目的:对黄芩属植物半枝莲Scutellaria barbataD.Don全草的抗肿瘤活性部位(三氯甲烷萃取物)的化学成分进行分离、鉴定。方法:采用反复硅胶、LH-20葡聚糖凝胶等柱色谱方法进行分离纯化,并通过理化常数测定与谱学分析鉴定其化学结构。结果:得到7个化合物,分别鉴定为木犀草素(1),柚皮素(2)、半枝莲酸(3),2,4-二羟基苯甲酸(4),香豆酸乙酯(5),β-谷甾醇(6),胡萝卜苷(7)。结论:化合物4、5为首次从半枝莲中分离得到。  相似文献

目的:探讨喜脑宁冻干粉(XNN)对全脑缺血的保护作用及其可能机制.方法:采用四动脉结扎法建立大鼠全脑缺血/再灌注模型,观察XNN对脑缺血再灌注过程中脑电图及翻正反射恢复时间、脑含水量、脑指数、脑组织匀浆MDA和LD含量、SOD,LDH和NOS活性、海马ET含量的影响;观察XNN对ADP诱导的血小板聚集率的影响;采用Fura-2/AM负载法,观察XNN对血小板内静息[Ca~(2+)]_i;和CaCl_2诱导血小板内[Ca~(2+)]_i,浓度变化的影响.结果:在全脑缺血再灌注模型上,XNN可以缩短翻正反射恢复时间,促进脑电图的恢复;降低脑含水量、脑指数;提高脑组织SOD,LDH活性,降低LD,MDA含量和NOS活性;降低海马组织ET含量.XNN对ADP诱导的血小板聚集有一定的抑制作用,能降低血小板内静息期[Ca~(2+)]_i,并对CaCl_2诱导的血小板内[Ca~(2+)]_i的升高有抑制作用.结论:XNN对全脑缺血具有一定的保护作用.  相似文献

目的:比较14种常用抗肿瘤中药的体外抑瘤效果。方法:采用四甲基偶氮唑兰(MTT)法分别测定了14种常用抗肿瘤中药的水提液、醇提液对人胃癌细胞株AGS、人胃癌细胞株BGC-823体外增殖反应的影响。结果:该14种中药中有10味体外对人胃癌细胞株AGS、人胃癌细胞株BGC-823体外增殖均有阳性抑制作用;且该10味中药醇提液体外对人胃癌细胞株AGS、人胃癌细胞株BGC-823体外增殖抑制作用优于水提液;有2味中药醇提浸膏对人胃癌细胞株AGS、人胃癌细胞株BGC-823体外增殖抑制作用均为强阳性;而其余2味中药对2株细胞抑制率为阴性。结论:14种中药中10味体外对胃癌细胞抑制率大于50%,但提取工艺对抑瘤效果存在较大影响,应尽量采用醇提工艺提取。  相似文献

Oxidized low-density lipoprotein receptor 1(OLR1)is upregulated in neurons and participates in hypertension-induced neuronal apoptosis.OLR1 deletion exerts protective effects on cerebral damage induced by hypertensive-induced stroke.Therefore,OLR1 is likely involved in the progress of intracerebral hemorrhage.In this study,we examined the potential role of OLR1 in intracerebral hemorrhage using a rat model.OLR1 small interfering RNA(10μL;50 pmol/μL)was injected into the right basal ganglia to knock down OLR1.Twenty-four hours later,0.5 U collagenase type VII was injected to induce intracerebral hemorrhage.We found that knockdown of OLR1 attenuated neurological behavior impairment in rats with intracerebral hemorrhage and reduced hematoma,neuron loss,inflammatory reaction,and oxidative stress in rat brain tissue.We also found that silencing of OLR1 suppressed ferroptosis induced by intracerebral hemorrhage and the p38 signaling pathway.Therefore,silencing OLR1 exhibits protective effects against secondary injury of intracerebral hemorrhage.These findings suggest that OLR1 may be a novel potential therapeutic target for intracerebral hemorrhage.  相似文献

The Corona Virus Disease 2019 (COVID-19) pandemic has huge impacts on the world, including human health and economic decline. The COVID-19 has severe infectivity, especially the elderly with chronic diseases will cause various complications after infection and accelerate the disease process. In addition, COVID-19 will also affect their mental health. Therefore, the mental health of elderly patients with chronic diseases cannot be ignored. The aim of this study was to investigate the well-being level of elderly people with chronic disease during COVID-19 postpandemic period in Beijing and analysis related influencing factors, so as to provide a basis for improving the well-being level of elderly chronic patients during the postpandemic period.Elderly patients with chronic diseases who met the inclusion criteria in 5 different administrative regions in Beijing were selected to carry out a questionnaire survey. The contents of the questionnaire included general data, the Memorial University of Newfoundland Happiness scale and the awareness situation of the COVID-19 pandemic. A total of 500 questionnaires were distributed by WeChat and 486 valid questionnaires were collected. The t test and one-way analysis of variance were used to compare Memorial University of Newfoundland Happiness scores between 2 or more groups, multiple linear regression analysis was used to conduct multiple factor analysis to explore the related factors about well-being level of elderly chronic patients.A total of 109 cases (22.43%) were evaluated high well-being level, 319 cases (65.64%) were evaluated moderate well-being level and 58 cases (11.93%) were evaluated low well-being according to the Memorial University of Newfoundland Happiness (MUNSH) scores rating. The multiple linear regression indicated that the education level, number of chronic diseases, medical expenses, frequency of children''s visits, taking care of grandchildren or not, and group activity frequency significantly affected the well-being of patients with chronic diseases during COVID-19 postpandemic period in Beijing (P < .05).Most elderly patients with chronic diseases had moderate or above sense of well-being during postpandemic period, but we should still pay attention to the mental health of those elderly chronic patients with low education level, much comorbidity, more medical expenses, less visits by children, not take care of grandchildren and never participate in group activities.  相似文献

The wastewater from ready-mixed concrete plants is currently being recycled as concrete mixing water. It has attracted significant attention from the construction industry and researchers since it promotes sustainable development through environmental protection, energy-saving, and emissions reduction. This article review first introduces the nature of wastewater in ready-mixed concrete plants in different regions. Then the effects of solid content in water on various properties of concrete, including working performance, durability and microscopic properties, are reviewed, respectively, when concrete is mixed with wastewater instead of tap water. Furthermore, the microscopic mechanism of action in concrete mixing with wastewater is discussed, and future work is recommended. This review provides fundamentals on the study of the properties of concrete after wastewater is mixed into concrete.  相似文献

目的:对早孕绒毛的滋养细胞进行原代培养,利用HBV感染血清对滋养细胞进行体外感染,探讨细胞在体外培养条件下对HBV的感染率及HBV在细胞中的复制状态.方法:利用胰蛋白酶和胶原酶联合消化培养法原代培养早孕胎盘中的滋养细胞,并进行免疫组化鉴定,获得纯度较高的滋养细胞后进行体外的HBV感染,通过细胞免疫化学染色、ELISA及PCR法分别检测细胞上清中的HBsAg,细胞中的HBVDNA及cccDNA.结果:原代培养的滋养细胞,细胞生长良好,特异性角蛋白一7阳性,纯度高.原代滋养细胞在与HBV阳性血清共同孵育后均未出现明显的细胞病变,感染后24h和48h在滋养层细胞细胞浆中可见HBsAg的阳性表达,被感染细胞呈散在分布.感染后48h起可检测到弱阳性的HBsAg及HBVDNA,感染后细胞内未检测出cccDNA.结论:利用联合酶消化法建立了胎盘中滋养细胞的体外培养系统.HBV能够在体外感染原代培养的滋养细胞,对被感染细胞的生长无明显影响.HBV可能不在滋养细胞中复制.  相似文献

间隙连接蛋白(Cx)在保证细胞间物质信息传递及维持皮肤屏障稳态方面发挥着重要作用,其基因突变,甚至表达水平异常均会引起多种疾病,严重影响患者生活质量。在编码人类Cx家族的21个基因中,与Cx基因突变相关的临床伴随疾病至少有14种。其中,Cx43分布最广,不仅在大多数器官组织中均有报道,也是伤口愈合、皮肤角质化,以及皮肤肿瘤发展等重要生理病理过程中的关键调控节点。本文总结了近年来Cx43基因(GJA1)在皮肤屏障中的作用、GJA1基因突变相关皮肤疾病及其潜在致病机制这几个快速发展领域中的研究成果,以期为Cx43临床伴发疾病防治及相关研究提供参考。  相似文献
