Differential effects of the pyrethroid tetramethrin on tetrodotoxin-sensitive and tetrodotoxin-resistant single sodium channels

The differential effects of the pyrethroid tetramethrin on tetrodotoxin-sensitive (TTX-S) and tetrodotoxin-resistant (TTX-R) single sodium channel currents in rat dorsal root ganglion (DRG) neurons were investigated using the outside-out configuration of patch-clamp technique. Channel conductances were 10.7 and 6.3 pS for TTX-S and TTX-R sodium channels, respectively, at a room temperature of 24–26°C. The single-channel current of TTX-S sodium channels at the test potential of −30 mV was −1.27 ± 0.25 pA, and was not changed after exposure to 10 μM tetramethrin (−1.28 ± 0.23 pA). The open time histogram of TTX-S single-channel currents could be fitted by a single exponential function with a time constant of 1.27 ms. After exposure to 10 μM tetramethrin, the open time histogram could be fitted by the sum of two exponential functions with time constants of 1.36 ms (τ_fast) and 5.73 ms (τ_low). The percentage of contribution of each component to the population was 62% for the fast component representing the normal channels and 38% for the slow component representing the tetramethrin modified channels. The amplitudc of TTX-R single-channel currents was slightly changed from −0.72 ± 0.14 to −0.83 ± 0.07 pA by 10 μM tetramethrin. The open time histogram of TTX-R single-channel currents could be fitted by a single exponential function with a time constant of 1.92 ms. In the presence of 10 μM tetramethrin, the open time histogram could be fitted by the sum of two exponential functions with time constants of 2.07 ms (τ_fast) and 9.75 ms (τ_slow). The percentage of contribution of each component was 15% for the fast, unmodified component and 85% for the slow, modified component. Differential effects of tetramethrin on the open time distribution of single sodium channel currents explains the differential sensitivity of TTX-S and TTX-R sodium channels.     

雷公藤总甙(TⅡ)中雷藤氯内酯醇的高效液相色谱分析

建立了雷公藤总甙(T_1)中雷藤氯内酯醇的高效液相色谱分析方法。采用μ-Bondapak C_(18)柱和46％甲醇-水流动相,紫外检测波长219nm。考察了雷藤氯内酯醇的稳定性,雷藤氯内酯醇在0.5～10.0μg/ml范围内线性良好,回收率在98％～101％之间。     

冠心病、高脂血症和低高密度脂蛋白血症患者载脂蛋白Al基因的多态性

本实验用分析基因限制性内切酶片段长度多态性(Restriction fragment length olymorphisms,RFLPs)技术,检测到中国人载脂蛋白AI(Apolipoprotein AI。Apo AI)基因3′端存一PstI多态位点,并发现该多态位点与低高密度脂蛋白(HDL)血症及冠心病有较密切的关系。     

男性不育患者的心理健康状况及相关因素探讨

目的　探讨男性不育患者的精神状况及与之相关的因素。方法　对 90例男性不育患者和 93例已婚有子女的健康男性进行症状自评量表和一般情况问卷调查。结果　男性不育患者的心理刺激重 ,症状多。不育患者的精神压力与职业、文化程度、每周性生活次数密切相关。农民、文化层次较低及性生活次数多的男性不育患者精神负担重 ,主要表现为人际关系敏感、抑郁、焦虑或偏执等症状。结论　建议在对不育男性尤其是农民不育患者进行不育治疗的同时给予心理健康服务和生育知识指导 ,以缓解患者的精神压力。     

钛接圈在不同烘烤温度下对镍铬合金浇注质量的影响

目的：研究钛接圈在不同烘烤温度下浇注镍铬合金的质量关系。方法：选择临床骨融合种植义齿二期手术后典型病例石膏模型，在基桩上固定纯钛接圈，常规制作桥架蜡型，使桥架接圈周围嵌体蜡厚度为１ｍｍ，常规内、外包埋，放置２４ｈ后，分４组不同温度烘烤，ａ组烘烤升温至６００℃，ｂ组升温至６５０℃，ｃ组升温至７００℃，ｄ组升温至８００℃。每组试件１０个，共４０个试件。用远红外温度测试仪控制合金浇注温度为１３０６℃，自然冷却后，电镜观察。结果：钛接圈烘烤温度６００℃、８００℃时镍铬合金支架断口出现二次断裂及气孔机会多，当６５０℃、７００℃时二次断裂出现机会少。结论：在常规牙科操作中，钛接圈烘烤温度为６５０℃、７００℃时，浇注镍铬合金较好。     

荧光定量聚合酶链反应在尖锐湿疣诊断中的应用

目的　探讨荧光定量聚合酶链反应 (FQ - PCR)对尖锐湿疣 (CA )的诊断价值。方法　 FQ- PCR检测病理确诊的 CA患者 36份标本和健康人 84份标本 ;FQ- PCR及病理诊断同期检测临床送检病例 2 73份标本 ,并做诊断性试验评价。结果　 36例病理确诊的 CA患者 HPV6和 HPV11的 DNA FQ- PCR全部阳性 ,平均拷贝数为 1.0× 10 7± 1.0× 10 2 / m l;84例健康人 FQ- PCR全部阴性 ,平均拷贝数为 3× 10± 2× 10 / ml。 FQ- PCR与病理诊断符合率为 10 0 % ,CA患者与健康人拷贝数的差异有显著性 (P<0 .0 5 )。FQ- PCR与病理诊断对比得出 :灵敏度为 10 0 % ,特异度为 92 % ,误诊率为 0 .0 8,漏诊率为 0 ,准确度为 98.9% ,阳性预测值为 98.8% ,阴性预测值为 10 0 % ,阳性似然比为 12 .5 ,阴性似然比为 0。结论　 FQ - PCR能准确定量 ,灵敏度高 ,特异性强 ,快速 ,简便 ,可作为尖锐湿疣早期诊断的指标     

顺丁烯二酸水溶液的热反应动力学

研究了不同温度、浓度条件下，顺丁烯二酸（顺酸）非催化反应网络，即顺酸可生成反丁烯二酸（反酸）和苹果酸，反酸与苹果酸存在可逆反应。建立了该反应网络的动力学模型。测定了不同反应条件下顺酸、反酸和苹果酸浓度随时间变化的规律，据此，动力学参数进行了估值。     

A Comparative Study of Dihydroartemisin in Compounds in Treatment of Uncomplicated Falciparum Malaria in Kampong of Cambodia

Objective: To compare the safety and efficacy of two compounds of dihydroartemisinin(DHA) -Artekin and Artekin (T) in the treatment of uncomplicated falciparum malaria. Methods:The regimen of 8-tablet for 2 days of Artekin and Artekin (T) were applied to 100 patients with uncomplicated falciparum malaria, who were randomly divided into two groups. Each group contained 50 cases. The cure rate, the mean parasites clearance time, the mean fever clearance and side-effects were observed to assess the safety and efficacy of the compounds used. Results: The mean parasites clearance time was 31. 7±9.0 hours in the Artekin group and 32. 8±8. 8 hours in Artekin (T) group respectively; the mean fever clearance time was 12. 7±7. 2 hours in Artekin group and 16. 5±7. 9 hours in Artekin (T) group; there were no recrudescence case in both groups within the 28 days of follow-up, the cure rates in Artekin group and Artekin (T) groups were 100%. Onychomycoses due to Microascus cirrosus (syn. M. desmosporus)

Microascus cirrosus is very rarely the aetiological agent of onychomycosis. We report two additional cases of toenail infections caused by this fungus.     

肉毒碱对实验性心肌损伤线粒体呼吸酶的影响

以大剂量维生素Ｄ造成心肌损伤模型，以肉毒碱作为影响因素，通过电镜细胞化学方法，观察了肉毒碱对心肌线粒体细胞色素Ｃ氧化酶和琥珀酸脱氢酶的作用。结果表明，肉毒碱具有提高线粒体呼吸酶活性及保护线粒体膜结构的应用，从而保护了呼吸链的完整性，改善了心肌氧化磷酸化功能。
