Synthesis and Reactions of 4-Alkyl-2-phenyl-3,4-dihydro-5H-1,4-benzodiazepine-5-ones The synthesis of the 4-alkyl-2-phenyl-3,4-dihydro-5H-1,4-benzodiazepine-5-ones (1a) by reductive cyclisation of the corresponding 2-nitro-N-alkyl-N-phenacylbenzamides (5) is reported. The different reactions of the new 4-alkyl-derivatives and the known 4-unsubstituted compounds of this ring system are described.     

Sigmoid colon injuries caused by blunt abdominal trauma]

Circumferential seromuscular stripping of the sigmoid colon with denudation of the mucosal tube over a length of 4 to 10 cm has been observed in four patients after blunt abdominal trauma. Three of the patients had been driving a motor car, with two of them wearing safety belts. One patient was injured when he was caught between steel bars at work. In two cases the sigmoid lesion was associated with a tear in the mesoileum, leading to ischemic bowel necrosis in one. In one patient the lesion was found 5 days after the accident when he presented the symptoms of diffuse peritonitis caused by secondary perforation of the denuded mucosal tube. Treatment consisted in resection, anastomosis and proximal colostomy in two, and resection with end colostomy and distal mucous fistula in the remaining two cases.--As long as perforation of the denuded mucosa has not occurred, symptoms may be minimal. Liberal use of peritoneal lavage in polytraumatized patients is therefore important in order to minimize the likelihood of overlooking this type of bowel lesion.

The enzyme, Qβ replicase, responsible for the replication of the RNA of Escherichia coli pahge Qβ, is composed of four nonidentical subunits, three of which, I, III, and IV, are coded for by the bacterial genome, while subunit II is phage-specific.     

Histopathological changes in autotransplanted baboon kidneys after 24-hour hyperbaric, hypothermic preservation perfusion

Baboon kidneys were preserved at 4 C and 3 atm for 24 hr. Thirty-two kidneys were perfused with G.S.H. replacement solution or Plasmalyte B during preservation. Fifteen kidneys were flushed with the same solution prior to preservation but not perfused during preservation. Twenty-six kidneys were flushed with serum prior to preservation and also not perfused during preservation. The preserved kidneys were autotransplanted to the iliac fossa. Histopathological studies of the kidneys were undertaken between the second and twenty-fourth day after reinsertion. It was found that preservation perfusion was frequently accompanied by glomerular thrombosis, glomerular and interstitial fibrosis as well as endarteritis in many kidneys. These changes were rarely observed in unperfused preserved kidneys. The observed renal alterations are similar to those described in the literature as chronic pyelonephritis in long-term studies of preserved dog kidneys.     

Enzyme pattern-directed chemotherapy. Effects of antipyrimidine combinations on the ribonucleotide content of hepatomas.

Using combinations of three antipyrimidine drugs, chosen on the basis of expected antihepatoma selectivity, we examined the effects on pyrimidine ribonucleotide pools in rat hepatomas (Morris 8999 and 3924A) and host livers. d-Galactosamine gave a marked decrease in UTP in hepatomas, 3-deazauridine caused decreases in CTP concentration, and these two agents together gave very pronounced (up to 86 per cent) decreases in hepatoma CTP content. Pyrazofurin caused decreases in UTP in both hepatoma and host liver, but in combination with d-galactosamine, pyrazofurin decreased hepatoma CTP by up to 79 per cent without decreasing the host liver CTP. The combination of pyrazofurin and d-galactosamine also gave a significantly greater depression of UTP levels in hepatoma 8999 than in host liver. Uridine and orotic acid both restored the d-galactosamine-induced depletion of UTP in liver and hepatoma 8999.
