability to allow appropriate parameter estimation.
