Quantitative and qualitative composition of hemopoietic tissue and the intensity of proliferation and differentiation of committed hemopoietic precursors are studied in mice treated with adriamycin, cyclophosphane, or 5-fluorouracil. It is shown that intense maturation of colony-forming units results in rapid regeneration of hemopoiesis even under conditions of reduced proliferation of these cells, whereas inhibition of their differentiation suppresses restoration of bone marrow cellularity despite intense proliferation of hemopoietic precursors. The effect of different doses of sheep's red blood cells (SRBC) on endogenous colony formation and immunogenesis was studied in CBA and C57BL mice. The absolute and relative numbers of antibody-forming cells (AFC) was shown to increase with an increase in the dose of SRBC from 2×107 to 2×108, and to decrease with a dose of 2×109 in mice of both lines. With an increase in the dose of antigen, the number of endogenous splenic colonies (ESC) of hematopoietic cells also increased. Interlinear differences in the number of ESC persisted. It is suggested that the mechanism of stimulation of the hematopoietic series is based on a unique type of blockade of the reticuloendothelial system by foreign red blood cells, leading to increased proliferative activity of the stem cells.Laboratory of Regulation of Immunopoiesis, Institute of Clinical and Experimental Medicien, Siberian Branch, Academy of Medical Sciences of the USSR, Novosibirsk. Summary Efferent neurons in isolated pedal ganglia of the pteropodial mollusc Clione limacina were filled with Lucifer Yellow through the wing nerves. Then the ganglia were illuminated with intense blue light which resulted in the complete inactivation of these neurons. After inactivation of efferent neurons, interneurons of the pedal ganglia continued to generate the locomotor rhythm.  相似文献

本实验室设计合成了三种不同LA/GA比例的mPEG修饰PLGA(PELGA,含15%mPEG),为了评价它们的血液相容性,我们以硅化玻璃试管为阴性对照,未硅化的试管为阳性对照,参照国际标准(ISO10993)和《中华人民共和国国家标准GB/T16886医疗器械生物学评价》方法进行了体外评价实验。试验包括溶血率实验,血小板黏附实验,动态凝血时间实验,凝血时间实验,血浆复钙时间实验和凝血酶原时间实验等综合评价指标。结果表明,合成材料具有优良的血液相容性,材料制成的纳米粒有望应用于静脉注射。  相似文献

多孔聚磷酸钙(Calcium polyphosphate,CPP)生物陶瓷是一种新型的骨组织工程支架材料,国外有研究表明聚磷酸钙作为骨组织工程支架材料具有良好的生物相容性及可降解性。通过对原料煅烧过程的研究,探索了聚合度的计算方法,制备出不同聚合度的材料。通过对材料烧结温度的试验,制得了不同晶型的聚磷酸钙多孔材料。在Tris—HCl缓冲液中进行的体外降解实验表明,CPP多孔支架材料是可控降解的,不同聚合度和晶型的支架材料力学强度和降解性能不同。随着聚合度的增加,材料的力学强度增大,降解速率变小;非晶态的CPP17d就可以完全降解,7晶型的CPP25d可完全降解,β和α晶型的CPP降解缓慢,30d分别降解了大约12%和5%。因此聚磷酸钙是一种很有前途的骨组织工程支架材料,凭借其独特的无机聚合物结构及降解性能,有望实现可控速率的降解。  相似文献

Shiga toxin (Stx) follows a complex intracellular pathway in order to kill susceptible cells. After binding to cell surface glycolipids, the toxin is internalized and trafficked in retrograde fashion to the endoplasmic reticulum (ER). From the ER lumen, the toxin must gain access to the cytoplasm, where it enzymatically inactivates the 28S rRNA, inhibiting protein synthesis. The host molecules involved in this pathway and the mechanisms utilized by the toxin to access the cytoplasm from the ER are largely unknown. We found that Stx is capable of energy-dependent transport across the ER lumen, as has recently been demonstrated for the cholera and ricin toxins. Genetic screening for molecules involved in Shiga toxin trafficking yielded a cDNA encoding a prematurely truncated protein. Characterization of this cDNA revealed that it encodes a novel Hsp40 chaperone, designated HEDJ or ERdj3, localized to the ER lumen, where it interacts with BiP, a molecule known to be involved in protein retrotranslocation out of the ER. We demonstrated that within the ER lumen Stx interacts with HEDJ and other chaperones known to be involved in retrotranslocation of proteins across the ER membrane. Moreover, sequential immunoprecipitation revealed that Shiga toxin was present in a complex that included HEDJ and Sec61, the translocon through which proteins are retrotranslocated to the cytoplasm. These findings suggest that HEDJ is a component of the ER quality control system and that Stx utilizes HEDJ and other ER-localized chaperones for transport from the ER lumen to the cytosol.  相似文献

Horizontal eye movements were recorded in eight healthy subjects during super-slow trunk rotation with respect to the space-stationary head. In some trials, subjects simultaneously indicated their perception of selfmotion by means of a joystick. Over the frequency range employed (0.007–0.05 cycles per second, ±20°), all subjects perceived the relative motion of head and trunk as a head rotation with respect to the stationary trunk. Eye movements were observed which were in phase with imaginary head rotation; their amplitude exceeded the amplitude of actual body rotation. The grasping of a rigid ground-based handle (1) produced a sensation of trunk rotation in space, (2) suppressed the sensation of imaginary head rotation in space and (3) gave rise to a significant decrease in amplitude of eye movements. The grasping of a stiff rod with non-zero compliance did not produce these effects. It is concluded that eye movements in response to body rotation with respect to the fixed head are not purely reflex reactions, but are influenced by the internal representation of body motion.  相似文献
