Role of microfilaments in the exocytosis of rat peritoneal mast cells   总被引:1,自引:0,他引:1  
When rat peritoneal mast cells were treated with the potent histamine releaser compound 48/80 in the presence of tetramethylrhodamine-labeled G-actin, the fluorescent G-actin particles were bound to the surface of extruded granules and to the cell surface. When rhodamine-phalloidin was incorporated into permeabilized rat mast cells in a Ca2+-free medium, rhodamine fluorescence was observed on the cell surface accompanied by serpentine ridges which appeared in the resting state. After perfusion with a cytosol-like solution containing Ca2+, rhodamine fluorescence appeared on the cell surface as a distinct network formation. In some cases, circular fluorescences which appeared to surround the extruded pores were observed in the cell membrane. These findings indicate the existence of actin filaments in the cell membrane and/or subplasmalemmal network. In whole-mount preparations, the granules were surrounded very densely with microfilaments of various widths. After exposure to compound 48/80, granules protruding through the cell membrane were wrapped in many filaments. The extruded granules located in the periphery of the cells were connected by many filamentous structures and disruptions in the middle of these connections were occasionally observed. In some cases, circular configurations of microfilaments were observed at the bottom of the extruded granules and in others dense gatherings of microfilaments were seen just beneath the granules, as if the latter were being pushed up and out of the cells.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The cross-reactivity of antibodies to adult T-cell leukemia (ATL)-associated antigens (ATLA) in human and monkey sera was investigated by indirect immunoperoxidase and immunoferritin methods using a human cell line (MT-2) carrying a type C virus (HTLV), two monkey cell lines (Si-1 and Si-3) carrying HTLV, and a monkey cell line (Si-2) carrying a type C virus isolated from an anti-ATLA-positive monkey. Anti-ATLA-positive but not-negative human and monkey sera gave positive immunoperoxidase reaction with all four virus-positive cell lines when studied by light microscopy. Electron microscopic findings revealed ferritin or peroxidase labeling of virus particles and plasma membranes of these four cell lines with antibody-positive but not-negative human and monkey sera. These results clearly indicate the cross-reactivity of anti-ATLA antibodies in human and monkey sera at light and electron microscopic levels, and the presence of antigenic determinants common to the surface of type C virus particles of human and monkey origin.  相似文献

The postnatal development of the three-dimensional structure and arrangement of the tendon cells in the calcaneal tendon of rats from 5 to 90 days of age were examined under the scanning and the transmission electron microscope (SEM and TEM). Exposed tendon cells were seen as winged bricks with a stellate profile in a cross section. Their slender perikarya were stacked successively in rows along the long axis of the tendon. The plate-like cytoplasmic processes, which were oriented along the perikaryal long axis, extended radially and joined those of the neighboring cells in rows. Therefore, the tubular channels for collagen fascicles were lined with the chained perikarya and their processes of the tendon cells. The cell rows were usually composed of many cells in developing stages, while short rows of one or two cells were observed in the fully developed stage. The cytoplasmic processes were the primary processes in the lateral extension, and their fine branches formed the secondary processes. The secondary processes were numerous at younger stages, showing a fine meshwork due to their mutual joining in the cross sections. In advanced stages, the meshes were coarse and the secondary processes were also perforated either with grouped fenestrations or large pores. In the fully developed stage, the secondary processes were fragmented on the perikarya, while the primary processes extended in the thin delicate sheets with large perforations. The findings in the present study suggest that the tendon cells are arranged in a three-dimensional network by their mutual joining. The tendon cells of the rat calcaneal tendon may not proliferate very much after birth, but do expand their nursing area in line with normal growth by an elongation of the main primary processes and a reduction of the secondary processes and perikaryal mass. The interfascicular clefts, which were caused by an intervention of either the processes at any developmental stages or the fragmented processes at a certain level of the tendon, may also play a role in the passage of tissue fluid.  相似文献

We investigated the relationship between the antinociceptive effect of the opiate agonist loperamide at the spinal level and its inhibitory effect on calcium influx. Intrathecal administration of loperamide showed a significant antinociceptive effect in the formalin test, which was not prevented by naloxone. On the other hand, no significant effects were observed by nicardipine, an L-type specific blocker, or by BAY K8644, an L-type specific agonist, suggesting no significant role of L-type calcium channels in nociceptive signal transduction. Loperamide suppressed the calcium influx in dorsal root ganglion neurons. As the antinociceptive effect of loperamide was not affected by naloxone or other calcium channel blocking toxins, and loperamide showed a direct inhibitory effect on calcium-influx, the analgesic effect of intrathecally injected loperamide might be due to its blockade of the voltage-dependent calcium channels at the terminals of the primary afferent fibers.  相似文献

Lymphoid cell lines were established from five different species of monkeys which were positive in antibodies cross-reactive with human T-cell leukemia virus type I (HTLV-I) and were shown to contain provirus sequences homologous to HTLV-I. Gene-specific probes of HTLV-I, gag, pol, env, pX, and LTR, hybridized efficiently with monkey DNAs from these cell lines under stringent conditions, indicating that the proviruses are very similar to HTLV-I along with whole viral genomes. However, the preliminary restriction mapping turned out the difference between simian retroviruses and HTLV-I and also among simian retroviruses. These findings suggest a common ancestor of simian and human retroviruses, but exclude the recent interspecies transmission between monkeys and humans.  相似文献

The authors approach the societal problem of the just distribution of health care resources by exploring one example-- the allocation of intensive care unit (ICU) beds between present patients and those awaiting admission--within the context of not expending a disproportionate share of resources for marginal benefit. They propose the metaphor of the societal "insurance policy" under which decisions regarding the investment of resources to meet various possible contingencies are made in advance. Summary Vincristine-induced crystalloid inclusions were examined in the neurons and neuronal processes of young rats by the electron microscope (EM) equipped with a tilting stage. Using a computer system that reproduces three-dimensional organization, an optical transformation method was applied to the microtubules and neurofilaments in an attempt to clarify the morphological appearance and internal pattern of crystalloid inclusions. The dimensional models obtained were compared with actual EM photographs, and the characteristic ultrastructural component and morphology were drawn out.Basically, a crystalloid inclusion is composed of four strands of intermediate 10 nm neurofilaments connected to one another by four side-arms producing a circular profile on a transverse section. These four side-arms seemed to arise from nodules within the filaments at regular intervals simulating a bead-like appearance. These data did not significantly differ from those obtained from EM images. Characteristically, these crystalloid inclusions began to appear 6 h after the administration of 10–3 M vincristine sulfate and persisted up to a period of 6 days. Beyond that, however, these inclusions were no longer demonstrable suggesting a transient state. This was in contrast to neurofibrillary tangles which appeared to be permanent changes.  相似文献
