We recently described a case of a Thrombospondin Type‐1 Domain containing 7A (THSD7A) associated membranous nephropathy in a female patient who was synchronously suffering from a THSD7A‐positive malignancy. We here investigated the role of THSD7A as a new potential tumor antigen by evaluating over 20 000 tissue spots in more than 70 different tumor entities by immunohistochemistry using tissue microarrays. THSD7A expression was highly variable in different neoplasias with differing staining patterns. Both gain and loss of THSD7A expression compared to expression status in non‐tumor tissue were linked to tumor‐specific markers in the different tumor entities and were of prognostic value. The potential role of THSD7A in tumor development and therapy needs further investigation.  相似文献

Dietary restriction extends lifespan in a variety of organisms, but the key nutritional components driving this process and how they interact remain uncertain. In Drosophila, while a substantial body of research suggests that protein is the major dietary component affecting longevity, recent studies claim that carbohydrates also play a central role. To clarify how nutritional factors influence longevity, nutrient consumption and lifespan were measured on a series of diets with varying yeast and sugar content. We show that optimal lifespan requires both high carbohydrate and low protein consumption, but neither nutrient by itself entirely predicts lifespan. Increased dietary carbohydrate or protein concentration does not always result in reduced feeding—the regulation of food consumption is best described by a constant daily caloric intake target. Moreover, due to differences in food intake, increased concentration of a nutrient within the diet does not necessarily result in increased consumption of that particular nutrient. Our results shed light on the issue of dietary effects on lifespan and highlight the need for accurate measures of nutrient intake in dietary manipulation studies.  相似文献

Background

Arsenic is a potent pollutant that has caused an environmental catastrophe in certain parts of the world including Bangladesh where millions of people are presently at risk due to drinking water contaminated by arsenic. Chronic arsenic exposure has been scientifically shown as a cause for liver damage, cancers, neurological disorders and several other ailments. The relationship between plasma cholinesterase (PChE) activity and arsenic exposure has not yet been clearly documented. However, decreased PChE activity has been found in patients suffering liver dysfunction, heart attack, cancer metastasis and neurotoxicity. Therefore, in this study, we evaluated the PChE activity in individuals exposed to arsenic via drinking water in Bangladesh.

Methods

A total of 141 Bangladeshi residents living in arsenic endemic areas with the mean arsenic exposure of 14.10 ± 3.27 years were selected as study subjects and split into tertile groups based on three water arsenic concentrations: low (< 129 μg/L), medium (130-264 μg/L) and high (> 265 μg/L). Study subjects were further sub-divided into two groups (≤50 μg/L and > 50 μg/L) based on the recommended upper limit of water arsenic concentration (50 μg/L) in Bangladesh. Blood samples were collected from the study subjects by venipuncture and arsenic concentrations in drinking water, hair and nail samples were measured by Inductively Coupled Plasma Mass Spectroscopy (ICP-MS). PChE activity was assayed by spectrophotometer.

Results

Arsenic concentrations in hair and nails were positively correlated with the arsenic levels in drinking water. Significant decreases in PChE activity were observed with increasing concentrations of arsenic in water, hair and nails. The average levels of PChE activity in low, medium and high arsenic exposure groups were also significantly different between each group. Lower levels of PChE activity were also observed in the > 50 μg/L group compared to the ≤50 μg/L group. Moreover, PChE activity was significantly decreased in the skin (+) symptoms group compared to those without (-).

Conclusions

We found a significant inverse relationship between arsenic exposure and PChE activity in a human population in Bangladesh. This research demonstrates a novel exposure-response relationship between arsenic and PChE activity which may explain one of the biological mechanisms through which arsenic exerts its neuro-and hepatotoxicity in humans.  相似文献

Post-installed rebars (PIRs) using mortar can offer bond strength at ambient temperature equal or higher to that of cast-in place rebars. However, high temperatures have the effect of weakening the bond, typically governed by the chemical and physical properties of the mortar which is often sensitive to temperature increase. Therefore, the behavior of PIRs in a fire situation becomes vulnerable. Moreover, after exposure of PIRs to high temperature, the heat transfer continues during the post-fire phase, which might endanger the construction after a fire event. In order to evaluate the evolution of the pull-out capacity during fire, Pinoteau et al. have developed the bond resistance integration method (Pinoteau’s RIM) to predict the bond resistance value of a rebar subjected to various temperatures in accordance with the fire exposure curves. Therefore, accurate temperature profiles during the post-fire phase are needed to ensure a correct calculation of the post-fire behavior of the PIR connection. This paper presents 3D finite element thermal simulations of PIRs in concrete exposed to ISO 834-1 fire conditions then cooled with ambient air. Numerical thermal profiles are then compared to the experimental results (i.e., post-fire pull-out tests). The proposed model provides guidelines for conducting numerical simulations to determine the thermal entry data necessary for predicting thermal profiles in PIRs during heating and cooling phases. Then, the post-fire pull-out capacity of PIRs in concrete is calculated using Pinoteau’s RIM, and compared to experimental post-fire pull-out results.  相似文献
