Background: The sudden infant death syndrome (SIDS) is still the main cause of postneonatal infant death. However, the causes and mechanisms of SIDS have never been completely elucidated. Catecholamines, via α2-adrenergic receptor (α2-AR) interactions, are known to influence brainstem autonomic and respiratory activity. Aims: To examine the catecholaminergic system abnormalities in SIDS victims, we investigated the alterations of α2-AR subtypes. Subjects and methods: We examined the developmental changes of α2-AR subtypes in the brainstem, especially in cardiorespiratory nuclei, in 21 SIDS victims and 17 age-matched controls by means of immunohistochemical methods. For statistical analysis, the χ2-test or Fisher’s exact probability test was performed. Results: There was a significant decrease in α2A-AR immunoreactivity in the solitary nucleus and ventrolateral medulla (VLM) in the medulla oblongata in SIDS victims compared with in control cases, but there were no significant differences of the α2B and α2C-AR immunoreactivity in the brainstem between SIDS victims and controls. Conclusion: α2A-AR immunoreactivity was selectively decreased in the solitary nucleus and VLM in the medulla oblongata in SIDS victims, so there was no possibility that it was secondary to chronic hypoxia or repeated ischemia. It may be related to some impairment of the cardiorespiratory neuronal system. Therefore, SIDS victims may be vulnerable to asphyxia, hypoxia, and/or hypercapnia, and fail to exhibit brainstem responses.  相似文献

This report presents data illustrating the neurophysiological features of efferent-afferent convergence on cortical neurons. During combination of stimulus of pyramidal tract axons with electrocutaneous reinforcement, some of the study neurons were found to change the parameters of their activity evoked by stimulation of this tract: evoked responses started to reproduce the structure of responses to the reinforcement. The most dynamic rearrangements of pyramidal tract responses were obtained in experiments in which the study neurons were included in a biotechnical complex with feedback, the complex consisting of neuron-computer-stimulator-animal and actually being an analog of a natural functional system. The role of efferent-afferent convergence on CNS neurons in the development of the results-of-action acceptor of a functional system for a voluntary behavioral act is discussed.  相似文献

The effects of 7 days of "dry" water immersion were investigated in six subjects. Changes in the contraction properties were studied in the triceps surae muscle. After immersion, the maximal voluntary contraction (MVC) was reduced by 18.9 % (P < 0.01), and the electrically evoked (150 impulses s(-1)) maximal tension during tetanic contraction (P(o)) was reduced by 8.2 % (P > 0.05). The difference between P(o) and MVC expressed as a percentage of P(o) and referred to as force deficiency was also calculated. The force deficiency increased by 44.1 % (P < 0.001) after immersion. The decrease in P(o) was associated with increased maximal rates of tension development (7.2 %) and relaxation. The twitch time-to-peak was not significantly changed, and half-relaxation and total contraction time were decreased by 5.3 % and 2.8 %, respectively, but the twitch tension (P(t)) was not significantly changed and the P(t)/P(o) ratio was decreased by 8.7 %. The 60 s intermittent contractions (50 impulses s(-1)) decreased tetanic force to 57 % (P < 0.05) of initial values, but force reduction was not significantly different in the two fatigue-inducing tests: fatigue index (the mean loss of force of the last five contractions, expressed as a percentage of the mean value of the first five contractions) was 36.2 +/- 5.4 % vs. 38.6 +/- 2.8 %, respectively (P > 0.05). While identical force reduction was present in the two fatigue-inducing tests, it would appear that concomitant electrical failure was considerably different. Comparison of the electrical and mechanical alterations recorded during voluntary contractions, and in contractions evoked by electrical stimulation of the motor nerve, suggests that immersion not only modifies the peripheral processes associated with contraction, but also changes central and/or neural command of the contraction. At peripheral sites, it is proposed that the intracellular processes of contraction play a role in the contractile impairment recorded during immersion.  相似文献

The effect of different doses of sheep's red blood cells (SRBC) on endogenous colony formation and immunogenesis was studied in CBA and C57BL mice. The absolute and relative numbers of antibody-forming cells (AFC) was shown to increase with an increase in the dose of SRBC from 2×107 to 2×108, and to decrease with a dose of 2×109 in mice of both lines. With an increase in the dose of antigen, the number of endogenous splenic colonies (ESC) of hematopoietic cells also increased. Interlinear differences in the number of ESC persisted. It is suggested that the mechanism of stimulation of the hematopoietic series is based on a unique type of blockade of the reticuloendothelial system by foreign red blood cells, leading to increased proliferative activity of the stem cells.Laboratory of Regulation of Immunopoiesis, Institute of Clinical and Experimental Medicien, Siberian Branch, Academy of Medical Sciences of the USSR, Novosibirsk. This study assessed the antimicrobial resistance of nasopharyngeal pneumococci isolated from children aged < 5 years in day-care centres and orphanages throughout Russia during 2001-2002. Swabs were collected from 2484 children in 43 day-care centres and eight orphanages in 11 cities of European Russia, and from 1669 children in 37 day-care centres and three orphanages in eight cities of Asian Russia, with a total of 1144 and 912 Streptococcus pneumoniae isolates being recovered in European and Asian Russia, respectively. All macrolide-non-susceptible (MICs 0.5-128 mg/L) and fluoroquinolone-non-susceptible (ciprofloxacin MICs > or = 4 mg/L) isolates were tested for resistance mechanisms and clonal relatedness. Non-susceptibility rates, by CLSI criteria, were 19.3%, 0.9% and 0.4% for penicillin G, cefotaxime and amoxycillin-clavulanate, respectively. Resistance to macrolides and lincosamides was also relatively low, i.e., < 7% for clindamycin and 14- and 15-membered macrolides. The highest rates of non-susceptibility were for tetracycline and co-trimoxazole (52.0% and 64.5%, respectively). No clones resistant to ciprofloxacin (MICs > or = 8 mg/L) were found, but 1.7% of isolates were non-susceptible (MIC 4 mg/L). No resistance was found to levofloxacin, gemifloxacin, telithromycin or vancomycin. Pulsed-field gel electrophoresis analysis showed no relationship between ciprofloxacin- and macrolide-non-susceptible isolates in European and Asian Russia. Resistance among macrolide-resistant isolates resulted mostly from the presence of erm(B) and mef(A), and from changes in L4; additionally, L22 mutations were common in isolates from Asian Russia. Non-susceptibility to quinolones was associated with mutations in parC and parE among European isolates. Asian Russian isolates had mutations in parC and gyrA, and alterations in parE were more common. There were substantial differences in non-susceptibility and mechanisms of resistance between pneumococci from Asian and European Russia, with orphanages appearing to be 'hot-spots' of resistance.  相似文献

We studied adhesive properties and physiological activity in vivo of cells from Lewis lung carcinoma and its metastases. These cells differed in tumorogenic activity and metastatic potential in the syngeneic system. In vivo non-metastasizing cells are characterized by a lower content of surface lectins to tetrasaccharides SiaLex [Neu5Ac2-3Gal1-4(Fuc1-3) GlcNAc] and SiaLea [Neu5Ac2-3Gal1-3(Fuc1-4)GlcNAc] and trisaccharide HSO3Lex [HSO32-3Gal1-4(Fuc1-3)GlcNAc] compared to cells forming metastases in the syngeneic system. Metastatic cells with low tumorogenic activity weakly expressed lectins to disaccharide ligands 6-SiaLac [Neu5Ac2-6Gal1-4Glc], 6-HSO3LacNAc, and A-di [GalNAc 1-3Gal] and trisaccharides H-type 1 [Fuc1-2Gal1-3GlcNAc and Lex [Fuc1-3(Gal 1-4)GlcNAc] compared to cells that initiated tumor formation in the syngeneic system (similarly to transplanted tumors). We hypothesized that cell receptors to these carbohydrate determinates are involved in the development and growth of primary tumors, while lectins to SiaLex, SiaLea, and HSO3Lex play a role in the progress of tumor process and metastasizing.  相似文献

Kosaki overgrowth syndrome is a recently described syndrome characterized by distinctive facial features, brain white matter lesions, and developmental delay. Germline activating heterozygous PDGFRB mutations have been reported in this condition. Systemic connective tissue‐type findings have been described in some individuals. We describe a 19‐year‐old Caucasian female with a history of hydrocephalus, Dandy–Walker malformation, cervical spine arachnoid cyst, progressive scoliosis, and overgrowth. Her physical exam included distinctive craniofacial dysmorphism, as well as soft and hyperextensible skin. Cardiovascular imaging during adolescence revealed saccular aneurysms in both coronary artery systems and subtle tortuosity of the cervical vertebral arteries. Exome sequencing trio analysis identified a de novo previously reported pathogenic variant in PDGFRB, c.1696T>C (p.[Trp566Arg]). Further functional studies included platelet‐derived growth factor cellular metabolic pathway activity that confirmed the variant causes a constitutive activation of the PI3K‐AKT pathway. This is the first report to characterize the activating nature of this PDGFRB variant. We also highlight the connective tissue findings seen in Kosaki overgrowth syndrome and recommend baseline echocardiographic evaluation in all individuals with this condition with particular emphasis on coronary arteries.  相似文献

The helix-coil transition in Phage T2 DNA in the presence of 6,4 · 10?3 mol/l Mn2+ is studied using light scattering and UV spectroscopy. The transition range is about 0,5°C. Near the temperature of the end of melting Tf the molecular weight Mw and the radius of gyration Rz of the complex are observed to decrease to about one half. At a temperature 0,1–0,25°C higher than Tf, Mw and Rz pass through a minimum, which implies that aggregation is preceded by unwinding of DNA strands. Thus, rise in temperature rather than Mn2+ -induced aggregation causes DNA + Mn2+ melting.  相似文献
