大肠癌组织p53基因突变及其与临床病理关系的探讨

应用非同位素标记ＰＣＲ－ＳＳＣＰ技术对３４例大肠癌组织ｐ５３基因第五外显子突变进行了检测。结果：５０％（１１／２２）结肠癌，８．３％（１／１２）直肠癌存在ｐ５３基因突变，低分化腺癌高于高、中分化腺癌（Ｐ＜０．０５），ＤｕｋｅｓＢ、Ｃ期高于ＤｕｋｅｓＡ期（Ｐ＜０．０５），ｐ５３基因第五外显子突变在已发生淋巴转移的大肠癌组织中呈现较强的趋势。提示：检测结肠癌原发灶ｐ５３基因第五外显子突变，对指导结肠癌的诊断、治疗及预后判断具有一定意义     

壮骨肾宝对实验性小鼠高脂血症的影响

目的观察由黄芪、白术、淫羊藿组成的复方制剂(壮骨肾宝)对实验性小鼠高脂血症的影响。方法用蛋黄乳液快速致小鼠单纯性高胆固醇血症和喂养高脂饲料致高脂血症模型 ,观察预防性给予不同剂量的壮骨肾宝后对血清总胆固醇(TC)、甘油三酯(TG)和高密度脂蛋白胆固醇(HDL -ch)的影响。结果壮骨肾宝中、高剂量能使血清TC降低(P<0.05),HDL -ch升高(P<0.01) ,但对TG的影响不大。结论壮骨肾宝有一定降血脂作用 ,并与剂量有关     

示波极谱法测定环境中痕量叶蝉散

本文研究了叶蝉散的水解产物邻异丙基酚在硼酸-NaOH－EDTA底液中与MBTH偶合生成物，在电位-0.35V（VS、S、C、E）处的极谱行为，加入等体积的乙醇可提高方法灵敏度和稳定性。方法灵敏度高，较比色法高一个多数量级；线性范围广，0．04～1．5PPm与峰高成正比；合并变异系数4.1％；平均回收率96％～99．2％；方法操作简便、快速，干扰少；样品分析结果与比色法无差异。     

Treatment of hemophilia A by living mother-to-son splenic transplantation. First case report in the world.

A case of severe hemophilia A was treated by living mother-to-son splenic transplantation. The recipient was a 9-year-old boy. After splenic transplantation, the level of blood factor VIII:C was increased, and no spontaneous hemorrhage occurred. The boy was followed up for over two years. This is a case of spleen allotransplantation with the longest function in the world.

     

一起幼儿园内空肠弯曲菌高感染情况的调查

作者报道了一起幼儿园内空肠弯曲菌高感染的传播。该幼儿园儿童空肠弯曲菌阳性率为23.0％(43/187)。腹泻病例空肠弯曲菌阳性率为18.6％(8/43),健康儿童带菌率为24.3％(35/144)。同期另一幼儿园健康儿童和门诊非腹泻患儿带菌率分别为11.3％(27/238)和7.5％(15/201),三者间差异显著(P<0.005)。通过流行病学调查及菌株的生物分型表明,传染源可能是两名患空肠弯曲菌肠炎的炊事员,他们与儿童的菌株为同一生物型。其传播可能是通过污染的食物或人与人接触而引起的。     

In situ hybridization of prostate-specific antigen mRNA in human prostate.

Prostate-specific antigen (PSA) mRNA was detected by in situ hybridization utilizing a 428 base pair [35S]-labelled cDNA probe from the 3' noncoding region of the PSA gene. Thirty six fresh surgical specimens were collected from patients undergoing radical retropubic prostatectomy for carcinoma of the prostate. Quantitative analysis of the levels of PSA mRNA in both the benign and malignant tissues was performed using an IBAS 2000 Image Analysis System. The results of this study demonstrated that there is a significant decrease in the expression of PSA mRNA in the carcinoma tissue when compared to the benign epithelium. The average binding (number of silver grains/1 x 10(4) microns. 2) for 20 specimens of malignant epithelium was 475 +/- 161 and 586 +/- 140 for 16 specimens of benign epithelium (p less than 0.05). Eleven patients had both benign and malignant tissue from the same surgical specimen available for study. From these paired specimens, the PSA mRNA expression was also significantly reduced in the malignant epithelium when compared to the benign epithelium, 445 +/- 162 and 588 +/- 135 respectively (p less than 0.005). The PSA protein was detected using a monoclonal antibody to PSA with an immunohistochemical staining technique. The PSA protein expression paralleled the expression of the PSA mRNA in the majority of the tissue sections. Many of the tumor specimens showed a heterogeneous expression of PSA, whereas all of the benign epithelium had a uniform high level of PSA expression. In conclusion, PSA mRNA and protein are located only within the glandular epithelial tissue, the expression of PSA protein parallels that of the PSA mRNA, and both the PSA protein and PSA mRNA are significantly decreased in the malignant epithelium when compared to benign prostatic epithelium.     

R波相关振动法血压测量

为了提高振地血压测量的准确性，提出了一咱基于心电Ｒ波与血压振动波之间相关性，识别振动信号的一种抗干扰方法＝－Ｒ波相关法，结合振动波形特征识别，该方法可有效地提取信号，消除干扰影响，该算法已在１６位８０９８单片机上实现，本文同时给出了程序的详细流程图。     

铸造桩核烤瓷全冠在后牙残冠和残根修复中的临床价值

目的：探讨铸造桩核烤瓷全冠在后牙牙体缺损修复中的价值。方法：选择后牙残根、残冠的164颗牙，在修复前行完善根管治疗术，再采用铸造桩核烤瓷冠的方法修复缺损。通过3～24个月的复查结果进行临床效果评价。结果：该修复体固位力强，边缘密合性好且耐磨，经过修复后2年复查，有效率达95．12％。结论：铸造桩核烤瓷全冠是一种效果良好的后牙残根、残冠的修复方式。     

热缺血供体同种异体犬肺移植术后早期肺功能的观察

目的　观察犬热缺血供体同种异体肺移植术后早期移植肺功能情况。方法　采用室温下热缺血 3 0min、LPD液灌注后 4℃保存 2 5h时的供肺 ,行右肺同种异体移植手术 7例 ,术前及监测恢复灌注 0 5 ,1,2及 4h的动脉血氧分压 (PaO2 )、肺动态顺应性 (Cdyn)、平均肺动脉压 (mPAP)、肺毛细血管楔压 (PCWP)和心输出量的变化 (CO)。结果　所有受体犬在观察期间均存活。恢复灌注 4h内的肺功能各指标变化差异无显著性 (P >0 0 5 ) ,或者虽然有统计学差异 ,但仍有较好的绝对水平。结论　热缺血 3 0min低温保存 2 5h的供肺可以维持生命 ,恢复灌注后 ,受缺血再灌注损伤的影响 ,在灌注后一段时间内 ,供肺功能下降 ,但仍可较好支持生命。     

In vitro differentiation of human adipose-derived adult stromal cells into neuron-like cells in hippocampal astrocyte conditioned medium

BACKGROUND: At present, researches on differentiating from human adipose-derived adult stromal cells (hADASC) to neuron-like cells are focus on inducing by artificial-synthetic compound solution; however, hippocampal astrocyte conditioned medium (HCAM) can induce in vitro differentiation from hADASC to neuron-like cells is still unclear. OBJECTIVE: To observe whether HCAM can induce in vitro differentiation from hADASC to neuron-like cells. DESIGN: Randomized control study. SETTING: Department of Neurology, Taixing People's Hospital; Central Laboratory, North China Coal Medical College. MATERIALS: Donor of adipose tissue was donated by female volunteers suffering from caesarean section in the department of obstetrics & gynecology in our hospital and aged 20－35 years. Adipose tissue was collected from subcutaneous tissue of abdomen during the operation. In addition, 8 male newborn Wistar rats within 24 hours with average body mass of 20 g were provided by Animal Institute of Chinese Academy of Medical Sciences. Rabbit-anti-human Nestin polyclonal antibody, rabbit-anti-human glial fibriliary acidic protein (GFAP) polyclonal antibody, rabbit-anti-human neuro-specific enolase polyclonal antibody and mouse-anti-human microtubal associated protein 2 (MAP-2) polyclonal antibody were provided by Wuhan Boster Company. METHODS: The experiment was carried out in the Central Laboratory of North China Coal Medical College from October 2004 to June 2005. hADASC was cultured with HCAM and its growth and morphological changes were observed under inverted phase contrast microscope. Immunocytochemistry, immunofluorescence and Western blotting were used to evaluate the expressions of Nestin, which was a specific sign of nerve precursor, neuro-specific enolase and MAP-2, which was a specific sign of nerve cell, and GFAP, which was a specific sign of neuroglial cells. MAIN OUTCOME MEASURES: Nestin, which was a specific sign of nerve precursor, neuro-specific enolase and MAP-2, which was a specific sign of nerve cell, and GFAP, which was a specific sign of neuroglial cells. RESULTS: On the 3rd day of culture, partial hADASC started deformation from slender shuttle-shape cells to neuron-like cells. It suggested that cells stretched out apophysis, which were mainly double-pole or multiple-pole cells. Five days later, immunohistochemical detection suggested that expression of Nestin (10.5±0.037) was found out in cells; meanwhile, expressions of GFAP (38.4±0.052) and neuro-specific enolase (NSE) (15.7±0.023) were also found out in cells; however, expression of MAP-2 was not observed. Western blot indicated that, 5 days after effect of HCAM, Nestin was found out in hADASC; meanwhile, expressions of GFAP and neuro-specific enolase were also found out; however, expression of MAP-2 was not observed. CONCLUSION: HCAM can induce the differentiation from hADASC to neuron-like cells in vitro.
