The reliability and validity of qualitative research findings are viewed with scepticism by some scientists. This scepticism is derived from the belief that qualitative researchers give insufficient attention to estimating reliability and validity of data, and the differences between quantitative and qualitative methods in assessing data. The danger of this scepticism is that relevant and applicable research findings will not be used. Our purpose is to describe an evaluative strategy for use with qualitative data, a strategy that is a synthesis of quantitative and qualitative assessment methods. Results of the strategy and factors that influence its use are also described.  相似文献

The syndrome of symmetrical peripheral gangrene is characterised by distal ischaemic damage in two or more extremities, without large vessel obstruction. Four patients with bilateral pedal ischaemia are described and their haemodynamic profiles presented. In all four cases the syndrome developed in association with noradrenaline administration, sepsis and DIC, despite a high cardiac output and a low calculated systemic vascular resistance index. Early treatment with epoprostenol was instituted in the final case and was successful.  相似文献

The sequential effects of removal of the corpus luteum, removal of the non-luteal ovary and sucking stimulus and the effects of season of the year on the release of luteinizing hormone (LH) and follicle stimulating hormone (FSH) have been determined in the tammar wallaby, Macropus eugenii. Plasma concentrations of LH and FSH were measured at 15 min intervals for 6 h in eight animals at four successive times in the breeding season (lactational quiescence); (A) while they were intact and lactating, (B) 10 days after removal of the ovary bearing the quiescent corpus luteum, (C) 21 days after bilateral ovariectomy and (D) 21 days after removal of the sucking stimulus, and (E) in the following non-breeding season (seasonal quiescence). Single blood samples were taken twice weekly during lactational quiescence. In the presence of ovarian tissue, basal concentrations of LH were low (0.94 ng mL-1) with pulses of low magnitude (1.3 ng mL-1) and low frequency (1.4 pulses per 6 h). There was no response to luteectomy but all three parameters increased after bilateral ovariectomy. Removal of the sucking stimulus affected the LH pulse frequency but seasonal differences were not evident. The pattern of release of FSH was not pulsatile. There was no response to luteectomy in basal concentrations of FSH but these rose significantly after bilateral ovariectomy (P less than 0.001) in lactational quiescence. There was no effect of removing the sucking stimulus but in seasonal quiescence concentrations were higher. The results indicate that non-luteal ovarian tissue is essential for the negative-feedback effects on LH and FSH secretion, that the pattern of release of LH, but not FSH, is pulsatile, and that there is no marked seasonal change in hypothalamo-hypophysial activity.  相似文献

We describe a ligand displacement immunoassay for measurement of theophylline in serum or plasma and show it to be clinically useful. A conjugate of theophylline-specific antiserum covalently bound to micro-crystalline cellulose and a beta-galactosidase-labeled theophylline derivative is prepared as a lyophilized reagent. Sample is added to the conjugate and, after 5-min incubation at room temperature, the bound and displaced enzyme-ligand are separated by brief centrifugation. The enzyme activity of the displaced enzyme-ligand is measured on a spectrophotometer or centrifugal analyzer. Linear calibration curves are obtained with appropriate sample dilution, allowing generation of the standard curve with a saline zero reference and a single calibrator. Performance of the assay compares favorably with that of other theophylline assays; comparison studies with use of 145 clinical specimens gave a coefficient of correlation with EMIT of 0.97.  相似文献

The collection characteristics of five types of substrates (collection surfaces) used in personal cascade impactors were evaluated for particle bounce in the laboratory with lead dioxide dust, and in the field with brass pouring fume and brass grinding dust. The substrates tested were uncoated stainless steel, silicon grease-coated stainless steel, oil-saturated Millipore membrane filter, oil-saturated Teflon membrane filter and oil-saturated sintered stainless steel. The use of coated and uncoated stainless steel plates to collect lead dioxide dust produced no difference in measured mass median diameter (MMD); however, with brass grinding dust, there was a 50% decrease in measured MMD when uncoated stainless steel substrates were used, as compared with coated stainless steel substrates. Oil-saturated Millipore membrane surfaces gave consistently lower MMDs than coated stainless steel surfaces. Coated and uncoated stainless steel gave similar MMDs when used to sample brass pouring fume. Oil-saturated Teflon membrane and oil-saturated sintered metal, surfaces for which the collection efficiency is presumed to be independent of the particle loading, gave MMDs similar to those measured for grease-coated stainless steel. The implications of these comparisons are discussed. It is concluded that bounce characteristics are strongly dependent on aerosol material and the suitability of collection surfaces needs to be determined by field evaluation.  相似文献

A simple and sensitive method for detecting low levels (0.5 and 1.0%, w/w) of co-eluting species in HPLC has been developed. This method is based on the subtraction of normalized peak up-slope and down-slope spectra from that of the apex. Visual inspection of the resultant “difference spectra” allows for a qualitative judgement regarding the integrity of the peak under consideration.  相似文献

Circulating progesterone and prolactin levels were measured in shot and live-caught wild red kangaroos using radioimmunoassays validated for the red kangaroo. The objective of the study was to correlate hormone profiles with reproductive status and determine if red kangaroos follow the general pattern elucidated for other macropodids. During Phase 2a lactation (<70 days) plasma progesterone concentrations were <189 pg/ml (n= 41). This value increased to >600 pg/ml (n= 32) during the transition to Phase 3 lactation (181 to 235 days) when the quiescent corpus luteum and embryo were reactivated. Progesterone concentrations then decreased to <300 pg/ml (n= 29) during dual lactation when females were suckling a neonate and a young at foot. Concentrations of prolactin during Phase 2a were <6 ng/ml (n= 17). Coincident with the period of reactivation of the diapausing blastocyst (181 to 235 days), plasma prolactin concentrations increased to 15 ng/ml (n= 32), then decreased and remained low through the subsequent stage of dual lactation. These results indicate that progesterone and prolactin profiles in wild red kangaroos follow patterns found previously in other macropodid species, the tammar and Bennett's wallabies.  相似文献

A quantitative study of synapse formation in the mouse olfactory bulb has been carried out using serial sections. Volumetric synaptic density as well as absolute number of synapses per olfactory bulb for eight distinct synaptic types have been determined at 15 different ages, from the beginning of synapse formation at embryonic day 14 (E14) to postnatal day 44 (P44). Synapses are first found in appreciable numbers at E15 when both axo-dendritic and a few dendrodendritic synapses occur in the presumptive glomerular layer. Initial synapse formation correlates closely with the reorientation of mitral cells from a primitive tangential to a defintive radial direction. Synapse formation by mitral cell dendrites occurs after mitral cell axons have grown into the future olfactory cortical areas, either simultaneous with or before synapse formation by these axons. Virtually all synaptic types detected in adults have been found on the day of birth, consistent with the idea that olfaction is an important sensory modality for newborn mice. Volumetric density of a given synaptic type generally increases 50–100 times during development while the absolute number increases about 1,000 times. Synapses in glomeruli develop more precociously than those in the external plexiform and internal granular layers, which correlates well with the time of origin and differentiation of the principal postsynaptic elements of these two divisions (mitral cells and internal granule cells) Correlation of the time of synapse formation of various synaptic types with their putative excitatory or inhibitory role determined in adult studies suggests that excitatory synapses generally form somewhat earlier, although throughout nearly all of synaptic development, both excitatory and inhibitory synapses are present. Reciprocal dendro-dendritic synapses in the external plexiform layer appear to have a special mode of formation. It is suggested that a granule-to-mitral dendro-dendritic synapse only forms next to an already existing mitral-to-granule synapse on the same gemmule.  相似文献
