Brain-derived neurotrophic factor (BDNF) stimulates neuronal growth and protects nigral dopamine neurons in animal models of Parkinson disease (PD). Therefore, BDNF is a candidate gene for PD. The authors investigated five single-nucleotide polymorphisms in 597 cases of familial PD. Homozygosity for the rare allele of the functional BDNF G196A (Val66Met) variant was associated with a 5.3-year older onset age (p = 0.0001). These findings suggest that BDNF may influence PD onset age.  相似文献

The role of genetics in Parkinson disease (PD) continues to be an area of considerable interest and controversy. We collected information involving the nuclear families of 948 consecutively ascertained PD index cases from the University of Virginia (UVA) Health System, the University of Medicine and Dentistry of New Jersey-Robert Wood Johnson (RWJ) School of Medicine, and Boston University (BU) School of Medicine. We performed a segregation analysis to assess evidence for the presence of a Mendelian pattern of familial transmission. The proportion of male (60.4%) and female (39.6%) cases, the mean age of onset (57.7 years), and the proportion of affected fathers (4.7%), mothers (6.6%), brothers (2.9%), and sisters (3.2%) were similar across the three sites. While most of the index cases were male, modestly more of the reported affected relatives were female. These analyses support the presence of a rare major Mendelian gene for PD in both the age-of-onset and susceptibility model. The age-of-onset model provides evidence for a gene that influences age-dependent penetrance of PD, influencing age of onset rather than susceptibility. We also found evidence for a Mendelian gene influencing susceptibility to the disease. It is not evident whether these two analyses are modeling the same gene or different genes with different effects on PD. The finding of significant genes influencing penetrance for PD raises the question of whether these may interact with environmental factors or other genes to increase the risk for PD. Such gene environment interactions, involving reduced penetrance in PD, may explain the low concordance rates among monozygotic twins for this disease.  相似文献

The management of cardiac arrhythmias has changed dramatically over the past several years. New drugs and devices are now available to treat various arrhythmias. Many new agents have been developed that rely on different electrophysiologic mechanisms to elicit their effect on the heart rhythm. Though often effective, these drugs also pose a risk because all of them have a variety of potential adverse effects associated with their use. Many of these adverse reactions are common to all antiarrhythmic drugs, whereas others are unique to particular agents. Volumetric rendering is a new approach to three-dimensional (3D) imaging that overcomes many of the drawbacks of currently available surface-rendering systems. Its application on the Pixar Imaging System in two cases of acetabular fracture was assessed to illustrate the features of the technique. The fast-computing architecture and large memory of this system allow rapid generation of a series of high-quality 3D images in each plane of rotation (x or spinal axis, z or somersaulting axis) that can be viewed as independent static images or as an animated real-time video loop. Editing to remove the normal contralateral hemipelvis enhances appreciation of acetabular abnormalities. Every pixel of computed tomographic data is preserved, allowing representation of both soft tissue and bone as translucent overlap. The presentation of data also allows detection of subtle abnormalities and features and minimizes the artifact generation common in surface-rendered images.  相似文献

The formation of condensed ring polycyclic aromatic hydrocarbons (PAHs) from the pyrolysis of ground tobacco in helium over the temperature range of 350-600 degrees C was investigated. PAH yields in the ng/g range were detected and the maximum yields of all PAHs studied including benzo[a]pyrene (B[a]P) and benzo[a]anthracene (B[a]A) occurred between 500 and 550 degrees C. The pathway to PAH formation in the 350-600 degrees C temperature range is believed to proceed via a carbonization process where the residual solid (char) undergoes a chemical transformation and rearrangement to give a more condensed polycyclic aromatic structure that upon further heating evolves PAH moieties. Extraction of tobacco with water led to a two fold increase in the yields of most PAHs studied. The extraction process removed low temperature non-PAH-forming components, such as alkaloids, organic acids and inorganic salts, and concentrated instead (on a per unit weight basis) tobacco components such as cell wall bio-polymers and lipids. Hexane extraction of the tobacco removed lipophilic components, previously identified as the main source of PAH precursors, but no change in PAH yields was observed from the hexane-extracted tobacco. Tobacco cell wall components such as cellulose, hemicellulose, and lignin are identified as major low temperature PAH precursors. A link between the formation of a low temperature char that evolves PAHs upon heating is established and the observed ng/g yields of PAHs from tobacco highlights a low temperature solid phase formation mechanism that may be operable in a burning cigarette.  相似文献

Quantitative in vitro autoradiography was used to study the distribution of [3H]hemicholinium-3 ([3H]HC-3) binding sites in the rat brain. Regional concentrations of HC-3 binding sites were corrected for regional tissue quenching of tritium in a number of brain structures. Specific binding of 10 nM [3H]HC-3 was highest in the interpeduncular nucleus, followed by the caudate-putamen, olfactory tubercle, amygdala, and the medial and lateral habenulae. There was a high positive correlation between regional HC-3 binding and choline acetyltransferase activity in rat brain; however, a novel pattern of the distribution of cholinergic terminals in the subnuclei of the interpeduncular nucleus was discovered. The apparent Kd in the 1-5 nM range and the pharmacological specificity of the HC-3 binding site agreed with data for choline uptake and for the HC-3 binding site as determined in membrane preparations. HC-3 autoradiography appears to be a useful anatomical marker for cholinergic terminals.  相似文献
