Targeted disruption of the murine tissue factor gene results in embryonic lethality 总被引:21,自引:9,他引:21
Tissue factor (TF) is an integral membrane glycoprotein that is believed to be the physiologic initiator of the blood coagulation cascade. Disruption of the mouse tissue factor gene leads to embryonic lethality between days E9.5-E11.5 of gestation. On E9.5, TF(-/-) embryos appear indistinguishable from their TF(+/+) and TF(+/-) littermates. By E10.5, TF(-/-) embryos are severely growth retarded, appear nearly bloodless, and are in most cases dead. Initial observations suggest that TF(-/-) embryos are dying of circulatory failure. Approximately 15% of the TF(-/-) embryos survive beyond E10.5, but none complete gestation. Heterozygotes appear normal and free of bleeding complications. 相似文献

Experimental animals that had been given excess iron in their diet were studied by magnetic resonance (MR) imaging in vivo and by magnetic resonance (MR) spectroscopy in vitro. Hepatic iron overload in patients with transfusional iron excess was studied by MR imaging, and isolated iron protein fractions were studied in vitro by MR spectroscopy. The spin echo image intensity of livers with iron overload was decreased because of the extreme decreases in T2 compared with normal; T1 was decreased only moderately. The relaxation rates 1/T2 and 1/T1 both showed a linear relationship to hepatic iron levels. Ferritin solutions showed moderate decreases in T2 and mild decreases in T1. The T2 relaxivity of ferritin, which is due to the iron core rather than the apoferritin protein shell, does not appear sufficient to account for the extreme decrease in T2 observed in hepatic iron overload. Low molecular weight cytosol iron is present in lower concentrations than ferritin but potentially has much greater relaxivity and may contribute to the MR findings. Background
Mathematical modelling and analysis is now accepted in the engineering design on par with experimental approaches. Computer simulations enable one to perform several 'what-if' analyses cost effectively. High speed computers and low cost of memory has helped in simulating large-scale models in a relatively shorter time frame. The possibility of extending numerical modelling in the area of breast cancer detection in conjunction with medical thermography is considered in this work.Methods
Thermography enables one to see the temperature pattern and look for abnormality. In a thermogram there is no radiation risk as it only captures the infrared radiation from the skin and is totally painless. But, a thermogram is only a test of physiology, whereas a mammogram is a test of anatomy. It is hoped that a thermogram along with numerical modelling will serve as an adjunct tool. Presently mammogram is the 'gold-standard' in breast cancer detection. But the interpretation of a mammogram is largely dependent on the radiologist. Therefore, a thermogram that looks into the physiological changes in combination with numerical simulation performing 'what-if' analysis could act as an adjunct tool to mammography.Results
The proposed framework suggested that it could reduce the occurrence of false-negative/positive cases.Conclusion
A numerical bioheat model of a female breast is developed and simulated. The results are compared with experimental results. The possibility of this method as an early detection tool is discussed.97.

Throughout the western United States, studies have identified various detrimental effects of contaminants to aquatic biota
from the use of agricultural drainage water for management of arid wetlands. However, little is known about the relative contributions
of contaminant loading from pollutants dissolved in water compared with those carried by drifting material (e.g., detritus) associated with drainage water. Consequently, we determined loading rates for contaminants dissolved in water and
those incorporated by drifting material for drainage (Diagonal Drain) as well as fresh (S-Line Canal) water used for wetland
management at Stillwater National Wildlife Refuge (SNWR), Nevada during the early, middle, and late periods of the irrigation
season (June through mid-November) in 1993. We found loading rates for trace elements throughout the irrigation season were
almost entirely (> 98%) associated with contaminants dissolved in the water rather than incorporated by drift. Although drift
contributed little to the total loading for trace elements to SNWR wetlands, contaminant concentrations were much greater
in drift compared with those dissolved in water. Loading rates for dissolved As, B, Hg, and total dissolved solids (TDS) differed
among periods for the Diagonal Drain. Along the Diagonal Drain, loading rates for dissolved As, B, Hg, Mo, unionized ammonia
(NH3-N), TDS, and Zn differed among its three sampling sites. B was the only trace element with differences in loading rates for
drift among periods from the Diagonal Drain. In contrast, loading rates for As, B, Cr, Cu, Hg, Se, and Zn in drift differed
among periods for the S-Line Canal. Along Diagonal Drain, loading rates in drift for B (middle and late periods), Cr, Cu,

The city of Bahía Blanca occupies a strategic place in Argentina south of the Pampean region in the north-east corner of the Patagonia. Since 1828, this city has been the historical and political border between Amerindian lands in the south, and the lands of European colonists. Nowadays, Bahía Blanca is an urban population mainly composed by descendents of immigrants from Spain and other European countries with apparently low admixture with Amerindians. In view of the unexpectedly high Amerindian admixture levels (about 46.7%) suggested by mtDNA data, and protein markers (19.5%), we analyzed a set of 19 Alu polymorphisms (18 autosomal, 1 of Chromosome Y) in a well-documented genealogical sample from Bahía Blanca. The genotyped sample was made up of 119 unrelated healthy individuals whose birth place and grandparent origins were fully documented. According to available genealogical records, the total sample has been subdivided into two groups: Bahía Blanca Original (64 individuals with all 4 gandparents born in Argentina) and Bahía Blanca Mix (55 individuals with one to three grandparents born out of Argentina). Allele frequencies and gene diversity values in Bahía Blanca fit well into the European ranges. Population relationships have been tested for 8 Alu markers, whose variation has been described in several Amerindian and European samples. Reynolds genetic distances underline the significant genetic similarity of Bahía Blanca to Europeans (mean distance 0.044) and their differentiation from Amerindians (0.146). Interestingly enough, when the general sample is divided, Bahía Blanca Original appears slightly closer to Amerindians (0.127) in contrast to Bahía Blanca Mix (0.161). Furthermore, the genetic relationships depicted through a principal components analysis emphasize the relative similarity of Bahía Blanca Original to Amerindians. Rats show signs of dependence on sugar when it is available intermittently, including bingeing, withdrawal, and cross-sensitization with amphetamine. In the current study, we sought to determine whether sugar-dependent rats would show increased intake of unsweetened ethanol and, conversely, whether intermittent access to ethanol would augment sugar consumption. In Experiment 1, with intermittent versus ad libitum access to ethanol, Sprague-Dawley rats were given escalating concentrations of ethanol (1%, 2%, 4%, 7%, and 9%) over the course of 20 days. Rats in the intermittent ethanol access group, with 12-h daily access, consumed more 4%, 7%, and 9% ethanol during the first hour of access, and more 9% ethanol daily, than did rats in the ad libitum ethanol access group. In Experiment 2, with ethanol as a gateway to sugar intake, the rats from Experiment 1 were switched to 10% sucrose with 12-h daily access for 1 week. Rats in the intermittent ethanol access group consumed significantly more sugar than was consumed by rats in a control group with no prior ethanol experience. In Experiment 3, with sugar as a gateway to ethanol to determine whether sugar dependence leads to increased ethanol intake, four groups were maintained for 21 days according to the following designations: intermittent access to sugar and chow, ad libitum access to sugar and chow, intermittent access to chow, or ad libitum access to chow. Four days later, all groups were switched to intermittent ethanol access, as described in Experiment 1. The group with intermittent access to sugar and chow consumed the most 9% ethanol, supporting the suggestion that sugar dependence alters a rat's proclivity to drink ethanol. These results may relate to the co-morbidity between binge-eating disorders and alcohol intake and the tendency of people abstaining from alcohol to consume excessive amounts of sugar. This work examines the effect of drug structure and ionization degree on the formation and properties of biocompatible layered double hydroxides (LDH) intercalated with ibuprofen and paracetamol. Ibuprofen (pKa = 5.3) is in its anionic form, whereas paracetamol (pKa 9.4) is only partially ionized at the synthesis pH (9.0), and thus intercalation is expected to be different in the two cases. Chemical analyses, X-ray diffraction, electron microscopy, infrared spectroscopy and thermal analyses were applied to characterize the materials. Dissolution kinetics and drug release kinetics were also investigated, in an ample range of pH (3.0–9.0) in NaCl solutions, and in physiological buffers (1.2, 4.5 and 6.8). All characterization techniques showed that an efficient intercalation of ibuprofen took place, resulting in a material with 30% of its weight corresponding to the drug. On the contrary, all techniques revealed a very poor intercalation of paracetamol (1.2%). The dissolution kinetics of LDHs was highly pH-dependent, being higher as pH decreased. The drug release kinetics, conversely, increased as pH increased. In physiological buffers the release rate depended not only on the pH but also on the type of buffer. This last behavior is useful to control the release in different parts of the digestive system. 相似文献
