Embryo transfer (ET) is the last stage of extracorporal fertilization during which the embryo is placed in the uterine cavity with a medium-filled catheter 2–3 days after in vitro fertilization. While fertilization in the laboratory occurs at very high rates (>:90%), the overall success of the procedure (i.e., take home baby) is still very low (<25%) and assumed to be mainly due to implantation failure. A computational model was developed to simulate ET within the uterine cavity by a fluid-filled catheter inserted into a two-dimensional channel with oscillating walls. The results showed that the speed at which the embryos are injected from the catheter dominates the procedure and controls the velocity of their transport within the uterine cavity. ET at excessively high injection speeds may lead to ectopic pregnancies, while uterine peristalsis affects transverse dispersion only during injection at low injection speeds. The presence of the catheter within the uterus does not affect flow patterns downstream of its tip. Materials are distributed throughout the body of mammals by fractal networks of branching tubes. Based on the scaling laws of the fractal structure, the vascular tree is reduced to an equivalent one-dimensional, tube model. A dispersion–convection partial differential equation with constant coefficients describes the heterogeneous concentration profile of an intravascular tracer in the vascular tree. A simple model for the mammalian circulatory system is built in entirely physiological terms consisting of a ring shaped, one-dimensional tube which corresponds to the arterial, venular, and pulmonary trees, successively. The model incorporates the blood flow heterogeneity of the mammalian circulatory system. Model predictions are fitted to published concentration-time data of indocyanine green injected in humans and dogs. There are various kinds of diagnostic examinations in the field of clinical cardiology. In this field, information concerning cardiac structure, dimensions (hypertrophy and dilatation) and cardiac functions are inevitably important. Noninvasive methods are desirable. Except for radiological examinations, electrocardiography and ultrasonic echocardiography are 2 main examinations in cardiology. Holter monitoring is one of the recent topics in clinical electro-cardiology. It is useful for the detection of rest angina or variant angina. Recently "silent myocardial ischemia" has been studied by using this method. This method is also useful for the study of arrhythmia, which is a major cause of sudden death. Quantitative analysis of arrhythmia in the studies of antiarrhythmic drugs can also be done by Holter monitoring. Late potential is also a topic in electro-cardiology. Ultrasonic echocardiography can detect the changes of cardiac structure and function. Doppler echocardiography, developed recently, has made it possible to estimate intracardiac pressure (especially pulmonary arterial pressure) noninvasively. With esophageal echocardiography findings in left atrium, such as thrombus or myxoma in left atrium can be obtained. The most appropriate tool must be selected from the many kinds of diagnostic examinations for efficient clinical diagnosis and therapy.  相似文献

The caecilians (Amphibia, Gymnophiona) constitute one of the least known groups of terrestrial vertebrates because most species live underground in quite inaccessible environments. Siphonops annulatus is an exclusively fossorial species and is the most extensively distributed caecilian in South America. Little is known of this order concerning circulating granulocytes, including their morphological and cytochemical structure and ultrastructure. This paper is part of a project covering the study of granulocytes in representative species of the order Amphibia. Blood extensions were carried out and submitted to Leishman, Toluidine Blue, Periodic acid Schiff, Sirius Red and hydrogen o-toluidine peroxide methods. Part of the samples was prepared for conventional transmission electron microscopy. Among granular leukocytes, mature and immature neutrophils and eosinophils were identified, plus basophils. The most frequent granulocyte encountered in S. annulatus peripheral blood is the neutrophil. This is a cell with a hyper-segmented nucleus and with a very clear cytoplasm when compared to the eosinophil, which presents large cytoplasmic acidophilic granules. On the other hand, the basophils present basophilic and metachromatic granules. Glycogen was detected in the cytoplasm of the neutrophils and eosinophils, while basic protein rich in amino acids was observed in the eosinophil’s granules. Myeloperoxidase activity was detected in the cytoplasm of the neutrophils and eosinophils. Neutrophils were ultrastructurally detected with three types of small granules: eosinophils with large and small spherical granules and basophils with large spherical granules with lamellate structures.  相似文献

The Sendai virus pi strain (SeVpi) isolated from cells persistently infected with SeV shows mainly two phenotypes: (1) temperature sensitivity and (2) an ability of establishing persistent infection (steady state). Three amino acid substitutions are found in the Lpi protein and are located at aa 1088, 1618, and 1664. Recombinant SeV(Lpi) (rSeV(Lpi)) having all these substitutions is temperature sensitive and is capable of establishing persistent infection (steady state). rSeVs carrying the fragment containing L1618V show both phenotypes. rSeV(L1618V), in which leucine at aa 1618 is replaced with valine, has the ability of establishing persistent infection, but is not a temperature-sensitive mutant, indicating that the ability of a virus to establish persistent infection can be separated from temperature sensitivity. The amino acid change at 1618(L-->V) coexisting with aa 1169 threonine is required for acquirement of a temperature-sensitive phenotype. Three amino acid substitutions are also found in the Ppi protein, but rSeV(Ppi) does not show these phenotypes.  相似文献

Electron microscopy has been the ‘gold standard’ of spatial resolution for studying the structure of the cell nucleus. Electron spectroscopic imaging (ESI) offers advantages over conventional transmission electron microscopy by eliminating the need for heavy-atom contrast agents. ESI also provides mass-dependent and element-specific information at high resolution, permitting the distinguishing of structures that are primarily composed of protein, DNA, or RNA. The technique can be applied to understand the structural consequences of epigenetic modifications, such as modified histones, on chromatin fiber morphology. ESI can also be applied to elucidate the multifunctional behavior of subnuclear ‘organelles’ such as the nucleolus and promyelocytic leukemia nuclear bodies. The authors dedicate this paper to the memory of Ying Ren (1961–2007). We all benefited from knowing her. Our research advanced through the technical creativity she provided.  相似文献

In immobilized decerebrate cats fictive locomotion was evoked by midbrain stimulation to analyse the efferent pattern to elbow and to distal forelimb muscles innervated by the deep radial nerve. The locomotor activity was assessed by recording nerve discharges and motoneuronal membrane potential changes. The elbow flexor and extensor motoneurons showed a reciprocal activity; the membranes were correspondingly depolarized and hyperpolarized. In the motor nuclei to the wrist and digit extensors the active phases changed systematically according to the radio-ulnar order of the muscles: the extensor carpi radialis (ECR) was flexor-coupled, the ulnaris (ECU) extensor-coupled, the digitorum communis (EDC), the lateralis (EDL) and the indicis proprius (EIP) displayed intermediate patterns. Intracellular recordings from these motoneurons revealed in all motor nuclei, except ECR, a double depolarization. The first occurred early and the second later in the flexor phase; a hyperpolarization was interposed. The second depolarization mainly determined the active phase. According to the radio-ulnar order of the muscles the onset and termination of the second depolarization were delayed. This was presumably due to the interposed hyperpolarization, which progressively increased in amplitude. The ECR exhibited a single depolarization, into which the double depolarization apparently merged. The other radial motor nuclei, supinator (Sup) and Abductor pollicis longus (APL) displayed complex patterns. Sup showed tonic discharges, flexor-type discharges or discharges extending both into the flexor and extensor phase, APL showed discharges similar to either EIP or Sup. Membrane potential changes were small in APL and Sup. Thus, the central locomotor network generates differentiated efferent activities in the distal forelimb muscles, the radio-ulnar order of the muscles being important for the generated pattern.  相似文献
