During orthodontic treatment, multinucleated clast cells carry out the resorption of mineralized tissues. Adhesion of clast cells to the mineralized tissues is mediated by transmembrane cell-surface glycoproteins called integrins, specifically by the alphavbeta3 integrin, which plays an important role in the process of bone resorption. The role of the alphavbeta3 integrin in bone resorption leading to osteoporosis has been demonstrated, but its role in alveolar bone and root resorption during orthodontic tooth movement is unknown. This study examined the expression of the alphavbeta3 integrin during experimental tooth movement. Tooth movement was achieved in 16 male Sprague-Dawley rats (each weighing 120-200 g) with elastic bands between their maxillary first and second molars. The molar-bearing segments were dissected and processed for histologic and immunohistochemical examination. The expression of alphavbeta3 integrin was examined with 2 primary antibodies: a polyclonal anti-alphav integrin subunit antibody and a polyclonal anti-beta3 integrin subunit antibody. Negative controls were similarly processed but without incubation with primary antibodies. The alphavbeta3 integrin was expressed both by osteoclasts associated with alveolar bone resorption and by odontoclasts associated with root resorption during experimental tooth movement. Furthermore, the beta3 integrin subunit was expressed by the epithelial rests of Malassez in the periodontal ligament. Negative controls did not show immunolabeling. The alphavbeta3 integrin adhesion receptor is expressed during experimental tooth movement and might be involved in the process of mineralized tissue resorption and the functions of the epithelial rests of Malassez.  相似文献

The objectives of this study were to investigate the structure of Invisalign appliances (Align Technology, Santa Clara, Calif) after intraoral exposure, and to qualitatively and quantitatively characterize the substances leached from the aligners after accelerated in vitro aging. Samples of Invisalign appliances were randomly selected from 10 patients before intraoral placement and after retrieval, and the prepared specimens were subjected to (1) bright-field optical reflection microscopy to study the surface morphology; (2) Fourier transform infrared microspectroscopy to characterize the in vivo changes in molecular composition induced on appliance surfaces, (3) scanning electron microscopy and energy dispersive X-ray microanalysis to identify the elemental composition of integuments formed on the surface, and (4) Vickers hardness (HV 200) testing. Another set of reference and retrieved appliances was subjected to artificial aging for 2 weeks, and the extracts were subjected to gas chromatography-mass spectroscopy. The retrieved appliances demonstrated substantial morphological variation relative to the as-received specimens involving abrasion at the cusp tips, adsorption of integuments, and localized calcification of the precipitated biofilm at stagnation sites. Buccal segments of retrieved appliances showed an increase in hardness, which might be attributed to mastication-induced cold work; however, the clinical implication of this effect on mechanotherapy is unknown. In vitro aged and retrieved appliances were found to leach no traceable amount of substances in an ethanol aging solution.  相似文献

BACKGROUND: Bone healing around endosseous dental implants is associated with peri-implant loading conditions. Therefore, the aim of this study was to evaluate histomorphometrically the bone response around unloaded, delayed, and immediately loaded implants with a progressive thread design that were placed in the posterior regions of the lower jaw in monkeys. METHODS: Nine adult monkeys (Macaca fascicularis) were used in this study. After extraction of the second premolars and first and second molars in the mandible, the bone was allowed to heal for a period of 3 months. Forty-eight 8 mm long implants with a diameter of 3.5 mm were placed according to the following protocol. In two of the monkeys, six implants were placed and left to heal submerged for 3 months (group A). In seven monkeys, 21 implants were placed in one side of the mandible and loaded after 3 months of submerged healing (group B). The group B implants were loaded with temporary resin bridges at the same time as another 21 implants that were loaded immediately (group C) after placement in the contralateral side of the mandibles of the same monkeys. The occlusion of group B and C implants was checked for optimal relationship of the resin bridges that were replaced 1 month later with metal bridges and loaded for an additional 2 months. The group A animals were sacrificed after 3 months of submerged healing without loading; group B and C animals were sacrificed after 3 months of implant loading. Specimens were examined histologically and histomorphometrically. RESULTS: All implants osseointegrated without presenting any gap in the metal-bone interface. Compact cortical bone was found in contact with the implant surfaces. Group A implants demonstrated in the interface cancellous bone with loose connective tissue. Group B and C implants showed a thick cortical plate with extensive bone trabeculae formation. There was a significant difference in bone-to-implant contact (BIC) between the various loading conditions. No significant difference (P < 0.05) was found between groups B and C. There was an increased area of bone (BA) within the threads as well as around the apices of group B and C implants. CONCLUSIONS: Implant loading might have stimulated increased bone formation and thus may be a key factor in influencing positive osseointegration. In addition, immediately loaded implants may osseointegrate in a similar manner as delayed loaded implants.  相似文献

PURPOSE: The purpose of this study was to estimate the degree of cure and monomer leaching of a light-cured and a chemically cured (no-mix) adhesive and to assess their biologic properties. MATERIAL: The degree of cure of adhesive specimens prepared with a procedure identical to the clinical bonding process was assessed by infrared spectroscopy. The adhesives were then immersed in normal saline solution for 2 months, and the residual monomer leached from the adhesives was quantitatively and qualitatively analyzed by liquid chromatography. The effect of the immersion media on human gingival fibroblasts' viability and proliferation was also evaluated with the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) and deoxyribonucleic acid (DNA) synthesis assays, respectively. The results were analyzed by analysis of variance and Tukey test (alpha = .05). RESULTS: No difference was found between the 2 adhesives with respect to their degree of cure and the amount of triethylene glycol dimethacrylate released; no diglycidyl dimethacrylate monomer was detected in the eluent. However, significant qualitative changes in the composition of the substances eluted from the 2 adhesives were observed. CONCLUSIONS: Whereas no cytotoxic effect was shown for either immersion media, a moderate reduction in the DNA synthesis was obtained by both adhesives, implying a minor cytostatic effect. Further research is required to assess the long-term biologic properties of adhesives, including potential estrogenic action.  相似文献

The clinical study was designed to study the vertical dimension of rest position and mandibular closure to maximum intercuspation of the teeth before, during, and after relaxation procedures on 14 subjects with stable dentitions. The findings call attention to the potential of both the Myomonitor and the Kinesiograph for research and clinical use, to the physiologic need for flexibility of the vertical dimension of rest position, and to the importance of measuring all three dimensions when examining the rest position.  相似文献

Bleaching tetracycline-stained teeth is the most challenging form of tooth lightening. This article reports on 44 subjects who bleached their tetracycline-stained teeth for 6 months using trays with reservoirs overnight in a half-mouth designed study and 2 of 3 different concentrations of carbamide peroxide (10%, 15% or 20%). The subjects were followed for 5 years. The area evaluated was the middle third of the teeth. More than 55% of tooth lightening occurred within 1 month; after 5 years, more than 65% of the maximum tooth whitening remained for all 3 gel concentrations. Tooth whitening can be accomplished with any of the 3 concentrations used.  相似文献
