Toxicity of perfluorinated fatty acids for human and murine B cell lines

The toxicity of two perfluorinated fatty acids, penta decafluoro-n-octanoic acid (PFOA) and nonadecafluoro-n-decanoic acid (NDFDA), on three mammalian B cell lines was evaluated. Cells were exposed to the perfluorinated molecules for either 24 or 48 hr under a variety of culture conditions. Immunoglobulin secretion and surface membrane expression were unaffected by both PFOA and NDFDA at sublethal concentrations. Lethal effects of PFOA and NDFDA are diminished by either lowering culture temperature (37 to 20 degrees C) or including fetal bovine serum or human serum albumin in media. At lethal concentrations, both PFOA and NDFDA possess detergent activity since they can release IgM in soluble form from a cell line that does not secrete immunoglobulins, and brief exposure (15 min) to 1-2 mM of both perfluorinated fatty acids results in solubilization of F4 cells equivalent to the anionic detergent deoxycholic acid. Our data suggest that at subtoxic concentrations, neither PFOA nor NDFDA alters expression or secretion of a differentiated gene product (IgM). At lethal levels, both chemicals cause increased solubilization of proteins from lymphoblastoid cell lines.

Die wässerigen Herzextrakte in niedrigeren Konzentrationen (11000 bis 1100000 Verdünnungen) hemmen—analog zu dem Ringereiweiß — die erholende Wirkung des Na-Oleinicum auf das hypodyname Froschherz. Nach faradischer Reizung des vagosympathischen Stammes erhöht sich der Eiweißgehalt der sich im Straubherzen befindenden Ringerschen Lösung.Mit 1 Textabbildung.     

Neoplastic obstruction of the trachea complicating pseudobulbar palsy

We have reported a case of pseudobulbar palsy with airflow obstruction initially attributed to chronic aspiration and chronic airflow obstruction caused by cigarette smoking. The finding of stridor led to a detailed evaluation of the obstruction. Flow-volume loops, fiberoptic bronchoscopy, CT scan of the upper thorax, and fine-needle biopsy of a lesion of the trachea were required to make the diagnosis of non-small-cell carcinoma involving the trachea.

Über Versuche mit radioaktiv (J131) markierten Antikörpern gegen Nucleinsäuren und Nucleoproteide aus hepatocellulärem Rattencarcinom und normaler Rattenleber

Zusammenfassung Durch wiederholte Injektion von Desoxyribonucleinsäure und Desoxyribonucleoproteiden aus normaler Rattenleber und einem transplantablen hepatocellulären Carcinom der Ratte wurden Kaninchenantiseren gewonnen. Aus diesen Seren wurden die -Globuline elektrophoretisch isoliert und nach Markierung mit J¹³¹ bei tumortragenden Ratten intravenös injiziert.Die Speicherung im Tumor lag in allen Fällen in etwa gleicher Höhe. Antikörper gegen DNP/Leber lagerten sich dagegen in der Leber in 6mal höherer Konzentration ab als normale -Globuline.Daraus ist zu schließen, daß in den dargelegten Versuchen die DNP aus Krebsgewebe gegenüber dem normalen Ausgangsgewebe Antigendefekte aufwiesen, die den Eiweißanteil betreffen. Darüber hinaus fehlten den DNP des Krebsgewebes Antigenkomponenten, die in Normalgeweben anderer Organe, z. B. der Niere, vorhanden waren.Im Gegensatz zu Antikörpern gegen DNP aus Tumor und Normal-gewebe verhielten sich -Globuline gegen DNS aus den beiden Geweben in ihrer Organverteilung gleich.Unsere bisherigen Resultate sprechen gegen die Vorstellung eines isolierten Angriffspunktes der malignen Entartung an den Cytoplasmagranula und gegen die Spezitität von Krebsantiseren, die mit Hilfe von Tumordesoxyribonucleoproteiden gewonnen wurden.Es wird eine Methode beschrieben, um insbesondere Leber und Niere weitgehend von Blut zu befreien.

Summary Rabbit sera were produced by repeated injections of desoxyribonucleic acid (DNA) and desoxyribonucleoprotein (DNP) obtained from normal rat liver and from a transplantable hepatocellular rat carcinoma. Gamma globulins were isolated electrophoretically from these sera, were labelled with I¹³¹, and injected intravenously into tumor bearing rats.The radioactivity in hepatoma was similar in all cases. But antibodies against DNP/liver were concentrated in the liver 6 times more than normal -globulins.From these experiments it may be concluded that DNP from carcinomatous tissue is antigenically deficient when compared to normal tissues. The deficiencies probably are only of the protein. Moreover, the carcinoma DNP lacked certain antigenic components that were present in normal tissues and organs, e. g. in the kidney.In contrast to antibodies against DNP from tumor and from normal tissue, -globulins against DNA from the two tissues behaved equally.The findings to date do not support the concept that malignant transformation affects primarily or solely the cytoplasmic granula. The results furthermore discount the specificity of tumor antisera produced against tumor-DNP.A method for clearing blood from the liver and kidney is described.
