We have investigated the effects of the angiotensin II type 1 receptor antagonist candesartan, and the angiotensin II converting enzyme inhibitor ramipril, on catecholamine release from the anaesthetized dog's adrenal gland. These drugs were given systemically in low and high doses. The gland was stimulated electrically (0.5-12 Hz) and by angiotensin II infusion (40 ng/kg/min). Electrical stimulation resulted in frequency-dependent increases in catecholamine release. Candesartan (0.8, 4.0 mg/kg) and ramipril (0.125, 0.625 mg/kg) increased basal catecholamine release along with decreases in blood pressure. Both drugs diminished direct nerve stimulation-induced catecholamine release. When both drugs were combined, their inhibitory effect was slightly enhanced. Candesartan blocked catecholamine release induced by angiotensin II. Ramipril was not tested in this respect. The percentage of noradrenaline released during electrical stimulation of the gland remained constant and ranged from 14% to 22%. Both drugs appear to act by blocking local modulation of catecholamine release by the chromaffin cells.  相似文献

Glucose-6-phosphate dehydrogenase (G6PD)-deficient subjects are susceptible to chemical-induced oxidative haemolysis. Little is known concerning the haemolytic properties of Chinese herbal medicine on G6PD-deficient subjects. Our objective was to investigate the pro-oxidative effect of 18 commonly used Chinese herbal medicine (CHM) on human G6PD-deficient red blood cells. G6PD-deficient (n=10) and normal (n=10) whole blood samples were incubated with water extracts of CHM. The resulting levels of reduced glutathione (GSH) and methaemoglobin (MetHb) were determined by biochemical assays. Rhizoma Coptidis significantly reduced GSH level by 48.9+/-5.4% (at 1 mg/mL) in the G6PD-deficient erythrocytes (P<0.001) compared with the respective control group without challenge. Similar dose-dependent responses were observed at higher concentrations of Cortex Moutan, Radix Rehmanniae, Radix Bupleuri, Rhizoma Polygoni Cuspidati and Flos Chimonanthi (P<0.01, 5-10 mg/mL). In addition, the levels of MetHb were elevated significantly when challenged with Rhizoma Coptidis (2.8 fold at 5 mg/mL) and Cortex Moutan (3.4 fold at 10 mg/mL). This is the first report on the pro-oxidative action of CHM on G6PD-deficient blood samples in vitro as demonstrated by the decrease of GSH and increase of MetHb. G6PD-deficient subjects should restrain from excessive consumption of these pro-oxidative herbs.  相似文献

Rotavirus gastroenteritis should soon be a vaccine-preventable disease. In a 10-year survey of rotavirus gastroenteritis conducted at the Prince of Wales Hospital (PWH), 2281 cases were detected of which 2213 (97%) occurred in children < 5 years old. A consistent epidemic occurred each winter during the months of December and January. Of all laboratory-confirmed cases, 78% were community-acquired with a mean hospital stay of 4.7 days. The estimated incidence of rotavirus-attributed hospitalization was 2/1000 children < 5 years old. Over the 10 years, rotavirus was responsible for one death, and contributory to three other deaths. On average each year, 195 children < 5 years old were hospitalized for a total of 917 days in PWH, accounting for an estimated expenditure of HK$2.8 (approximately US$0.4) million on hospitalization costs. The annual financial burden for rotavirus gastroenteritis for the whole of Hong Kong could be in excess of HK$9.6 (approximately US$1.2) million.  相似文献

A multicenter clinical study was conducted using iohexol, a second-generation nonionic contrast medium, for excretory urography performed in 130 children. Doses of iohexol (300 mg iodine/ml) ranged between 150 and 660 mgI/kg (0.5 and 2.2 ml/kg). Iohexol was tolerated well, and no significant adverse reactions occurred. Sixty-five iohexol urograms were evaluated to determine the minimum dose for adequate visualization of the kidneys and collecting systems. A dose greater than 300 mgI/kg (1.0 ml/kg) always resulted in a urogram of diagnostic quality, while visualization was insufficient for diagnosis in 10% of studies done with doses of 150-300 mgI/kg (0.5-1.0 ml/kg). Another 65 iohexol urograms were compared in a blinded manner with a similar number of studies performed using iothalamate meglumine at comparable iodine concentration and dose. Visualization of calyces and pelvoinfundibular structures achieved with iohexol was rated better with statistical significance, but there was no difference in visualization of the renal parenchyma or ureters. Use of iohexol in excretory urography may be advantageous in children who are at greatest risk for an adverse reaction to contrast media or in those most likely to benefit from use of a low osmolality contrast agent.  相似文献

The authors give a review mainly about that worm species of the endoparasites of dogs and cats which are more frequently dangerous for the people living in urban circumstances because of the dissemination faeces of these animals. The life cycle of the nematode Toxocara species in dogs and cats as the final hosts, in inadequate++ hosts like man and the consequences of the zoonoses caused by these worms are briefly discussed. The authors survey the epidemiology, clinical appearances and the prevalence of human infections according to the data of the Hungarian and foreign literature. The clinical symptoms and the danger of this parasitosis running with the increasing number of the pet animals are pointed out. It is emphasized that the wide-range explanatory work and the role of the owners of pets in the decrease of the risks are very important, moreover that organizing the struggle against zoonoses such as toxocarosis is mainly veterinary task, however, its control is common medical and veterinary interest. The recognition, identification, diagnosis, treatment and prevention of human infections can only be effective with the proper knowledge in this zoonosis. Toxocarosis is one of the most frequent helminthozoonoses of townspeople in Hungary.  相似文献

The metabolism of atriopeptin prohormone ANF1-126 was examined with the aid of two separate radioimmunoassays, one detecting the C-terminal atriopeptins and the other detecting a fragment of the prohormone N-terminus. Intact prohormone standards are recognized in both assays, whereas the C-terminal atriopeptins are only detected by the atriopeptin assay. Both atriopeptin and N-terminal fragment immunoreactivities were detected in rat plasma and were simultaneously elevated following intravenous administration of desamino-arginine-vasopressin. Atriopeptin immunoreactivity returned to basal levels within 60 min after desamino-arginine vasopressin administration, whereas the N-terminal fragment immunoreactivity remained elevated for more than 2 hr. Analysis of both acid-boiled and sodium dodecyl sulfate-boiled rat atrial extracts by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and immunoblotting revealed the presence of a single high molecular weight species which reacted to both antisera and which comigrated with atriopeptin prohormone standards. Western blots of plasma from desamino-arginine vasopressin-stimulated rats yielded both the low molecular weight C-terminal atriopeptin and a high molecular weight N-terminal fragment-reactive peak which was smaller than the prohormone standards and which did not possess atriopeptin immunoreactivity. A recombinant 128-amino acid atriopeptin prohormone construct, ANF1-126-Arg-Arg, was used as a model substrate for prohormone metabolism. ANF1-126-Arg-Arg was specifically cleaved followed incubation with thrombin to yield the 98-amino acid N-terminal fragment and the C-terminal atriopeptin, AP28-Arg-Arg. Processing of ANF1-126-Arg-Arg by reperfusion through an isolated heart or by incubation in serum yielded identical metabolites to those generated by incubation with thrombin. No significant metabolism was observed following incubation of the prohormone with rat plasma. We conclude that the rat heart contains the necessary enzyme to cleave both endogenous and exogenous prohormone to atriopeptin and that processing by blood enzymes is not required.  相似文献

目的:以组织工程血管脱细胞支架和仿生髓核组织工程细胞支架、周围神经去细胞神经基膜管为例,介绍组织工程脱细胞支架的免疫原性研究情况. 方法:应用计算机检索Medline 1997-01/2007-03关于免疫原性的文章.检索词“Immunotoxicology”并限定文章的语种类为English.同时利用计算机检索中国期刊全文数据库1997-01/2007-03的相关文章,限定文章语言种类为中文,检索词“组织工程,免疫原性”. 结果:主要组织相容性复合体Ⅰ免疫组织化学方法能够检测血管脱细胞支架的免疫原性;组织学观察及反转录-聚合酶链反应检测γ-干扰素、白细胞介素2、白细胞介素4、白细胞介素10 mRNA表达,可以反映仿生髓核组织工程支架的免疫原性;主要组织相容性复合体Ⅱ抗原可以反映出经过化学萃取后的去细胞预变性神经基膜血管结构保留较完整,免疫原性低. 结论:脱细胞可以极大地降低组织工程支架的免疫原性,从而使得组织工程产品有更广阔的使用前景.  相似文献

BACKGROUND: Most blood centers utilize a confidential unit exclusion (CUE) process, intended to reduce the risk of transfusion-associated infectious diseases by allowing high-risk donors confidentially to exclude their blood from use for transfusion. The effectiveness of this method remains controversial. STUDY DESIGN AND METHODS: Confirmatory or supplemental test results for antibodies to human immunodeficiency virus, human T-lymphotropic virus type I, and hepatitis C virus, as well as hepatitis B surface antigen and syphilis and screening test results for antibodies to hepatitis B core (antigen) and alanine aminotransferase levels were obtained for approximately 1.8 million units donated during 1991 and 1992 at five blood centers within the United States. The prevalences of these infectious disease markers in units that the donors confidentially excluded (CUE+) and units that the donors did not exclude (CUE-) were calculated and examined within demographic subgroups. RESULTS: Units that were CUE+ were 8 to 41 times more likely to be seropositive for antibodies to human immunodeficiency virus and hepatitis C virus, hepatitis B surface antigen, and syphilis and three to four times more likely to react for antibody to hepatitis B core (antigen) or to have elevated alanine aminotransferase levels than units that were CUE- (p < 0.001). The positive predictive value of CUE (the percentage of CUE+ units that were confirmed seropositive for any marker) was 3.5 percent, and the sensitivity of CUE (the percentage of confirmed-seropositive units that were CUE+) was 2.3 percent. CONCLUSION: The current CUE process has low sensitivity and apparently low positive predictive value, and in many cases, it appeared that donors misunderstood it. Yet, CUE was not a “random process,” as CUE+ units were more likely to be seropositive for any infectious disease marker than CUE- units. This suggests that efforts to improve the CUE system may be warranted. As risk factors for transfusion-transmitted infection become more difficult to identify by history-based screening, however, such efforts may have limited effect.  相似文献
