Gomisin N, which is a lignan isolated from Schisandra chinensis, has some pharmacological effects. However, the anti-inflammatory effects of gomisin N on periodontal disease are uncertain. The aim of this study was to examine the effect of gomisin N on inflammatory mediator production in tumor necrosis factor (TNF)-α-stimulated human periodontal ligament cells (HPDLC). Gomisin N inhibited interleukin (IL)-6, IL-8, CC chemokine ligand (CCL) 2, and CCL20 production in TNF-α-stimulated HPDLC in a dose-dependent manner. Moreover, we revealed that gomisin N could suppress extracellular signal-regulated kinase (ERK) and c-Jun N terminal kinase (JNK) phosphorylation in TNF-α-stimulated HPDLC though protein kinase B (Akt) phosphorylation was not suppressed by gomisin N treatment. In summary, gomisin N might exert anti-inflammatory effects by attenuating cytokine production in periodontal ligament cells via inhibiting the TNF-α-stimulated ERK and JNK pathways activation. 相似文献

Prosaposin, the precursor of the sphingolipid hydrolase activator proteins called saposins A, B, C, and D, is abundant in the nervous system and muscles. Besides its role as the precursor of saposins, prosaposin is reported to function as a neurotrophic factor, initiating neural differentiation and preventing neuronal cell death in vivo and in vitro. In this study, we examined the localization and synthesis of prosaposin in the rat cochlea. Intense prosaposin immunoreactivity was observed in the organ of Corti, stria vascularis, and spiral ganglion. In an immuno-electron microscopic study, prosaposin immunoreactivity was found mainly in lysosomal granules of the cells in these regions. In the lysosome, prosaposin does not always colocalize with cathepsin D, but was localized mainly in the dark area of the lysosome. Prosaposin mRNA was observed in these same regions. Our results suggest that prosaposin plays a role in homeostasis in the peripheral auditory system. 相似文献

Aim: Intrahepatic bile ducts are the targets for inflammation in primary biliary cirrhosis (PBC), but their pathogenesis is not known. Gram-positive bacterial DNA was detected recently in gallbladder bile of PBC patients. In the present study, we assessed the possible pathological role of lipoteichoic acid (LTA), the Gram-positive bacterial cell wall component, in PBC.Methods: Liver samples, obtained from 20 patients with PBC (stage 1–2 with CNSDC: stage 3–4 with loss of bile ducts = 10:10) and from 13 patients with chronic hepatitis due to hepatitis C virus (CH–C) with lymphocytic cholangitis, were subjected to immunohistochemical staining with polyclonal rabbit anti-LTA as the primary antibody. Serum reactivities to LTA were studied by ELISA. After 1 μg of purified LTA was placed in a 96-well microplate as an antigen, an antibody capture assay was carried out using serum samples from PBC (n = 20), CH–C (n = 13) and healthy subjects (n = 11).Results: LTA was localized around the sites of chronic non-suppurative destructive cholangitis (CNSDC) in the portal area in stage 1–2 PBC but was not detected in the portal area in CH–C. In stage 3–4 PBC, LTA was localized around sites of ductular proliferation at the periphery of portal tracts. IgM class anti-LTA serum titers were significantly higher in PBC than in CH–C. IgA class anti-LTA serum titers were significantly higher in PBC than in healthy subjects.Conclusions: In the PBC livers, the profile of immunoreactivity to LTA changed markedly as the disease progressed. Sera from PBC showed higher levels of anti-LTA titers than CH–C (IgM) or from healthy subjects (IgA). The LTA-mediated immune system might affect the initiation and/or progression of PBC. 相似文献

Objective To determine whether thallium-201 (201Tl) brain single photon emission computed tomography (SPECT) could supplement magnetic resonance (MR) imaging diagnostic information
by visual comparison of two separate data sets from patients with ring-like contrast-enhanced cerebral lesions.
Methods A combination of MR imaging and 201Tl brain SPECT sets obtained from 13 patients (10 men, 3 women) ranging in age from 26 years to 86 years (mean 61.0 years)
were retrospectively reviewed. A total of 12 patients had a solitary lesion, and the others had multiple lesions. All but
two intracranial foci were pathologically confirmed. The final diagnoses were six glioblastomas, two cerebral metastases from
lung cancer, and one each of abscess, resolving hematoma, primary central nervous system lymphoma, toxoplasmosis, and radiation
necrosis. The two separate image formats (MR images and SPECT) were shown to ten readers with practical experience. All of
the MR images for each patient were shown to each reader first. After interpreting them, the readers were shown the SPECT
images. Images were scored in terms of how benign or malignant the foci were on a 5-point scale from “definitely benign” to
“definitely malignant.”
Results The improvement in the performance of all ten readers was from 67.7% to 93.8% in mean accuracy (P = 0.0028) and from 0.730 to 0.971 in mean Az value (P = 0.0069) after they were shown the 201Tl brain SPECT images.
Conclusions
201Tl brain SPECT should substantially increase confidence in the diagnosis of intracranial lesions with ring-like contrast enhancement
when MR imaging does not permit differentiation between benign and malignant disease. 相似文献

Summary Magnetic resonance images of 128 patients with compressive lesions of the cervical spinal canal were reviewed to determine whether a high signal intensity lesion within the spinal cord was present on T2 and proton density weighted spin echo images. Such high signal intensity was observed in 24 cases or 18.8%. The incidence was higher in herniated disk (32.4%), atlanto-axial dislocation (28.6%), and ossification of the posterior longitudinal ligament (22.7%), whereas the abnormality was found sporadically in cervical spondylosis and vertebral body tumors. The high intensity lesion on T2 weighted images was generally observed in patients with constriction or narrowing of the spinal cord. The lesion was not demonstrated on T1 weighted spin echo images. Spinal cord constriction or localized narrowing seemed to be the most important portant predisposing factor in producing such a high signal intensity. The pathophysiologic basis of such an abnormality was presumed to be myelomalacia or cord gliosis secondary to a long-standing compressive effect of the spinal cord. 相似文献

The Physiological and Operative Severity Score for the Enumeration of Mortality and Morbidity (POSSUM) is widely used to predict surgical complications affecting various organs. However, there are few reports about objective evaluation methods for head and neck surgery. In this study, we retrospectively examined the association between POSSUM score and actual surgical complications of head and neck reconstruction surgery. In total, 711 patients who underwent head and neck reconstruction after cancer extirpation between January 2007 and January 2015 were studied. The predicted risk of complications was calculated using the POSSUM score and compared with the actual rate of perioperative complications. Perioperative complications occurred in 178 (25%) patients, comprising systemic complications in 52 (7%) patients, surgical site infection of the head and neck area in 78 (11%) patients and failure of the free flap in 55 (8.8%) patients. When patients were divided into a perioperative complication group and a no-complication group, a significant difference between the two groups was observed in the predicted postoperative rate calculated from the POSSUM score (p?.0001, odds ratio 1.03, 95% confidence interval?=?1.02–1.04). Furthermore, the cutoff value of the POSSUM score calculated from the receiver operating characteristic curve using Youden’s index was 43.0%. POSSUM is a useful risk indicator for head and neck reconstruction surgery. It is possible to objectively calculate the prediction level using a standard assessment method without adding burden to any medical facility. Therefore, patients may be considered at high risk for perioperative complications when the POSSUM score is 43.0% or higher. 相似文献

This preliminary prospective study was conducted to determine whether immobilization with the arm in external rotation would decrease the rate of recurrence after initial traumatic anterior dislocation of the shoulder. Forty patients with initial shoulder dislocations were assigned to (1) conventional immobilization in internal rotation (IR group, n = 20) or (2) a new method of immobilization in external rotation (ER group, n = 20). The recurrence rate was 30% in the IR group and 0% in the ER group at a mean 15.5 months. The difference in recurrence rate was even greater among those who were aged less than 30 years (45% in the IR group and 0% in the ER group). Immobilization with the arm in external rotation is effective in reducing the rate of recurrence after initial dislocation of the shoulder. 相似文献

In order to study the relations of malocclusion formation and feeding behavior as a scientific research project, we measured the 3-D morphology of 31 healthy newborns within 7 days of birth and 1 month later using dental casts. Nine characteristics were selected and the developmental changes were analyzed using specialized software for investigating palatal development in detail. The results revealed that the palatal width, 3 palatal characteristics relating to depth, maximum slope location, and mean slope increased significantly during the 1 month, but there were no significant changes observed in palate length, cuspid ratio, and maximum slope. There was a significant correlation between palate depth and maximum depth point location, suggesting that the deeper points tended to be located further back. No significant correlations were observed between newborns within 7 days of birth and 1 month after birth for cuspid ratio, maximum depth point location, and maximum slope. Our observation of inter-characteristics correlations revealed 7 significant (P < 0.01) combinations out of a total of 33 combinations were observed in newborns within 7 days of birth, while only 4 were observed in 1-month-old infants. This appears to have been because of morphological changes occurring between individuals due to growth. Thus, it was clarified that not only size but shape (cuspid ratio, maximum slope point location, and the mean slope) also changes over the first month after birth. 相似文献

The majority of lymphoid cells from a patient with non-Hodgkin's lymphoma with leukemic transformation were demonstrated to carry receptors for both sheep erythrocytes and complements by the combined rosette assay using neuraminidase-treated sheep erythrocytes and complement-coated zymosan beads. Most of them were considered morphologically lymphoblasts and were positive for acid phosphatase staining. Terminal deoxynucleotidyl transferase activity was not detected in these cells. Lymphoid cells from this patient did not respond to the stimulation with phytohemagglutinin-P, concanavalin-A, and pokeweed mitogen (PWM). When these cells were cultured with PWM for 7 days, no plasma cell was generated. Although only a few plasma cells were generated in the PWM-stimulated culture of normal purified B cells alone, the addition of the patient's cells to purified normal B cells resulted in a markedly enhanced generation of plasma cells in response to PWM, as was the case with normal T cells. But leukemic cells either from a patient with T-cell leukemia not having complement receptors or from a patient with null-cell leukemia showed no enhancing ability in B- cell differentiation. In addition, the culture supernates of the patient's cells obtained after 24-hr PWM stimulation had an ability to promote B-cell differentiation comparable in activity to those from the PWM-stimulated normal T cells. 相似文献

A restriction site polymorphism in the Lp(a) apolipoprotein gene (the LPA gene) is reported. The basis for the polymorphism is presence or absence of an MspI restriction site that appears to be 3' to the last kringle IV structure of the gene. The "1" gene (presence of the restriction site) has a frequency of 0.316 and the "2" gene (absence of the restriction site) has a frequency of 0.684. Both members of each of 67 monozygotic (MZ) twin pairs had the same genotype and there was Mendelian segregation of the DNA variants in 40 families with a total of 75 children. There was a lower proportion of people with genotype 1-1 in the top quartile than in the 3 bottom quartiles of the population distribution of Lp(a) lipoprotein levels but the difference did not reach statistical significance. 相似文献
