Lysine acetylisalicylate inhibits the protein synthesis in the rat gastric mucosa in vivo and in vitro

Lysine acetylsalicylate (LAS), a water soluble derivative of acetylsalicylic acid and parenterally administrable analgesic, locally inhibits the incorporation of 14C protein hydrolysate into proteins of the rat isolated gastric mucosa. A similar effect on protein synthesis was observed after s.c. administration of the drug, whereas DNA and RNA synthesis were not affected. Obviously, LAS inhibits the translation step during biosynthesis of proteins. It is conceivable that this effect contributes to the potential ulcerogenic action of acetylsalicylate and that parenteral administration is not a criteria to protect the stomach from salicylate-induced side effects.     

Distribution of pyrrolidinomethyl-tetracycline (rolitetracycline) and tetracycline in blood and various organs of mice measured by high pressure liquid chromatography

By means of a newly developed high-pressure liquid chromatographic method the organ distribution of tetracycline (TC) and pyrrolidinomethyl-tetracycline (PMT) has been studied. When mice were treated with 50 mg/kg i.v. TC or PMT these antibiotics could be detected in all organs investigated (liver, kidney, heart, lung, muscle, spleen). Especially high concentrations were found in liver and kidneys, where TC and PMT could be detected up to 6 h. In animals treated with PMT part of the PMT applied decomposed slowly yielding TC, which was found together with PMT in blood and all organs.

A high pressure liquid chromatographic method for the determination of tetracyclines in blood and organs of experimental animals

A high-pressure liquid chromatographic method has been developed for the quantitative analysis of tetracyclines from biological material. Tetracyclines can be extracted from serum or organ homogenates by means of acetonitrile containing buffer. The extract is chromatographed on a reversed-phase column and the tetracyclines are monitored spectrophotometerically at 357 nm. Tetracyclines, pyrrolidinomethyl-tetracycline, 4-epitetracycline, and doxycycline are thus separated within 8 min. The lowest concentration for the intact molecule detectable are 0.2 micrograms/ml serum or blood and 0.4 micrograms/g organ. In a comparative study the new extraction procedure and the chromatographic analysis yielded the same results as a microbiological method.     

Der Einfluß des Kaliumcyanid und einiger SH-Gruppen blockierender Stoffe auf die Atmung und den Gehalt von Hefezellen an Coenzym A,anorganischem Phosphat und einigen Adenosinphosphaten

Zusammenfassung Hemmstoffe des Coenzym A. und zwar die anorganischen und organischen Arsen- und Quecksilberverbindungen, Natriumarsenit, Sublimat, Oxarsan, Spirotrypan, Neosalvarsan und Salyrgan sowie Janusgrün B und Monojodacetat und der spezifische Blocker der Cytochromoxydase Kaliumcyanid wurden an Hefezellsuspensionen hinsichtlich ihrer Wirkung auf Atmung, Coenzym A-Aktivität, AMP, ADP und ATP und intracelluläre anP untersucht.Die meisten Stoffe führen innerhalb bestimmter Konzentrationsbereiche zur Entokopplung der von Lynen u. Mitarb. beschriebenen Beziehung zwischen Atmung und Coenzym A-Gehalt der Hefezellen. Denn der Coenzym A-Verlust geht nicht obligatorisch mit einer Senkung der Atmung, die Hemmung der Atmung durch geringe Cyanidkonzentrationen nicht mit einer solchen des Coenzym A einher.Die Abnahme des Coenzym A — sei sie die Folge der Blockierung der funktionellen SH-Gruppen des Coenzym A oder einer Störung der Coenzym A-Synthese — führt, solange die Sauerstoffaufnahme und der stationäre Gehalt an anP keine Änderungen erfahren, zur Erhöhung der ADP, AMP und ATP-Konzentrationen in den Hefezellen. Die Ursache der ATP- und ADP-Zunahme liegt vermutlich unmittelber im Coenzym A-Mangel, der die Hefezellen zur Einschränkung zahlreicher, vom Coenzym A abhängiger, energieverbrauchender Synthesen zwight, die Zunahme der Adenylsäure in einer vom Coenzym A-Mangel mittelbar induzierten Hemmung der Polynucleotidsynthesen. Kaliumcyanid und Sublimat in höheren Konzentrationen schädigen die Atmung und setzen dadurch den Gehalt der Hefezellen an Coenzym A, anP, ATP und ADP herab, während AMP eine der ATP- und ADP-Abnahme entsprechende Zunahme erfährt.Mit 3 Textabbildungen     

Changes in serum transaminases, SDH and liver morphology after treatment with trypanocidal diamidines in mice

The effects of 2 recently developed trypanocidal agents, Diamidinophenylindol (DAPI) and Diimidazolinophenylindol (DIPI) at doses of 10-30 micrograms/g intraperitoneally (i.p.) on serum GOT, GPT and sorbitol dehydrogenase (SDH) levels and on liver morphology have been investigated in mice. Pentamidine served as reference drug. Both agents caused dose-dependent increases in serum transaminases and SDH, and discrete morphological changes of the liver, e.g., fatty degenerations, azinoperipheral vacuolisation and alterations of the nuclei, at least at the higher dosage.
