The bovine UK and simian SA11 rotaviruses are commonly used VP7-type reference strains. Since the surface protein VP3 is a significant neutralization antigen, it is important to fully characterize the VP3 types associated with current reference strains. Here we present the complete nucleotide and predicted amino acid sequence of VP3 from UK rotavirus (VP7 type 6) and compare it to the published sequences of SA114fm and RV-5. We also compare the deduced amino acid sequence covering the trypsin cleavage region of UK VP3 to 25 other available sequences. The UK protein is clearly different from that of bovine NCDV (another commonly used VP7 type 6 strain) and represents a second VP3 type associated with bovine rotaviruses. Our SA11 sequence differs from that determined by Lopez et al. [1985, Virology 144, 11-19; later referred to as SA114fM by Lopez et al. (1986, Virology 154, 224-227], their sequence being very similar to the published sequence of NCDV VP3. The significance of these results with regard to virus serotypes is discussed. Finally, in analyzing the nucleotide sequence surrounding the initiation codon, a potential hairpin-loop structure was identified which may be involved in translational regulation.  相似文献

This study compared autonomous self-regulation and negative self-evaluative emotions as predictors of smoking behavior change in college student smokers (N = 303) in a smoking cessation intervention study. Although the two constructs were moderately correlated, latent growth curve modeling revealed that only autonomous regulation, but not negative self-evaluative emotions, was negatively related to the number of days smoked. Results suggest that the two variables tap different aspects of motivation to change smoking behaviors, and that autonomous regulation predicts smoking behavior change better than negative self-evaluative emotions.  相似文献

Although motivational interviewing (MI) has been shown to be effective in changing health behaviors, its effects on smoking cessation have been mixed. The purpose of the present study is to assess factors of motivation and self-efficacy as they mediate the relationship between MI and smoking cessation. This is a secondary analysis of an MI based smoking cessation randomized trial. MI counseling was associated with change in smoking behaviors during a 12 months intervention but was not related to autonomous motivation, controlled motivation, or self-efficacy at baseline and 6 months, the hypothesized mediators. This study confirmed the pathway to quit smoking through increase in self-efficacy.  相似文献

Spindle-shaped viruses are a dominant morphotype in hypersaline waters but their molecular characteristics and their relationship to other archaeal viruses have not been determined. Here, we describe the isolation, characteristics and genome sequence of His2, a spindle-shaped halovirus, and compare it to the previously reported halovirus His1. Their particle dimensions, host-ranges and buoyant densities were found to be similar but they differed in their stabilities to raised temperature, low salinity and chloroform. The genomes of both viruses were linear dsDNA, of similar size (His1, 14,464 bp; His2, 16,067 bp) and mol% G+C (approximately 40%), with long, inverted terminal repeat sequences. The genomic termini of both viruses are likely to possess bound proteins. They shared little nucleotide similarity and, except for their putative DNA polymerase ORFs, no significant similarity at the predicted protein level. A few of the 35 predicted ORFs of both viruses showed significant matches to sequences in GenBank, and these were always to proteins of haloarchaea. Their DNA polymerases showed 42% aa identity, and belonged to the type B group of replicases that use protein-priming. Purified His2 particles were composed of four main proteins (62, 36, 28 and 21 kDa) and the gene for the major capsid protein was identified. Hypothetical proteins similar to His2 VP1 are present in four haloarchaeal genomes but are not part of complete prophages. This, and other evidence, suggests a high frequency of recombination between haloviruses and their hosts. His1 and His2 are unlike fuselloviruses and have been placed in a new virus group, Salterprovirus.  相似文献

A probable new Helicobacter species was isolated from the blood of a 14-month-old aboriginal child who presented with vomiting, diarrhea, fever, and dry cough. The most similar 16S rRNA gene sequence was that of Helicobacter fennelliae CCUG 18820(T) but the new sequence differed from it by at least 32 base substitutions and by the presence of a large (353-nucleotide) intervening sequence.  相似文献

A full-length ds cDNA copy of UK bovine rotavirus gene segment 9, which codes for a nonstructural protein, has been cloned into the PstI site of pBR322, and its sequence has been determined by cloning into bacteriophage M13mp8. Gene 9 is 1076 nucleotides long and contains a single, long, open-reading frame capable of coding for a protein of 313 amino acid residues. The possible function of this nonstructural protein in virus replication is discussed.  相似文献

Sequences of VP9 genes from short and supershort rotavirus strains   总被引:3,自引:0,他引:3  
Segment 10 genes from a short (RV-5, serotype G2) and a supershort (B37, a new G serotype) strain were cloned and their sequences compared to the (corresponding) segment 11 sequences of Wa, SA11, and UK rotaviruses. The determined nucleotide sequences were 817 (RV-5) and 947 (B37) bases in length and showed extensively conserved 5' noncoding and protein coding regions. The major open reading frame codes for a protein of 200 (RV-5) or 198 (B37) amino acids, and the newly proposed second open reading frame can code for a protein of 92 amino acids. Compared to long strain gene segments, the base sequences of the short and supershort strains were found to contain extended, AT-rich 3' noncoding regions which were not significantly homologous to each other, to other parts of the VP9 gene, or to other rotavirus genes that have been sequenced. The function(s) of these 3' regions is not apparent.  相似文献
