人工冬虫夏草联合糖皮质激素对大鼠肺纤维化的干预作用 总被引:1,自引:0,他引:1
目的:探讨人工冬虫夏草(虫草菌液)联合糖皮质激素对博莱霉素诱导的大鼠肺纤维化的干预作用.方法:50只SD大鼠,随机分为5组:正常对照组、模型组、虫草菌液组、强的松组、虫草菌液联合强的松组(联合组).采用气管内注射博莱霉素建立模型,次日起分别给药灌胃治疗,正常组和模型组给予生理盐水.第28天处死动物,肺组织标本行HE染色及MASSON染色进行病理形态学观察、Ashcroft纤维化评分,碱水解法检测肺组织HYP含量,免疫组化检测肺组织CTGF蛋白表达,酶联免疫法检测肺泡灌洗液上清TGF-β1含量.结果:与模型组相比,虫草菌液、强的松组能够降低肺组织纤维化评分,减少肺组织胶原沉积、HYP含量,下调肺组织CTGF蛋白表达,降低BLAF中TGF-的含量;联合组分别与虫草菌液组、强的松组比较,能够进一步减少肺纤维化评分、胶原面积、HYP含量,进一步下调CTGF蛋白、TGF-蛋白表达.结论:虫草菌液、强的松对博莱霉素诱导的大鼠肺纤维化有治疗作用,且虫草菌液联合强的松能产生协同治疗效果. 相似文献

As the most prevalent route of delivery, oral administration has the challenge of potentially low bioavailability in part
because P-glycoprotein (P-gp) in the intestinal tract affects absorption. Therefore, absorption enhancers or P-gp inhibitors
are strategies to solve this problem. The aim of the present study was to investigate the effects of borneol on transportation
of colchicine and rhodamine123, two P-gp substrates, in rats. In vitro transportation was assessed with a diffusion chamber system with isolated rat intestines. Different concentrations of borneol
(10, 40 and 80 μg/mL) were prepared in solutions with two P-gp substrates compared with blank solutions. The in vivo effects on colchicine were assessed by a pharmacokinetic study. Borneol enhanced the absorptive transport of two P-gp substrates,
which was relevant to the concentration. A pharmacokinetic study showed that in the presence of borneol, a significant increase
in Cmax and AUC0→8 of colchicine occurred when compared to colchicine alone. The study showed that borneol affected two P-gp substrates in the
intestine, possibly by inhibiting the effects of P-gp and enhancing intestinal absorption of drugs. Therefore, borneol could
be developed as a P-gp inhibitor and absorptive enhancer. 相似文献

The McCune-Albright syndrome (MAS) is characterized by a clinical triad of polyostotic fibrous dysplasia, café-au-lait hyperpigmented macules, and hypersecretory endocrinopathies. Acromegaly is an uncommon manifestation of the endocrine disturbance associated with MAS, and the role of surgery in managing these cases has been a topic of debate. The authors present the case of a 35-year-old man with MAS who was also diagnosed with acromegaly, hyperprolactinemia, and pituitary macroadenoma. The patient had an 18-year history of fibrous dysplasia involving the right frontal bone and ribs as well as multiple endocrinopathies, but no cutaneous hyperpigmented macules. An oral glucose tolerance test demonstrated partial suppression of plasma levels of growth hormone (GH). The patient underwent transsphenoidal resection of the pituitary tumor, performed with assistance of neuronavigation, and tolerated the procedure well. After the surgery, both prolactin and GH levels returned to normal. These results suggest that neuronavigation-assisted transsphenoidal surgery can safely remove pituitary adenomas associated with MAS and successfully treat the underlying endocrine abnormalities. 相似文献

MicroRNAs (miRNAs) are important regulators of cell fate decisions, while the miRNAs and their targets in the regulation of stem cell differentiation are largely unidentified. Here we report novel functions of miR-125b/Lin28 axis in the regulation of mouse embryonic stem cell (mESC) lineage specification and cardiomyocyte differentiation. With a MicroRNA Array screen, we identified a number of miRNAs significantly changed during ESC differentiation, among which miR-125b showed a marked reduction during early differentiation. The abundantly expressed miR-125b in undifferentiated mESCs was dramatically downregulated to a level hardly detected during differentiation day 3 to 5, with a concomitant upregulation of Lin28. Ectopically expressing miR-125b did not alter characteristics of undifferentiated mESCs, whereas it impaired the endoderm and mesoderm development, but not the ectoderm, and inhibited cardiomyocyte formation. We further demonstrate that miR-125b targeted the 3'-untranslated region of Lin28 and reduced the abundance of Lin28 at both mRNA and protein levels. Moreover, phenotypes of miR-125b overexpressing cells were mimicked by downregulation of Lin28 and rescued by reintroduction of Lin28. In addition, the impaired cardiogenesis in miR-125b-introduced cells was greatly recovered when mimicking endoderm environment by cultivation with the condition medium from a visceral endoderm-like cell line, END-2. These results reveal that the miR-125b/Lin28 axis is an important regulator of early lineage specification and cardiomyocyte differentiation of ESCs. 相似文献
