Neonatal hypoxia–ischemia (HI) is a common occurrence in preterm and low‐birth‐weight infants, and the incidence of low‐birth‐weight and preterm births is increasing. Characterization of brain injury after HI is of critical importance in developing new treatments that more accurately target the injury. After severe HI, neuronal cells undergo necrosis and secondary apoptosis of the surrounding cells as a result of neuroinflammation. We sought to characterize the biochemical pathways associated with cell death after HI. Bax, a cell death signaling protein, is activated after HI and translocates to the nucleus, endoplasmic reticulum, and mitochondria. The translocation patterns of Bax affect the resultant cell death phenotype (necrotic or apoptotic) observed. Although Bax is known to oligomerize once it is activated, less is known about the factors that control its translocation and oligomerization. We hypothesize that Bax kinase‐specific phosphorylation determines its oligomerization and intracellular localization. Using well‐established in vivo and in vitro models of neonatal HI, we characterized Bax oligomerization and multiorganelle translocation. We found that HI‐dependent phosphorylation of Bax determines its oligomerization status and multiorganelle localization, and, ultimately, the cell death phenotype observed. Understanding the mechanisms of Bax translocation will aid in the rational design of therapeutic strategies that decrease the trauma resulting from HI‐associated inflammation. © 2013 Wiley Periodicals, Inc.  相似文献

To assess mercury exposure and potential risk, total mercury (THg-H) and methylmercury (MeHg-H) in hair were studied in 160 adults. The study group consisted of 60 individuals living in the north central coast of Venezuela. A section of the area was known to be contaminated with mercury from a chlor-alkali plant installed near one of the tributary rivers of the Caribbean Sea. The study group was selected from 4 inclusion criteria points. The control group was composed of 100 individuals selected from Carabobo state with no known exposure to Hg. A questionnaire was designed to collect demographic, health information, work activities and fish consumption habits. Hair samples were analyzed for THg. Samples with THg-H > 5 microg/g were also analyzed for MeHg. The mean THg-H was 1.88 +/- 1.50 and 0.99 +/- 0.87 microg/g for the study and control groups, respectively. The study group was statistically higher than control individuals, however, no statistical differences of THg-H were found between each of the 4 categories of both groups. Mean MeHg-H value was 3.67 +/- 1.25 microg/g. Associations were made between Hg-H and several variables. No significant relationship was noted between Hg-H levels and clinical symptoms. R analyses and t-tests were used to determine associations between questionnaire variables and THg-H. The main predictors of THg-H levels in the study group were fish consumption and frequency. As both groups presented relatively low values for THg-H and MeHg-H, the results of this study indicate that Hg exposure did not exceed safe levels. However, a more in-depth exposure assessment should be conducted to more accurately assess this exposure, specifically in terms of Hg content in water and fish sampling.  相似文献

The International Agency for Research on Cancer (IARC) evaluates causes of cancer with help from independent international experts in an open and transparent manner. Countries, research and regulatory agencies, and other organizations adopt IARC evaluations for communication of human cancer hazards, and for strategies to prevent cancer. Scientists worldwide endorse IARC cancer evaluations and process. Those with economic interests, however, challenge IARC's cancer evaluations, most recently for glyphosate and red and processed meats, and are conducting a campaign including intervention from US Congressional Representatives to discredit IARC's review process and to undermine financial support—a campaign intimidating to IARC and Working Group members. Challenges to scientific interpretations serve to advance science and should be resolved by scientific experts who do not have conflicts of interest. Such interference does not bode well for the free flow of scientific information that informs and protects the public from risks of cancer.  相似文献

Cardiac tumors in children are rare and the majority are benign. The most common cardiac tumor in children is rhabdomyoma, usually associated with tuberous sclerosis complex. Other benign cardiac masses include fibromas, myxomas, hemangiomas, and teratomas. Primary malignant cardiac tumors are exceedingly rare, with the most common pathology being soft tissue sarcomas. This paper provides consensus-based imaging recommendations for the evaluation of patients with cardiac tumors at diagnosis and follow-up, including during and after therapy.  相似文献

Primary hepatic malignancies are relatively rare in the pediatric population, accounting for approximately 1%–2% of all pediatric tumors. Hepatoblastoma and hepatocellular carcinoma are the most common primary liver malignancies in children under the age of 5 years and over the age of 10 years, respectively. This paper provides consensus-based imaging recommendations for evaluation of patients with primary hepatic malignancies at diagnosis and follow-up during and after therapy.  相似文献
