观察血管紧张素Ⅱ 1型受体拮抗剂氯沙坦对球囊成形术后兔血管内膜增生和基质金属蛋白酶 2及其抑制剂表达的影响 ,探讨氯沙坦治疗血管再狭窄的可能机制和作用。 2 4只新西兰兔随机分为 3组 :对照组、模型组及氯沙坦组 ,后两组持续高胆固醇喂养 ,实验 1周后行腹主动脉内膜剥脱术 ,9周后对狭窄部位行球囊成形术 ,氯沙坦组于球囊成形术前 1周开始口服氯沙坦 10mg (kg·d)。 13周末取腹主动脉行病理形态学观察和基质金属蛋白酶2及组织型基质金属蛋白酶 2抑制剂的免疫组织化学分析。结果发现 ,高胆固醇饲养 9周后 ,模型组、氯沙坦组血清总胆固醇及低密度脂蛋白胆固醇均明显增加 (P <0 .0 1) ,且未发现氯沙坦干预后对血脂水平有影响。 13周后氯沙坦组较模型组内膜厚度减少 65 % ,内膜面积减少 3 4% ,内膜厚度与中膜厚度比减少 3 2 % ,内膜面积与中膜面积比减少 2 8%。免疫组织化学分析显示 ,基质金属蛋白酶 2表达因氯沙坦的干预无明显变化。而组织型基质金属蛋白酶 2抑制剂表达明显减少 (P <0 .0 5 ) ,基质金属蛋白酶 2 组织型基质金属蛋白酶 2抑制剂显著增加 (P <0 .0 1)。提示氯沙坦在不影响血脂的情况下 ,可明显抑制血管损伤后内膜增生 ,其机制可能与抑制组织型基质金属蛋白酶 2抑制剂表达 ,恢复基质金属蛋 相似文献

本研究旨在探索红景天苷对骨髓MSC增殖及干细胞因子(SCF)分泌的影响.应用密度梯度离心和贴壁培养法,体外分离并扩增MSC,应用流式细胞术和成脂及成骨诱导法进行MSC鉴定,并观察红景天苷对MSC增殖、细胞周期及分泌SCF的影响.结果表明,红景天苷可影响MSC的增殖,以1.5 mg/ml浓度刺激MSC增殖作用最强,在24、48、72 h内存在一定的时间依赖性,并且红景天苷明显增加S、G1/M期细胞比例,尤以1.5 mg/ml作用明显.应用1.5 mg/ml红景天苷与MSC共培养48 h,其培养上清中SCF含量明显上调(P<0.01),其mRNA表达也明显增加(P<0.01).结论:在一定剂量浓度下红景天苷对MSC有明显增殖促进作用,并可提高MSC的CSF基因表达和分泌. 相似文献

AIM: To analyze the clinicopathological features of Brunner‘s gland adenoma of the duodenum.METHODS: A rare case of Brunner‘s gland adenoma of the duodenum was described and related literature was reviewed.RESULTS: Brunner‘s gland adenoma of the duodenum appeared to be nodular hyperplasia of the normal Brunner‘s gland with an unusual admixture of normal tissues, including ducts, adipose tissue and lymphoid tissue. We suggested that it might be designated as a duodenal hamartoma rather than a true neoplasm.CONCLUSION: The most common location of the lesion is the posterior wall of the duodenum near the junction of its first and second portions. It can result in gastrointestinal hemorrhage and duodenal obstruction. Endoscopic polypectomy is a worthy treatment for benign Brunner‘s gland adenomas,as malignant changes in these tumors have never been proven. 相似文献

Angiotensin II type 1 receptor (AT1-R) and nuclear factor-kappaB (NF-κB) in the paraventricular nucleus (PVN) play important roles in heart failure (HF); however, the central mechanisms by which AT1-R and NF-κB contribute to sympathoexcitation in HF are yet unclear. In this study, we determined whether interaction between AT1-R and NF-κB in the PVN modulates neurotransmitters and contributes to NAD(P)H oxidase-dependent oxidative stress and sympathoexcitation in HF. Rats were implanted with bilateral PVN cannulae and subjected to coronary artery ligation or sham surgery (SHAM). Subsequently, animals were treated for 4 weeks through bilateral PVN infusion with either vehicle or losartan (LOS, 10 μg/h), an AT1-R antagonist; or pyrrolidine dithiocarbamate (PDTC, 5 μg/h), a NF-κB inhibitor via osmotic minipump. Myocardial infarction (MI) rats had higher levels of glutamate (Glu), norepinephrine (NE) and NF-κB p65 activity, lower levels of gamma-aminobutyric acid (GABA), and more positive neurons for phosphorylated IKKβ and gp91phox (a subunit of NAD(P)H oxidase) in the PVN when compared to SHAM rats. MI rats also had higher levels of renal sympathetic nerve activity (RSNA) and plasma proinflammatory cytokines (PICs), NE and epinephrine. PVN infusions of LOS or PDTC attenuated the decreases in GABA and the increases in gp91phox, NF-κB activity, Glu and NE, in the PVN of HF rats. PVN infusions of LOS or PDTC also attenuated the increases in RSNA and plasma PICs, NE and epinephrine in MI rats. These findings suggest that interaction between AT1 receptor and NF-κB in the PVN contributes to oxidative stress and sympathoexcitation by modulating neurotransmitters in heart failure. 相似文献

Previous studies have shown that dietary acid load (DAL) harms bone health, but the evidence is inconsistent and insufficient. This study examined the relationships between DAL and the risk of hip fracture. This case–control study contained 1070 pairs of 1:1 age-, city-, and gender-matched incident cases and controls (mean age, 71 years) recruited in Guangdong, China. Dietary information was collected using a validated 79-item food frequency questionnaire through face-to-face interviews. DAL was estimated based on established algorithms for the potential renal acid load (PRAL) and net endogenous acid production (NEAP). Higher PRAL and NEAP were dose-dependently associated with a higher risk of hip fracture in both the conditional logistic regression model and restricted cubic spline analysis after adjusting for potential covariates. The multivariate-adjusted odds ratios and 95% CI of hip fracture for tertiles 2 and 3 (vs. 1) of DAL were 1.63 (1.18, 2.25) and 1.92 (1.36, 2.71) for PRAL and 1.81 (1.30, 2.53) and 2.55 (1.76, 3.71) for NEAP in all participants (all p-trends < 0.001), respectively. Subgroup analyses showed more pronounced associations in participants with a lower body mass index. Our findings suggested positive associations between the estimated DAL and the risk of hip fractures in the elderly Chinese population. 相似文献

Major histocompatibility complex class I chain-related A (MICA) is a highly polymorphic gene located within the MHC class I region of the human genome. Expressed as a cell surface glycoprotein, MICA modulates immune surveillance by binding to its cognate receptor on natural killer cells, NKG2D, and its genetic polymorphisms have been recently associated with susceptibility to some infectious diseases. We determined whether MICA polymorphisms were associated with the high rate of Schistosoma parasitic worm infection or severity of disease outcome in the Dongting Lake region of Hunan Province, China. Polymerase chain reaction-sequence specific priming (PCR-SSP) and sequencing-based typing (SBT) were applied for high-resolution allele typing of schistosomiasis cases (N = 103, age range = 36.2-80.5 years, 64 males and 39 females) and healthy controls (N = 141, age range = 28.6-73.3 years, 73 males and 68 females). Fourteen MICA alleles and five short-tandem repeat (STR) alleles were identified among the two populations. Three (MICA*012:01/02, MICA*017 and MICA*027) showed a higher frequency in healthy controls than in schistosomiasis patients, but the difference was not significantly correlated with susceptibility to S. japonicum infection (Pc > 0.05). In contrast, higher MICA*A5 allele frequency was significantly correlated with advanced liver fibrosis (Pc < 0.05). Furthermore, the distribution profile of MICA alleles in this Hunan Han population was significantly different from those published for Korean, Thai, American-Caucasian, and Afro-American populations (P < 0.01), but similar to other Han populations within China (P > 0.05). This study provides the initial evidence that MICA genetic polymorphisms may underlie the severity of liver fibrosis occurring in schistosomiasis patients from the Dongting Lake region. 相似文献
