The microinjection of hypertonic sodium chloride into the third ventricle elicits natriuresis and kaliuresis in rats following water loading. These responses were blocked by atropine or phentolamine microinjection into the MSA. The data suggest the interaction between periventricular areas sensible to changes of sodium concentration in the cerebrospinal fluid (CSF) and MSA. Cholinergic and alpha-adrenergic synapses of MSA are involved in this interaction which regulates renal sodium excretion.  相似文献

The aim of this work was to study the role of the dorsal noradrenergic ascending pathway (DNAP), which originates in the locus coeruleus (LC) on the preovulatory surge of luteinizing hormone (LH) follicle-stimulating hormone (FSH) and prolactin (PRL) by producing bilateral electrolytic lesions (cathodal or anodal) in this nucleus. LC lesions were placed at 11.00 h on proestrus in female rats with regular 4-day estrous cycles. Intact rats, sham-operated as well as animals with missed lesions served as controls. In Experiment I, anodal current was applied and hourly blood samples were withdrawn (from 13.00 to 17.00 h) via a jugular catheter from conscious, freely moving rats for determination of plasma LH, FSH and PRL concentrations. In Expt. II, Expt. I was repeated using cathodal current and collecting blood samples hourly from 13.00 to 18.00 h. In both experiments the animals were sacrificed on the next morning when the occurrence of ovulation was checked. The medial septal area (MSA), medial preoptic area (MPOA), and medial basal hypothalamus (MBH) were dissected and assayed for norepinephrine (NE), dopamine (DA) and 5-hydroxyindoleacetic acid (5-HIAA) content. Experiment III was performed in order to test if a hormonal discharge occurred immediately after lesion placement. Blood samples were collected immediately before and 15, 30, 60 and 90 min postoperatively (from 11.00 to 12.30 h). Either anodal or cathodal lesions blocked the proestrous surge of LH, FSH and PRL. The hypothesis that the lesions advanced or delayed these hormonal surges was rejected since we found no increases in the hormonal levels from 11.00 to 12.30 or from 13.00 to 18.00 h, and ovulation was not observed on the following morning in the lesioned animals. Since control, sham-operated and missed-lesion groups exhibited LH, FSH and PRL surges and ovulation, this blockage appears to be caused by the destruction of the LC neurons. Also, this blockade was correlated with a decrease in the NA content in the MPOA and MBH, but not in the MSA, whereas the DA and 5-HIAA content were not changed in all groups examined. The results lead us to suggest that the integrity of noradrenergic afferent input from the LC to luteinizing hormone-releasing hormone neurons in the MPOA and MBH is essential for triggering the preovulatory surge mechanisms for gonadotrophins and PRL.  相似文献

The objective of the study was a quantitative examination of Proprioceptive Neuromuscular Facilitation (PNF) exercise in simultaneous combination with FES of lower extremity muscles in comparison to voluntary movement, training with PNF alone, or training with FES alone. Two subjects were monitored during a one‐month rehabilitation period. The PNF pattern included flexion, adduction, and external rotation of the hip, knee flexion, and dorsiflexion with inversion of the ankle, a pattern similar to the swing phase of walking. Quantitative measurements were conducted by using goniometers on the hip, knee, and ankle joints. Major changes were found in the hip angle. Improvements in goniograms were greatest during the first week, smaller during the second week, and showed only a slight positive trend in the last two weeks. The measurements made two months after the start of training showed somewhat lower values in comparison to previous sessions.  相似文献

Fatigue of electrically activated paralysed muscles is a major factor limiting the duration of functional electrical stimulation (FES) supported paraplegic standing. Fatigue can be significantly delayed by changing the posture. Since paralysed individuals are deprived of proprio- and exteroception from the lower limbs they are not aware of the posture and loading of their paralysed legs. If suitable cognitive feedback (CF) information about posture in the sagittal and frontal planes is provided, they might be able to successfully exercise posture switching. A two-dimensional electrocutaneous CF system was developed. Relative limb loading and the location of the weighted centre of pressure were selected as informational variables. Discrete encoding schemes in the form of spatial and frequency codes were employed and the informational signals were divided into three sub-regions. The ability to correctly interpret the CF was investigated using one- and two-dimension tracking tests in three paralysed subjects, each of whom were studied over five consecutive days. All three subjects were able to use the CF in one-dimension tests. Two subjects could do the same also in two-dimension tests. The encoding scheme which was developed to communicate the selected biomechanical variables proved to be easily understood and thus appropriate for use in paraplegic standing.  相似文献

The anteroventral region of the third ventricle (AV3V) is critical in mediating osmotic sensitivity. AV3V lesions increase plasma osmolality and block osmotic-induced vasopressin (VP) and oxytocin (OT) secretion. The aim was to evaluate the effects of AV3V lesions on neurosecretion under control/water replete conditions and after 48 h dehydration. The focus was on central peptidergic changes with measurement of OT and VP content in the hypothalamic paraventricular (PVN) and supraoptic (OT) regions and the posterior pituitary. AV3V-lesioned rats exhibited an elevated plasma osmolality and higher OT content in SON and PVN. There was an increase in VP content in PVN, but no change in SON. As predicted, the plasma peptide response to dehydration was absent in lesioned animals. However, dehydration produced depletion in posterior pituitary VP in lesioned animals with no change in OT. No changes in nuclear VP and OT levels were seen after dehydration. These results demonstrate that AV3V lesions alter the VP and OT neurosecretory system, seen as a blockade of osmotic-induced release and an increase in basal nuclear peptide content. The data indicate that interruption of the osmotic sensory system affects the central neurosecretory axis, resulting in a backup in content and likely changes in synthesis and processing.  相似文献

The aim of this study was to describe and validate a method to evaluate the preovulatory surges of gonadotropins in rats submitted to anesthesia and implantation of a jugular vein cannula in the morning of proestrus and to withdrawal of serial blood samples in the afternoon of the same day. In experiment I, to choose an adequate anesthetic, cycling female rats were anesthetized in the morning of proestrus (10:00-11:00 h) with tribromoethanol, ketamine/xylazine or tiletamine/zolazepam and a Silastic cannula was implanted into the jugular vein. Blood samples (0.6 ml) were withdrawn hourly between 12:00 and 18:00 h of the same day and, on estrus morning, the rats were decapitated and the number of ova was counted. The preovulatory gonadotropin surges as well as ovulation occurred in rats anesthetized with tribromoethanol, while they were prevented by ketamine/xylazine or tiletamine/zolazepam. To investigate if the jugular cannulation under tribromoethanol anesthesia and serial blood sampling performed in experiment I altered the magnitude of the gonadotropin surges and the number of ova, intact rats (control) or rats anesthetized with tribromoethanol followed or not by jugular vein cannulation were decapitated at 16:00 h of proestrus and in the morning of estrus. The magnitude of preovulatory gonadotropin surges and the number of ova were not different among groups. Thus, since neither tribromoethanol nor surgical procedures or serial blood sampling altered the preovulatory gonadotropin surges or the ovulation process, this method seems to be suitable for this sort of study in rats.  相似文献

Two groups of rats with electrolytic lesions of the medial and upper septal area (MUL) or, alternatively, of the anteroventral portion of the third ventricle (AV3V) and a third group of sham-operated rats were water loaded and received three carbachol injections into the locus coeruleus according to the following schedule: 1) prelesion, 2) on the second postlesion day and 3) on the seventh postlesion day. Both MUL and AV3V lesions inhibited the carbachol-induced natriuresis on the second postlesion day. Recovery was almost complete after MUL but not after AV3V lesion on the seventh day. Water deprivation also reduced the carbachol-induced natriuresis but passive hydration of AV3V animals did not avoid the impairment induced by the lesion. Transient seizure phenomena such as clonic convulsions, salivation and analgesia subsequent to carbachol injection were not altered by the lesions.  相似文献

The effects of perinatal picrotoxin (0.75 mg/kg) on heterosexual and homosexual behavior of male rats, sexually experienced or not, were studied. The following data were obtained: (1) at birth, body weight and anogenital distance were not modified by the treatment; (2) during lactation, both treatment and sex interfered with body weight as well as in adult age; (3) as experimental animals were trained, the heterosexual behavior was improved; (4) picrotoxin treatment reduced lordotic response of homosexual behavior in inexperienced male rats and (5) the heterosexual experience with female rats inhibited homosexual behavior of both experimental and control animals. These results suggest that perinatal maternal picrotoxin exposure improved heterosexual behavior in male rats and the sexual experience reveals this effect. In addition, picrotoxin did not induce feminization in experimental inexperienced rats. Finally, the sexual experience per se promotes changes in brain regions related to male behavioral and sexual aspects.  相似文献

BACKGROUND: Leptin has been proposed to be involved in central control of adiposity and fat distribution but the role of this peptide is controversial. The aim of our study was to test the relationship between serum leptin and body composition, fat distribution, and some biochemical markers such as fasting insulinemia and lipoproteins in a population of healthy Italian postmenopausal women. METHODS: One hundred and twenty-three postmenopausal women (62.1+/-8.7 years) were evaluated. Body composition (fat and lean mass) was assessed by dual-energy X-ray absorptiometry (DXA). Two regions of interest were determined for regional fat analysis. Serum leptin and insulinemia were measured by radioimmunoassay, lipoproteins with colorimetric methods and apolipoproteins nephelometrically. RESULTS: Plasma leptin levels are strongly related to total fat mass, in grams (r=0.73, p<0.001) or as a percentage of soft tissue (r=0.75, p<0.001), and to adiposity, calculated as ratio between lean and fat mass (r=0.76, p<0.001). A significant correlation was also found between serum leptin and central fat distribution (r=0.29, p<0.01). As concerns biochemical markers, serum leptin was significantly related to fasting insulin (r=0.38, p<0.001), total cholesterol (r=0.29, p<0.01), Apolipoprotein-B (r=0.35, p<0.001), and triglycerides (r=0.22, p<0.05). When corrected for total fat mass, the partial correlation coefficients remain significant for percentage of total body fat (r=0.27, p<0.01), adiposity (r=0.23, p<0.01), and fat proportion in android region (r=0.18, p<0.05). CONCLUSIONS: These data indicate that leptin levels are related to adiposity and fasting insulin levels; indeed fast insulin mantains significant correlation with leptin (r=0.23, p<0.01) after controlling for fat mass. Android distribution of fat mass in elderly women is associated with leptin concentration.  相似文献
