The pattern of amine oxidation was studied in human blood vessels at various stages of development, and in tissues with muscular layers made up predominantly or exclusively of smooth muscle. Specific activity of benzylamine oxidase, present in all vascular tissues examined, was higher in vessels than in other tissue; the enzyme, in organs rich in nonvascular smooth muscle, though lower than in blood vessels, was significantly higher than in striated muscle or connective tissue. The localization of benzylamine oxidase activity in smooth muscle, as opposed to connective tissue, may have important physiological implicacations.  相似文献

We employed a morphological assay of outer segment collapse to determine if growth factors or other supplements directly affect dissociated rod photoreceptors in vitro. The morphological changes in outer segments were correlated with the light responsiveness of rods. Time-lapse video microscopy was used to observe the collapse of rod outer segments from isolated single cells and small clumps of cells. A consistent pattern of outer segment collapse into the inner segment was observed, yielding a convenient assay of the effects of neurotrophic factors on photoreceptor functional maintenance. The functional state of rods, defined as light-responsiveness, was measured with suction electrode recordings and matched with the various stages of outer segment collapse. Ciliary neurotrophic factor (CNTF) and glial cell-line-derived neurotrophic factor (GDNF) at a high concentration, yielded statistically significant improvements in rat outer segment survival times. Basic fibroblast growth factor (bFGF), which rescues photoreceptors in several rodent models of retinal degeneration, produced a significant increase in survival time in the presence of the cofactor heparin. In 4 out of 10 cases using human tisue, bFGF also yielded a significant increase in survival times. When brain-derived neurotrophic factor (BDNF) was applied to rat rods, outer segment survival times did not change. Outer segments collapsed more quickly when either pigment epithelial cell derived factor (PEDF) or sugarN-acetyld-galactosamine (NAD-gal) were present. Our results show that rod photoreceptors can respond to bFGF, GDNF and CNTF in vitro and provide evidence for a direct effect of these neurotrophic factors on rods. The rapid collapse of isolated photoreceptors in this model provides a convenient means for testing various neurotrophic agents and the induced cellular responses.  相似文献

The immunological basis of resistance to Brugia pahangi infection in rats was studied. Infections were investigated in athymic rnu/rnu rats and in rats treated with the immuno-suppressive agents cyclosporin A (CsA) or cyclophosphamide (Cy). The recovery of adult worms in normal rats was 1-2% in comparison to 12.2% recovery in athymic rats. CsA and Cy treated rats did not have increased adult worm burdens. Microfilarial (Mf) levels (expressed as Mf per ml per adult worm) were highly elevated in both athymic and Cy treated rats but not in CsA treated rats. IgG and IgM levels to B. pahangi antigens were severely depressed in both athymic and Cy treated rats. IgG levels but not IgM levels were abrogated in CsA treated rats. These results implied that control of larval establishment or adult killing, and regulation of Mf levels are separate T-cell dependent mechanisms and act independently of IgG antibody. Control of Mf levels is associated with a specific IgM response which is Cy sensitive but CsA resistant.  相似文献

Two cases of elastosis perforans serpiginosa were studied by direct immunofluorescence. In one case, homogeneous deposits of IgM, C3 and C4 on the abnormal elastic fibres in the papillary dermis were demonstrated. In addition, in both cases, along the basement membrane zone, coarse linear deposits of fibrinogen and properdin were observed inside and near the transepidermal channels. Groups of cytoid bodies stained for IgM and occasional weak linear deposits of IgG, IgA, IgM and C3 were also observed. The immune deposits on the abnormal elastic fibres might indicate antigenic alteration and activation of humoral immunological mechanisms in the process of their elimination. The other findings may reflect a non-specific binding by damaged tissue structures.  相似文献
