A novel temperate bacteriophage was isolated from a Bacillus cereus cereulide-producing strain and named vB_BceS-IEBH. vB_BceS-IEBH belongs to the Siphoviridae family. The complete genome sequence (53 kb) was determined and annotated. Eighty-seven ORFs were detected and for 28, a putative function was assigned using the ACLAME database. vB_BceS-IEBH replicates as a plasmid in the prophage state. Accordingly, a 9-kb plasmid-like region composed of 13 ORFs was identified. A fragment of around 2000 bp comprising an ORF encoding a putative plasmid replication protein was shown to be self-replicating in Bacillus thuringiensis. Mass spectrometry analysis of the purified vB_BceS-IEBH particle identified 8 structural proteins and enabled assignment of a supplementary ORF as being part of the morphogenesis module. Genome analysis further illustrates the diversity of mobile genetic elements and their plasticity within the B. cereus group. 相似文献

A close relationship between tumor angiogenesis, growth, and carcinomatosis has been observed. Netrin-4 (NT-4) has been shown to regulate angiogenic responses. We aimed to examine the effects of NT-4 on colon tumor angiogenesis, growth, and carcinomatosis. We showed that NT-4 was expressed in human colon cancer cells (LS174). A 20-fold increase in NT-4 expression was stably induced by NT-4 pcDNA in LS174 cells. In vivo, a Matrigel angiogenesis assay showed that NT-4 overexpression altered vascular endothelial growth factor (VEGF)/basic fibroblast growth factor-induced angiogenesis. In nude mice with LS174 xenografts, NT-4 overexpression inhibited tumor angiogenesis and growth. In addition, these NT-4-involved inhibitory effects were associated with decreased tumor cell proliferation and increased tumor cell apoptosis. Using an orthotopic peritoneal carcinomatosis model, we demonstrated that NT-4 overexpression decreased colorectal cancer carcinomatosis. Moreover, carcinomatosis-related ascites formation was significantly decreased in mice transplanted with NT-4 LS174 cells versus control LS174 cells. The antiangiogenic activity of NT-4 was probably mediated by binding to its receptor neogenin. Netrin-4 had a direct effect on neither in vitro apoptosis and proliferation of cultured LS174 cells nor the VEGF-induced acute increase in vascular permeability in vivo. We propose that NT-4 overexpression decreases tumor growth and carcinomatosis, probably via an antiangiogenic effect, underlying the potential therapeutic interest in NT-4 in the treatment of colorectal cancer growth and carcinomatosis. 相似文献

Recent studies indicate that cognitively intact individuals experience frequent rightward collisions while walking through
narrow doorways. Such a directional bias has been attributed to an attentional bias in spatial perception. However, these
studies did not investigate the involvement of any motor factor that could affect the directional bias in the body. In the
present study, three experiments were conducted to quantify the impact of the leading foot when crossing a doorway threshold
on the directional bias in the body midpoint when passing through the doorway. Participants walked through the perceived center
of a relatively wide doorway. Measurements of the deviation of the upper-body midpoint from the center of the doorway demonstrated
that the leading foot had a very strong influence on the directional bias. Some participants showed rightward deviation irrespective
of which foot was used to step through the doorway (Experiment 1). However, a consistent rightward bias in the body was not
observed in other experiments. Both the movement of one hand (Experiment 2) and covert visual attention to one side of the
door (Experiment 3) caused contralateral deviation of the body. It is likely that the movement of the hand and a visual stimulus
serve as an attentional cue and are effective to avoid neglect of the ipsilateral side; as a result, the body midpoint is
deviated to the contralateral side. From these findings, we conclude that the directional bias in locomotor trajectories while
passing through a doorway results from the combination of a motor factor, particularly the leading foot, and attentional/brain
