AIM:to study the prevalence of human papillomavirus(HPV) in esophageal carcinoma in tangshan,China,a high-incidence area.METHODS:Formalin-fixed,paraffin-embedded tissue specimens from 198 patients who were pathologically diagnosed with esophageal squamous cell carcinoma from 2011 to 2013 were obtained from a pathology department in Tangshan.DNA was extracted from all198 specimens to detect HPV by polymerase chain reaction(PCR).β-globin PCR was performed to check the quality of the DNA extraction procedure.PCR was performed to detect a wide range of HPV types,and type-specific PCR was performed to detect HPV types16 and 18.Negative and positive controls were used for HPV 16 and 18 detection.RESULTS:the DNA extraction method in this study appeared to be more effective than other previously reported methods.After DNA extraction,more than98%of the tissue specimens had an acceptable result in the DNA qualification test(β-globin PCR).the overall prevalence of HPV in tumor tissues by GP6+/GP5+PCR was 79.79%,and the prevalence of HPV types16 and 18 was 40.40%and 47.47%,respectively.PCR demonstrated the presence of HPV,and direct sequencing confirmed the HPV genotypes.All HPVpositive PCR products were checked by DNA sequence analysis using DNAman and compared with the known HPV sequences listed in the basic Local Alignment Search tool database to evaluate the HPV types.this analysis confirmed the presence of HPV types 16 and18.CONCLUSION:DNA of high-risk HPV types 16 and 18is present in esophageal tumors,implicating HPV as a possible etiologic factor for esophageal squamous cell carcinoma.  相似文献

The main aim of this study was to propose a new statistical method for evaluation of spatial malignancy distribution within Magnetic Resonance Spectroscopy (MRS) grid in Glioblastoma Multiforme patients. Voxels with different malignancy probabilities were presented as a novel MRS-based Malignancy Probability Map (MPM). For this purpose, a predictive probability-based clustering approach was developed, including the two following steps: (1) Gaussian Mixture Model, (2) Quadratic Discriminate Analysis coupled with Genetic Algorithm. Clustered probability values from two methods were then integrated to exploit the MPM. Results show that the suggested method is able to estimate the malignancy distribution with over 90% sensitivity and specificity. The proposed MRS-based MPM has an acceptable accuracy for providing useful complementary information about regional diffuse glioma malignancy, with the potential to lead to better detection of tumoral regions with high probability of malignancy. So, it also may encourage the use of additional information of this map as a tool for dose painting.  相似文献

Nasopharyngeal carcinoma (NPC), although not very common in many parts of the world, is a major concern insome countries, including Iran. Molecular studies are very helpful to provide essential information regarding underlyingcarcinogenetic mechanisms. Here, considering NPC proteomic approaches, established biomarkers were designated forprotein-protein interaction network construction and analysis with corresponding plug-ins. A network of reported proteinmarkers was constructed and topological and biological process features were investigated. Centrality analysis showedthat JUN, CALM1, HSB1, and SOD1 are more important than other differentially expressed proteins in an interactingpattern. What is more, by extending the network, Tp53, PRDM10, AKT1, ALB, HSP90AA1, and EGFR achieved thehighest values for NPC network strength. It can be concluded that these proteins as well as their contributing processes,particularly in a second network, may be important for NPC onset and development. Targeting these candidate proteinsmay allow novel treatment approaches following appropriate validation.  相似文献

Objective: CD44 is an important cell adhesion molecule that plays a key role in growth, invasion, proliferation andmetastasis of cancer cells. CD44 protein over-expression is associated with a poor prognosis of gastric cancer (GC) andprevious studies have shown that CD44 gene polymorphisms could affect survival and recurrence. In this study, wetested the hypothesis that polymorphisms impacting on the CD44 signaling pathway may predict clinical outcomes inpatients with GC. Materials and Methods: DNA was extracted from blood of 150 healthy individuals and formalin-fixedparaffin-embedded (FFPE) tumor tissue of 150 patients. The two polymorphisms rs187116 and rs7116432 werestudied by RFLP-PCR and sequencing techniques. Results: There was a strong significant correlation between singlenucleotide polymorphisms (SNPs) in the CD44 gene, tumor recurrence, and overall survival (p <0.0001). The existenceof a significant relationship between tumor recurrence and overall survival was proved in this study, with at least oneallele G for the polymorphism rs187116 and at least one allele A for polymorphism rs7116432. Conclusion: These resultsprovide evidence of a relationship between CD44 gene polymorphisms and clinical outcomes in our GC patients.This result could help identify individuals with GC who have a high risk of tumor recurrence.  相似文献

This study aimed to investigate the effect of olive oil-rich diet on omentin and adiponectin concentrations. This cross-over randomized trial included 17 overweight women. Participants were assigned to consume either a usual (16% saturated fatty acids [SFA] and 8% monounsaturated fatty acid [MUFA]) or an olive oil-rich diet (16% MUFA and 8% SFA) for 6 weeks crossing over after a 2-week washout period. There was no significant difference in the changes of omentin between two dietary interventions. However, in the adjusted model for polyunsaturated fatty acids and fat mass, usual diet tended to decrease omentin levels whilst olive oil-rich diet tended to increase (?56.1?±?32.0 versus 40.6?±?32.0?ng/mL; p?=?.056). Adiponectin levels increased during two periods, but changes were greater during olive oil-rich diet with a trend toward significance (4.8?±?3.0 versus 13.4?±?3.0?μg/mL; p?=?.06). Consumption of olive oil-rich diet tended to increase omentin and adiponectin in comparison with the usual diet.  相似文献
