研究表明:肿瘤坏死因子(TNF)可剂量依赖地引起牛肺动脉内皮细胞乳酸脱氢酶释放率(LDH%)升高,促进中性粒细胞向内皮细胞粘附,并可抑制内皮细胞增殖和DNA合成。蛋白激酶C(PKC)抑制剂1-(5-异喹啉磺酰基)-2-甲基哌嗪(H-7)和槲皮素一方面可剂量依赖地抑制TNF对内皮细胞的直接损伤,另方面又可通过抑制TNF诱导的中性粒细胞对内皮细胞粘附增加,减轻TNF对内皮细胞的间接损伤作用,同时还可抑制TNF对内皮细胞增殖和DNA合成的影响,从而间接加强内皮细胞对损伤的自我修复。 相似文献

Human neutrophil elastase is thought to play an important role in connective tissue destruction in diseases such as emphysema and arthritis. In this article, it is demonstrated that the elastase activity of mature human peripheral blood neutrophils can be rapidly increased in vitro by treatment of the cells with lipopolysaccharide from E. coli and that this increase is inhibited by corticosteroid pretreatment of the cells and by inhibitors of protein synthesis. Alterations in intracellular enzyme content of the mature polymorph in response to bacterial products or other stimuli may be important for amplification of the inflammatory response. 相似文献

Clinical transplantation requires cold storage of tissue for several hours. We have examined the elemental content in exocrine and endocrine cells in mouse pancreas after cold storage by X-ray microanalysis, and in parallel carried out morphological studies. Tissue was stored at 4 degrees C for 4-12 h in Normal Krebs-Ringer's (high Na+/K+ ratio), Modified Krebs-Ringer's (low Na+/K+ ratio), Euro-Collins, University of Wisconsin (UW) solution, and seven modified version of UW solution. Incubation in Normal Krebs-Ringer's solution caused significantly increased Na and decreased K concentrations in contrast to incubation in other solutions. The cellular concentration of Na and Cl followed the concentration in the storage solution. Changes in the endocrine cells were similar to, but less pronounced than those in exocrine cells. Calcium was retained best in UW and some variants of UW, and least in Euro-Collins. This may indicate differences in preservation of secretory granules. Also, morphological studies showed that endocrine cells were less affected than exocrine cells. In conclusion, the only factor determining the intracellular concentration of diffusible ions after cold tissue storage is the ionic composition of the extracellular medium. The cost of autologous transfusions using semiautomated instruments in 52 orthopedic cases, 75 coronary artery bypass graft (CABG) cases, and 218 aortic aneurysm cases was compared to the cost of equal amounts of homologous blood. While none of the orthopedic cases reached cost equivalence (median cost deficit per case, +97), 31 percent of the CABG cases (median cost deficit per case, +61) and 56 percent of the thoracic aortic aneurysm cases (mean cost surplus per case, +30) did so. In most cases, the major orthopedic and CABG procedures do not reach cost equivalence and might be served better by other means of autologous blood recovery.
