The lysis of antigen presenting cells (APCs) by cytotoxic T lymphocytes (CTLs) may be one mechanism whereby an immune response is downregulated by Staphylococcus superantigens. Disappearance of monocytes/macrophages from staphylococcal enterotoxin A (SEA)-activated peripheral blood mononuclear cell (PBMC) cultures, but not from control PBMC cultures was seen by flow cytometry. Recently, adenosine triphosphate (ATP) has been described as an effector molecule in CTL-mediated lysis of some murine tumor target cells. We have also shown that ATP caused the lysis of human macrophages, and that treatment of cells with interferon gamma (IFN gamma) rendered macrophages significantly more sensitive to ATP than untreated cells. To show that this purine nucleotide may play a role in modulating the immune system, we generated human CTLs that were stimulated with SEA, and used them as effector cells against SEA-pulsed autologous macrophages. CTLs were found to specifically lyse SEA-pulsed macrophages, while control, unpulsed, macrophages were unaffected. The addition of hexokinase, an enzyme that hydrolyzes ATP, significantly abrogated the killing of SEA-pulsed cells during the assay. In examining the mechanism of cytotoxicity, electron microscopy showed that macrophages incubated with both ATP and CTLs underwent necrosis, rather than apoptosis. From these results, it is suggested that ATP is released from CTLs during antigen presentation, and that IFN gamma- activated macrophages, which are inherently more sensitive to this mediator, are readily lysed and therefore removed from circulation, thus downregulating an immune response.  相似文献

Background Skin cancer surgery is usually performed on the face, and more specifically on and around the midfacial apertures. Each defect generated by the exeresis of a face tumor can be repaired by various advancement, rotation or transposition flaps techniques. Therefore the choice of the closure axis of a defect and its cicatricial and orificial consequences is decisive. Results The closure of a defect is usually made by symmetrically suturing its edges, across the incision axis according to the rule of halves. However, the closure axis of a defect is intentional and characterized by the subcutaneous suture axis which determines the induced tension orientation, and thus the possible displacement of the aperture free margins. The horizontal stretching principle guarantees the lack of impact on the 3 major apertures of the midfacial frame: eye, nostril and mouth. Conclusions This biomechanical concept is decisive to repair midfacial defect with functional and aesthetic results. It also provides objective arguments for the reparative techniques prioritization and the ability to codify those to be recommended as first‐line treatment in the surgical management of the face cutaneous tumours.  相似文献

Summary A comparative study of serial anatomic sections in the transverse, frontal and sagittal planes with corresponding MRI sections of the pelvis allowed the authors to define the most suitable sectional planes and MRI modes for a morphologic study of the levator ani muscle. This study shows the value of MRI examination in the assessment of anorectal malformations.

Résumé A partir de l'étude comparative de coupes anatomiques dans les plans transversal, frontal, sagittal et des coupes IRM correspondantes du petit bassin, les auteurs déterminent quels sont les plans de coupe et les modes IRM les plus performants pour l'étude morphologique des muscles élévateurs de l'anus. Cette étude montre l'intérêt de l'examen IRM dans le bilan des malformations ano-rectales opérées.

The Opacity Lensmeter is a new instrument permitting the measurement of lens opacity. This preliminary report was performed on 98 patients (195 phakic eyes). Age, sex, lens opacity and retinal sensibility (using automated perimetry with the Octopus 2000 R) were assessed. A correlation between age and lens opacity was found in a simple linear regression model on 186 eyes (available data): opacity = 0.00423 square (age) + 5.18. An another more precise correlation was found in 138 eyes, excluding those with anterior segment abnormalities and corneal opacity: opacity = 0.0035 square (age) + 5.17. No statistical differences were observed between lens opacity and sex nor were left/right differences significant. No conclusion was possible concerning retinal sensibility and measured lens opacity because of the small sample (25 eyes). This easily handled instrument may be helpful in daily practice to evaluate opacity in lens pathology. Furthermore, it should be useful for the interpretation of the respective role of cataract and retinal damage in P.O.A.G. loss of retinal sensibility.  相似文献

The decarboxylation of naproxen (1H) and its salt (1-) was achieved by means of chemical [Ce(IV) or S2O8(2-)] and electrochemical oxidation. The product patterns were compatible with mechanisms involving single-electron transfer from the pi-system or the carboxylate moiety. The results are discussed in connection with the involvement of electron-transfer processes in the reported phototoxicity of naproxen.  相似文献
