卡西霉素对血小板在脑微血管内皮细胞粘附的影响及药物的阻断作用

体外实验表明calcimycin0.01μmol·L^-1刺激牛脑微血管内皮细胞25min,使兔血小板与脑微血管内皮细胞的粘附率增高17.1%。PAF受体拮抗剂WEB20860.1,1.0和10.0μumol·L^-1对血小板在脑微血管内皮细胞上粘附的抑制率分别为9.0,22.9和23.1%。DMPP0.1,1.0,10.0μmol·L^-1及Tet0.1,1.0,10。Oμmol·L^-1的抑制率分别为9.7,15.6,22.1%和7.8,15.6及24.6%,提示DMPP和Tet对脑血管有保护作用。     

若干卤代靛玉红衍生物的合成

靛玉红用于临床治疗慢性粒细胞白血病已有多年,疗效明确。但溶解度差,影响在体内的吸收。近年来为了提高疗效降低肠胃道副作用,在亚氨基和羰基上合成了一系列化合物,并筛选出一些有抗肿瘤作用的活性物质,在靛玉红芳环上的卤代物,虽有Eller-Pandrand等作过一些工作,但未见用于抗肿瘤上,本文在芳环上引进卤素拟增加溶解度,共合成了6个卤代靛玉红,经药理抗肿瘤筛选,在平行试验中,化合物Ⅰ,Ⅱ,Ⅲ和     

Staphylococcus epidermidis bacteremia from transfusion of contaminated platelets: application of bacterial DNA analysis

Septicemia is a rare complication of platelet transfusion. A case is reported of transfusion-associated septicemia in a 66-year-old man who received a 10-unit pool of platelets. During transfusion, he experienced rigors, wheezing, dyspnea, and fever. A total of four blood cultures drawn 10 and 36 hours after discontinuation of the transfusion grew Staphylococcus epidermidis. Culture of the residual platelet pool yielded S. epidermidis with a colony count of 10(5) organisms per mL. Strain identity of all four blood isolates and the platelet pool isolate was confirmed by gel electrophoresis of EcoRI and HindIII restriction digests of whole-cell DNA. There have been 31 prior reported cases of platelet transfusion-associated septicemia, of which 9 have been caused by coagulase-negative staphylococci. Systemic reactions to platelet transfusions should prompt consideration of transfusion-associated bacteremia as the cause.     

血小板激活因子对大鼠脑血管通透性的影响及药物的保护作用

颈内动脉注射血小板激活因子(PAF),再给伊文思蓝,可见脑实质染色程度加深,而颈内动脉只注射伊文思蓝,脑实质未见染色。而我们合成的新药SZ-1可剂量依赖性地抑制PAF诱导的脑实质伊文思蓝染色程度的加深。在体外培养的脑微血管平滑肌细胞上,PAF能显著刺激¹⁴C-花生四烯酸的释放,而SZ-1能剂量依赖性地抑制这种释放,提示PAF在脑内产生的损害除与其他因素相关外,还与其刺激花生四烯酸释放有密切关系,SZ-1对PAF引起的脑部损害有保护作用。     

5-取代-4-甲基-1,3-二氢-2H-咪唑-2-酮类化合物的合成及其强心作用

本文设计并合成了5-取代-4-甲基-1,3-二氢-2 H-咪唑-2-酮类化合物23个,其中19个尚未见报道。中间体5-(4-溴甲基苯甲酰基)-4-甲基-1,3-二氢-2H-咪唑-2-酮(Ⅴ)为一新化合物。在关键中间体4-甲基-1,3-二氢-2H-咪唑-2-酮(Ⅳ)的合成中,设计了由β-羟基丙胺一步氧化直接合成氨基丙酮的新方法。药理初筛表明:Ⅱ₁,Ⅱ₂,Ⅱ₄,Ⅲ₄,Ⅲ₆,Ⅲ₇,Ⅲ₈和RMI 82,249(1)具有不同程度的正性肌力作用,其中4个未知化合物Ⅱ₁,Ⅱ₂,Ⅱ₄和Ⅲ₄的最大效应高于已知对照物(1)。Ⅱ₄既增加心肌收缩力,又减慢心率,值得进一步研究。

Deletion of the human retinoblastoma gene in primary leukemias

As an initial step in evaluating the role of tumor suppressor genes in leukemogenesis, we surveyed primary leukemia cells from 130 patients for possible deletion of the retinoblastoma susceptibility (Rb) gene by Southern blot analysis. Two of them clearly showed homozygous deletion of Rb alleles. The first patient was a pre-B acute lymphoid leukemia (ALL) associated with a cytogenetic translocation: t(14;16)(q24;q22). The deletion was located at the 3' portion of the Rb gene, very close to the site of Rb gene deletion recently identified in an ALL cell line. The absence of Rb110 protein was further confirmed by Western blot analysis. The second patient was a chronic myelomonocytic leukemia (CMMoL), terminated in acute blastic transformation. Deletion of the 5' portion of Rb gene was found in leukemic cells in the chronic stage. The results indicated that inactivation of the Rb gene occurred in certain cases of leukemia. Its significance warrants further study.     

蝙蝠葛碱和粉防己碱对[3H]WEB 2086与体外牛脑前动脉平滑肌细胞结合的影响(英文)

用标记的血小板活化因子拮抗剂[³H]WEB 2086,在培养的牛脑前动脉平滑肌细胞上鉴定了血小板活化因子受体。结果表明在25℃时该细胞上存在两种与配基具有不同亲和力的受体结合位点,其中K_d-1=22.8±5.0 nmol·L^-1,K_d-2=186+20.5 nmol·L^-1;B_max-1=2.1±0.3 pmol/10⁴细胞,B_max-2=12.1±1-5 pmol/10⁶细胞。蝙蝠葛碱和粉防己碱均能抑制[³H]WEB2086与上述细胞的结合。
