21－羟化酶CYP21B基因Ile~（172）→Asn错义突变

为了解先天性肾上腺皮质增生症患者的２１－羟化酶ＣＹＰ２１Ｂ基因中Ｉｌｅ~（１７２）→Ａｓｎ错义突变的发生率，根据放大受阻突变体系（Ａｍｐｌｉｆｉｃａｔｉｏｎｒｅｆｒａｃｔｏｒｙｍｕｔａｔｉｏｎｓｙｓｔｅｍ，ＡＲＭＳ）的要求，设计了３种引物：５＇ｄ（ＴＴＧＧＧＡＧＡＣＴＡＣＴＣＣＣＴＧＣＴＣＴ）３＇（共同引物）、５＇ｄ（ＡＧＧＴＧＡＧＧＴＡＡＣＡＧＡ）３＇（正常引物）、５＇ｄ（ＡＧＧＴＧＡＧＧＴＡＡＣＡＧＴ）３＇（突变引物），在７例患儿中进行了检测，发现具有本突变者３例。对其中一例进行的家系分析，结果提示：这组引物有快速、简便的优点，不需使用同位素就能对具有Ｉｌｅ~（１７２）→Ａｓｎ变异的高危家庭成员作产前诊断。     

In vitro and in vivo effects produced by the immunomodulating agent RU 41740 on human polymorphonuclear leukocytes in the elderly

The activity of RU 41740, a glycoprotein extract from Klebsiella pneumoniae has been investigated on some polymorphonuclear (PMN) functions. Chemotaxis, random migration and oxidative metabolism (assessed by chemiluminescence, O2 consumption and O2- generation) were studied in parallel. PMN were collected from adult and aged human volunteers. Experiments were performed either in vitro or in vivo in a double blind placebo assay. In both PMN populations RU 41740 enhanced oxidative metabolism either in in vivo or in vitro experiments. However, a higher and dose-related activity was observed on PMN collected from the younger subjects whereas maximal effective concentration was reached earlier with PMN collected from aged subjects. RU 41740 did not modify random migration but inhibited chemotaxis of PMN collected from the younger population in a dose-related manner. These data corroborated previous results observed on PMN collected from various animal species and suggested an interaction of RU 41740 on PMN membrane. Moreover drug-induced macrophage and lymphocyte stimulation might also explain, at least in part, the in vivo effects described in this study. Thus RU 41740 could partly account for the protective effects exerted against bacterial and fungal infections through its activity on PMN functions.     

Die Wertigkeit der Adenosintriphosphatase Reaktion zur Diagnostik von essentiellen Paraproteinämien

Zusammenfassung Bei 26 Fällen mit essentieller Paraproteinämie wurde die cytochemische Aktivität der Adenosintriphosphatase (ATPase) in den Plasmazellen des Knochenmarks untersucht. 15 Fälle zeigten eine erniedrigte und 11 Patienten eine normale ATPase-Aktivität. Die Untersuchungen zeigen, daß die Erniedrigung der ATPase für die Diagnostik des Plasmocytoms nicht spezifisch ist.Herrn Prof. Dr. W. Hoffmeister, Mannheim, zum 65. Geburtstag.     

前列腺素E1对大鼠卵泡壁发育和分化影响的形态学观察

本文应用组织学方法和电镜对PGE1引起未成熟大鼠卵泡颗粒细胞和卵泡膜外层细胞的形态学变化作了研究。结果表明:外源性PGE1刺激了颗粒细胞的发育和黄体化,同时也促进了卵泡膜外层细胞的发育和从成纤维细胞向平滑肌细胞分化。文中对有关PGE1在排卵过程中的作用机制等问题进行了讨论。     

Production of specific anti-rat 5-HT1A receptor antibodies in rabbits injected with a synthetic peptide

Polyclonal antibodies were raised by the repeated injection of rabbits with a synthetic peptide corresponding to a highly selective portion (amino acid residues 243 to 268) of the amino acid sequence of the rat 5-HT1A receptor. The anti-peptide antiserum allowed the immunoprecipitation of 5-HT1A receptors but not of other 5-HT1 sites solubilized from rat hippocampal membranes. Immunoautoradiographic labelling of rat brain sections with the anti-peptide antiserum was superimposed with the autoradiographic distribution of 5-HT1A sites labelled by the selective radioligand [3H]8-OH-DPAT.     

Heparin fragments modulate the collagen phenotype of fibroblasts from radiation-induced subcutaneous fibrosis

Acute local gamma irradiation of porcine skin induces, as in human skin, an extensive and mutilating sclerosis characterized by continuous expansion of the fibrosis invading the adjacent muscle and by accumulation of the macromolecular components of the extracellular matrix. Collagen synthesis, content, and types were studied in the presence of heparin fragments (100 micrograms/10(6) cells) in the culture medium, by measuring the incorporation of the radiolabeled precursor [3H]proline into confluent primary cultures of porcine fibroblasts obtained from normal and irradiated fibrotic dermis. Enhancement in collagen biosynthesis and deposition and preferential increase in collagen type III synthesis were observed in fibrotic fibroblast cultures when compared to those in normal dermis fibroblasts. The total collagen synthesis and the rate of collagen hydroxylation appear unmodified by heparin fragments both in normal and in fibrotic fibroblast cultures. But heparin fragments induce a 10- and 2-fold decrease, respectively, in collagen type III and type V syntheses by fibrosis fibroblasts. As only minor effects upon collagen type III and V are observed in cultures of normal dermis fibroblasts, these results highly suggest that heparin fragments are capable of specifically modulating the collagen phenotype of fibroblasts derived from radiation-induced dermis fibrosis and thus are able to regulate the fibrotic process.     

Interpretation of low-level Plasmodium infection rates determined by ELISA for anophelines (Diptera: Culicidae) from Egyptian oases

Plasmodium infection rates determined by enzyme-linked immunosorbent assay (ELISA) were compared for Anopheles sergentii (Theobald) and An. multicolor Cambouliu in Siwa Oasis, Egypt, an area with low-level Plasmodium vivax transmission, and in Bahariya and Farafra, two other Egyptian oases which appear to be free of malaria. Initial testing indicated that 4.4% (23 of 518) and 0.8% (4 of 518) of the An. sergentii were positive for P. vivax and P. falciparum, respectively, and that 1.4% (1 of 71) of the An. multicolor were positive for P. falciparum. However, after two confirmational tests, only 1.2% (6 of 518) of the An. sergentii remained consistently positive for P. vivax. Initial ELISA absorbance was not a useful predictor of potential false positive reactions in the P. vivax assay. Paradoxically, the six ELISA-positive An. sergentii were from the two malaria-free oases. This study raises the question of whether ELISA-positive reactions for anopheline vector species provides unequivocal evidence for transmission in areas of low malaria endemicity.     

面神经锥体段的解剖学及其临床意义

在80侧成人颞骨干标本上,对面神经锥体段的埋藏位置及深度,面神经与面神经隐窝、鼓室窦三者之间的关系及其变异类型进行了应用解剖学研究。提出了耳科后鼓室进手术路中对面神经锥体段的予测方法。经90例手术验证有实用价值。     

选择性髂内动脉插管灌注对盆腔恶性肿瘤介入性治疗的手术研究与探索

本文旨在探讨盆腔恶性肿瘤的介入性治疗问题。对45例患有盆腔恶性肿瘤的患者,采用Seldinger法,经双侧股动脉插管,先端选择性导入对侧髂内动脉;或一侧导入肠系膜下动脉,另侧导入髂内动脉,留置导管12小时,使用突击剂量持续灌注化疗药物。并经手术、B超、CT、内窥镜及指检等方法进行化疗前后的对比观察。结果:其中病灶消失持续1个月以上者9例、病灶缩小50％以上并持续超过一个月者23例、缩小不足50％且增大未达25％者9例、增大超过25％以上者4例,总有效率为71.1％。10例患者出现了脱发及皮肤色素沉着,1例患者出现下肢动脉栓塞,10例出现了不同程度的消化道反应等。结论:本法损伤小,操作简便安全,可多次重复术式:选择性强,药效持久均衡且毒副作用小;可提高手术切除率,也为不能手术的癌肿提供一种较理想的治疗手段。     

应用双抗体夹心ELISA检测人血清中Clq含量

本文采用抗人Clq单克隆抗体和多克隆抗体建立双抗体夹心ELISA,检测人血清中Clq含量。244份正常人血清标本检查结果,Clq正常值为287±56μg/ml。检测28份SLE患者血清有4份Clq呈明显下降.26份SLE病人血清同时用CH50试验检测总补体含量,发现二者呈正相关。因此,该技术有可能作为有关疾病如SLE等的临床诊断及疗效监测的辅助方法。
