The alar plate of the prosencephalon differentiates into a tectum-like structure when transplanted into the mesencephalon around the 10-somite stage. Here, we report on the projection pattern of the retinal ganglion cells to the transplants. Optic nerve fibers were labeled with horseradish peroxidase (HRP) and 3H-proline, and the innervation of the optic nerve fibers to the chimeric tectum was analyzed by HRP histochemistry on whole-mounted specimens, by autoradiography and by electron microscopy on embryonic day 16. In the chimeric tectum, the transplant was distinguished from the host by difference in nuclear structure between the quail and the chick cells. It was shown that the transplant had the laminar pattern of the optic tectum when the transplant was integrated into the host mesencephalon. The whole-mount HRP histochemistry showed that the optic nerve fibers extend to the transplants. Autoradiography showed that the distribution pattern of silver grains was similar in both the host and the transplant. These results may indicate that the optic nerve fibers turn to the transplant and terminate on the transplant. Electron microscopy further confirmed that optic nerve fibers ended by making synaptic contacts with the dendrites in the transplant region of the tectum. These results indicate that the transplant with the laminar pattern of the optic tectum is a true tectum receiving input from the eye.  相似文献

Hepatic angiomyolipoma (AML) is very rare and only about 80 cases have been reported. The tumor is fundamentally heterogeneously composed of the three tissue components of blood vessels, smooth muscle cells (SMC), and fat cells. Two cases of hepatic AML are reported here, both of which are histologically composed predominantly of a fat cell element and resembled true lipoma (lipomatous AML). However, careful examination of both tumors revealed the presence of a small amount of epithelioid SMC, especially around blood vessels. Immunohistochemical study using monoclonal antibody for melanoma (HMB-45) clearly revealed a small amount of HMB-45-positive SMC around the blood vessels and scattered in the diffuse fat cell growth in both tumors. Since no liver tissue components or primary liver tumors are reactive with HMB-45 except AML cells, the presence of HMB-45-positive cells within the tumor clearly established the diagnosis of hepatic AML. Any fatty tumor or focal fatty lesion of the liver that superficially resemble true lipomas should be tested for the presence of HMB-45-positive SMC in the tumor to differentiate it from AML.  相似文献

Although most kinase-defective growth factor receptor proteins are associated with pathogenic conditions, a kinase-defective Eph-family receptor protein, EphB6, is expressed in normal human tissues. We generated monoclonal antibodies specific for human EphB6 to characterize its expression on human hematopoietic cells. A very small population of normal human peripheral white blood cells (0.57 +/- 0.07%, n = 12) expressed EphB6. The EphB6-positive cells were CD2+, CD7+, CD3+ and CD4+ or CD8+ lymphocytes, but they did not express CD19 or CD11b. In human bone marrow, only 1.5 +/- 0.19% of lymphocytes expressed EphB6. Compared with the expression in peripheral lymphocytes, prominent expression of EphB6 protein was demonstrated in CD4+CD8+ double-positive mouse thymocytes. The T-cell lineage-specific expression was strictly conserved in human leukemia/lymphoma cells. Among T-cell-derived leukemia cells, the expression level of EphB6 seemed to decrease with maturation of the cells. These results suggest that EphB6 expression is regulated in T-cell development.  相似文献

A positive regulator was identified within a 2.3 kb fragment of the 6.4 kb mouse bacteremia region (mba region) of the virulence pKDSC50 plasmid of Salmonella choleraesuis. Sodium dodecyl sulphate polyacrylamide gel electrophoresis showed that Escherichia coli K-12 carrying the recombinant plasmids of the 2.3 kb fragment produced Mba1 protein with a molecular mass of 32 kDa. The recombinant plasmids carrying a 4.1 kb fragment, the other part of 6.4 kb region, produced Mba2 (32 kDa), Mba3 (70 kDa) and Mba4 (29 kDa) proteins. All three proteins were expressed by using the lacZ promoter under isopropyl thiogalactoside induction. In contrast to this, Mba3 protein was overexpressed independently of the lacZ promoter when the 2.3 kb fragment coexisted either in cis or trans. These results suggest that Mba1 is a trans-acting positive regulator for the expression of the Mba3 protein of mba region of pKDSC50.  相似文献

To examine early intercellular junction changes caused by treatmentwith 9, 10-dimethyl-l, 2-benzanthracene (DMBA), rat lingualepithelium was cultivated in isolation and observed by electrophysiological,freeze-fracture and whole-mount electron microscopy. Electrophysiologicalmeasurements showed a transient decrease in membrane potentialof -10.2 mV 6 h after the treatment. It returned to almost thesame level as that of the control group 1 day later. Six hoursafter treatment, input resistance decreased rapidly to 5.3 Mbut increased to 18.0 M 12 h after treatment. Transient reductionof input resistance and membrane potential occurred prior tothe decrease in the coupling ratio 6 h after treatment withDMBA. In freeze-fracture replicas, the number of gap junctionsdecreased by 45% of the control value 6 h after treatment withDMBA. At 12 h and thereafter, the number and area of gap junctionssubsequently decreased by 60–80% of the control value.Alterations in the number and area of desmosomes were similarto those of the gap junctions. The formation of epithelial cytoskeletons,partially devoid of the 2–4 and 5–8 nm filamentswas also observed. A decrease in the density of filament networksbeneath the plasma membranes was especially apparent. Treatmentwith a carcinogen brought about morphological cellular changesas early as 6 h after treatment, and such early changes mighttrigger metabolic cellular abnormalities. Affected cells appearto move away from normal cells in a process of repeated destructionand revision of intercellular junctions, and cytoskeletons.  相似文献

PurposeThe aim of this study was to assess the association between the implementation of abdominal angiography and outcome among pediatric patients with blunt splenic or hepatic injury.MethodsThis was a retrospective observational study, with a study period of 14 years, from January 2004 to December 2017. Blunt-trauma patients with splenic or hepatic injury who were less than 19 years old were included in this study. We used propensity-score-(PS) matching analysis to assess the relationship between abdominal angiography and in-hospital mortality.ResultsIn total, 639 patients were eligible for analysis, with 257 patients included in the abdominal-angiography group and 382 patients in the no-abdominal-angiography group. After PS matching, 224 patients from each group were selected. In the PS matched patients, in-hospital mortality was lower in the abdominal-angiography group than in the no-abdominal-angiography group (4.9% vs. 11.2%, odds ratio 0.416, 95% confidence interval 0.177–0.903).ConclusionIn this population, the implementation of abdominal angiography was significantly associated with lower in-hospital mortality among pediatric patients with blunt splenic or hepatic injury compared with nonimplementation of abdominal angiography.Type of studyPrognosis study.Level of evidenceIII  相似文献
