中药提取物逆转K562/ADM细胞的耐药作用及其分子机制的探讨

Objective To investigate the reversal effect of the monomer of traditional Chinese medicine on muhidrug resistance(MDR) and its possible mechanism in K562/ADM cell line in vitro. Methods With different concentrations of baicalin, geniposide administered to K562/ADM cells, the proliferation of K562/ ADM cells was detected by the MTY assay. Expression of mdr-1 mRNA, Topo Ⅱ mRNA was measured by semi-quantitive RT-PCR. Results Thatbaicalin and geniposide could increase the sensitivity of K562/ADM cells to adriamycin, multiples of reversion were 1.95 times and 1.46 times. The proliferation of K562/ADM cells was in-hibited obviously by baicalin and geniposide, the level of mdr-1 mRNA expression was down-regulated and the Topo Ⅱ mRNA was up-regulated(P<0.01 ). Conclusion Baicalin and geniposide may reverse the multi-drag-resistance of K562/ADM cells, which was related to the down-regulation of mdr-1 expression and up-reg-ulation of Topo Ⅱ beta expression.     

1998年延安地区细菌耐药性监测

报道 1998年延安地区 12 30株临床分离菌对常用抗菌药物的耐药性。采用药敏纸片扩散法 ,检出革兰阳性菌 353株 ( 2 8.7% ) ,革兰阴性菌 734株( 61.3% ) ,以大肠杆菌、凝固酶阴性葡萄球菌为多见。革兰阳性菌对青霉素的耐药率达 98% ,对头孢菌素类药耐药率为 10 %～ 2 0 %。革兰阴性菌对氨苄青霉素的耐药率为 80 % ,对头孢菌素类药耐药率为 30 % ,对喹诺酮类药物耐药率约为 10 %～ 2 0 %。

无痛性骨髓穿刺术的临床应用

目的：探讨无痛性骨髓穿刺术的临床应用。方法：分析应用丙泊酚静脉麻醉后行骨髓穿刺术103例（无痛组）的完成情况、不良反应、患者的满意度与同期常规骨髓检查术103例（对照组）比较,判断两种检查方法的差异。结果：无痛组患者满意度显著高于对照组（P〈0.01）。结论：无痛性骨髓穿刺术是一种安全、舒适的检查手段,优于常规骨髓穿刺术,值得临床推广。     

麻辛平喘口服液对组胺致痉豚鼠气管条舒张作用实验研究

目的观察麻辛平喘口服液对抗由组胺引起豚鼠离体气管条的收缩作用。方法制备豚鼠离体气管螺旋条,用组胺引起气管螺旋条收缩,观察麻辛平喘口服液对组胺诱发的离体气管条的收缩反应的舒张作用。结果麻辛平喘口服液能使组胺致痉的豚鼠气管平滑肌松弛,解痉百分率为(121.16±31.97)%。结论麻辛平喘口服液对组胺致痉豚鼠气管条具有舒张作用。     

犬MC2R基因5'-UTR、3'-UTR序列测定、分析及其 5'侧翼启动区序列分析

目的 分离犬MC2R基因CDS部分序列和该基因5'、3'非翻译区,并对5'、3'非翻译区和5'侧翼的启动区进行分析.方法 本研究采用反转录PCR(RT-PCR)和RNA连接酶介导的RACE (RLM-RACE)技术和BLAST分析软件.结果 得到了5'、3'非翻译区和部分CDS片段的序列,他们的大小分别为168 bp、1 366 bp和987 bp,并预测了大约1 500 bp的5'侧翼的启动区域.结论 对它们进行分析显示,该基因至少由两个外显子(exon1和 exon2)组成,exon1 和exon2的一部分编码5'非翻译区(5'-UTR),exon2其余的部分编码整个编码区.其启动区有inr, SF-1, SP1, CRE, PPRE, AP-1等多个顺式作用元件,这些为犬MC2R表达调控研究提供研究奠定基础.
